The effect of estrus induction by cabergoline on gonadotropin and steroid hormone responses was examined in anestrous bitches. Eleven beagles were used in the study; seven were included in the estrus induction group and four were included in the spontaneous estrus group. Cabergoline was orally administered to the estrus induction group at 5 µg/kg once daily for four weeks, or until hemorrhagic discharge was detected. The inter-estrus interval in the estrus induction group was significantly shorter than the previous estrus interval. Bitches that showed proestrus within four weeks of treatment showed increased luteinizing hormone (LH) pulse frequency and, subsequently, increased estradiol (E2) levels. Prolactin (PRL) levels declined promptly after treatment, except in one bitch that did not show proestrus during the cabergoline treatment period. There was a significant correlation between the time to proestrus induction and the reduction in PRL levels. A positive correlation was found between the LH levels two weeks after cabergoline administration and PRL reduction. This study demonstrates that an abrupt reduction in PRL is likely to be important for initiation of estrus in bitches. A reduction in PRL indirectly leads to an increase in LH pulse frequency, which regulates follicular development in bitches. However, if the period from the end of the previous estrus to the cabergoline treatment is short, it may take some time to show proestrus without increasing E2 levels, even if the LH level increases after cabergoline administration.  相似文献

Improving drought tolerance of container seedlings of Japanese larch is of high importance to afforestation. We hypothesized that adequate nitrogen (N) and limited water supply would increase the tolerance of container seedlings to water-deficit stress, circumventing photoinhibition, by means of (i) enhanced photosynthetic capacity with higher leaf N and (ii) decreased water loss from leaves with lower biomass allocation into aboveground parts. Container seedlings of Japanese larch were grown under the treatment combinations of adequate (+?N: 300 mg N container?1) or limited (??N: 150 mg N container?1) N and adequate (+?W: daily irrigation) or limited (??W: twice-a-week irrigation) water. Then, seedlings were subjected to a progressive drought treatment. Higher leaf N was observed in container seedlings grown under?+?N and???W. During progressive drought, lower stomatal conductance and net photosynthetic rate were observed in leaves with higher leaf N at a given predawn leaf water potential. Furthermore, the maximum efficiency of PSII photochemistry (Fv/Fm) was lower in leaves with higher leaf N, suggesting that higher leaf N might impair intrinsic tolerance to drought at the leaf level contrary to expectations. Conversely,???N and???W seedlings with lower shoot biomass delayed soil drying as a whole-plant response via a reduction in leaf transpiration, leading to delayed photoinhibition as indicated by a decline in Fv/Fm. To circumvent stress at the initial stage of water deficit, lower leaf N via limited N regime and smaller shoot biomass driven by limited N and water regimes would be important.

The purposes of this study were 1) to examine changes in the indocyanine green (ICG) clearance by feeding and 4-day fasting in dry cows, and (2) to investigate the relationship between ICG clearance and blood chemistry profile in postpartum cows. In 3 dry cows, post-feeding ICG half-life (T(1/2)) was shorter than the pre-feeding value, and post-fasting T(1/2) was longer than pre-feeding and post-feeding values. In 16 lactating cows, T(1/2) value at 2 weeks postpartum showed positive correlations with AST, T-Bil and γ-GTP. These results suggested that ICG clearance correlated with T-Bil and liver enzymes can be sensitive and accurate diagnostic marker for impaired liver function in dairy cows. In addition, ICG clearance is greatly affected by feeding and fasting.  相似文献

We used chlorophyll fluorescence to examine photosynthetic responses to excess Mn accumulation in leaves of four tree species differing in successional traits. Betula ermanii Cham. (Be) and Alnus hirsuta Turcz. (Ah) were studied as representatives of early-successional species. Ulmus davidiana Planch. var. japonica (Rehder) Nakai (Ud) was selected as a mid-successional species, and Acer mono Maxim. var. glabrum (Lév. et Van't.) Hara (Am) was chosen as a late-successional species. In Be, Ah and Am, high foliar concentrations of Mn had little effect on maximum photochemical efficiency of photosystem II (PSII), as indicated by the values of dark-adapted F(v)/F(m), whereas a significant decrease was observed in Ud. Photochemical quenching (qP) and the excitation capture efficiency of open PSII (F'(v)/F'(m)) decreased with increasing leaf Mn concentration at photosynthetic steady state after a 15-min exposure to 430 &mgr;mol m(-2) s(-1) PPFD. Compared with early-successional species, these decreases were greater in mid- and late-successional species leading to lower effective quantum efficiencies of PSII (DeltaF/F'(m) = qP x F'(v)/F'(m) = (F'(m) - F)/F'(m)). To determine the extent of photoinhibition, F(v)/F(m) of the illuminated leaves was remeasured after a 15-min dark period. Compared with the dark-adapted F(v)/F(m), we observed a significant decrease in F(v)/F(m) in Am leaves containing high concentrations of Mn. These chlorophyll fluorescence studies indicate that the early-successional species Be and Ah have a higher tolerance to excessive accumulations of Mn in leaves than the mid- and late-successional species Ud and Am.  相似文献

We evaluated the effects of elevated carbon dioxide concentration ([CO2]) and two nutrient regimes on stem growth rate, annual ring structure and temporal variations in photosynthetic characteristics of seedlings of Japanese larch (Larix kaempferi (Lamb.) Carr.). Seedlings were grown in phytotron chambers in an ambient (360 ppm) or an elevated (720 ppm) [CO2] in two nutrient regimes for one growing season. Elevated [CO2] reduced stem height and increased stem basal diameter compared with ambient [CO2]. The effect of elevated [CO2] on growth tended to be greater at high-nutrient supply than at low-nutrient supply. Elevated [CO2] had no significant effect on ring width or the number of tracheids per radial file. There was no obvious difference in cell wall thickness or the relative area of the cell wall between seedlings grown in ambient or elevated [CO2]. Although growth in elevated [CO2] resulted in a slight increase in cell diameter, the increase had a relatively minor effect on the relative area of the cell wall. Net assimilation rate increased in response to elevated [CO2]; however, the increase in whole-crown photosynthetic rate (Total Agrowth) in seedlings in the elevated [CO2] treatment was minimal because of the smaller specific needle area and acclimation of the photosynthetic characteristics of the needles to the growth [CO2]. In conclusion, we observed no obvious enhancement in the capacity for carbon fixation in Japanese larch seedlings grown in the presence of elevated [CO2] that might be attributable to changes in stem growth. However, elevated [CO2] caused changes in the temporal pattern of stem growth and in some anatomical features of the tracheids.  相似文献

A primary cultured cell line named CHKS was established from a hepatocellular carcinoma (HCC) of a dog showing a high level of serum alpha-fetoprotein (AFP). CHKS secreted a 66 KDD AFP into the growth medium regardless of the presence or absence of fetal bovine serum (FBS). Cloning CHKS with limiting dilution produced 4 clones, CHKS-1, -2, -3, and -4, which secreted 826, 471, 70, and less than 10 ng/ml, respectively, of AFP into the culture medium. In culture, these cell lines were similar in morphology and proliferation pattern to epithelial cells and positive to periodic acid-Schiff (PAS) staining. The presence of mRNA for canine albumin was demonstrated by nested PCR. The doubling times of the clone cell lines were 21, 45, 36, and 35 h, saturation densities 34, 18, 22, and 24 x 10(4)/cm(2), and plating efficiencies 18, 45, 46, and 45%, respectively. Chromosome analysis of these cell lines showed near triploidy. These results show that CHKS and its clones have hepatic cell functions and are useful for carcinogenetic and clinical studies of canine HCC.  相似文献

Blood estrone sulfate (E1S), estrone (E1), estradiol (E2) and progesterone (P4) in newborn piglets were measured to clarify the relationships among birth and placental weight, vitality of offspring and litter size. First, the association between vital status (normal, weak and stillborn) from 165 newborn piglets of 18 litters and steroid concentrations; second, steroid concentrations from 152 newborn normal piglets and litter size; and third, steroid content in fetal placenta from 50 newborn normal piglets of six litters and litter size, were investigated. In the normal group, the birth and placental weight were significantly higher than those in the other groups. Blood E1S levels in the stillborn group were significantly lower, whereas E1, E2 and P4 were significantly higher compared to the normal group. Blood and placental E1S levels in the small litter group were significantly higher than those in the other groups. However, there was no significant difference among the three litter size groups in the levels of steroid hormones in maternal blood. These results indicate that vitality of newborn piglets is related to E1S concentration of neonate, to birth weight and placental weight. However, steroid hormone concentrations of newborn piglets were greatly affected by the number of littermates.  相似文献

Transdermal absorption of lidocaine was determined by measuring plasma lidocaine concentrations following skin application of 5% lidocaine patches. Two lidocaine patches were placed on the ventral abdominal midline of seven dogs for 72 hours. Lidocaine was detectable in plasma 12 hours after patch application, and it reached steady-state concentrations between 24 and 48 hours. Plasma lidocaine levels decreased dramatically at 60 hours post-application. Low plasma lidocaine concentrations remained for 6 hours after patch removal. No clinically significant side effects were noted.  相似文献

Characterization of CD34+ cells in canine bone marrow, umbilical cord blood, and peripheral blood was performed by flow cytometric analysis. The ratio of CD34+CD45hi cells, which are absent in human blood, was high in the CD34+ cell fraction, but 98% of these was suggested B-cells. The remaining CD34+CD45lo cells may comprise canine hematopoietic progenitor cells, and these cells accounted for 0.23 +/- 0.07% of the fraction in cord blood, 0.30 +/- 0.07% in bone marrow, and 0.02 +/- 0.01% in peripheral blood.  相似文献
