Leptin, a pleiotropic hormone regulating food intake and energy expenditure, has been shown to directly modulate human polymorphonuclear neutrophil (PMN) functions or indirectly through the action of tumor necrosis factor-alpha (TNF-alpha). Bovine PMN have considerable different characteristics from human PMN. For example, it does not respond to N-formyl-Methionyl-Leucyl-phenylalanine, a well known human PMN activator. In the present study, we tested the effects of leptin and TNF-alpha on superoxide production and degranulation of bovine peripheral PMN, in which both long isoform of leptin receptor (Ob-Rb) and TNF receptor 1 were expressed. Human leptin, human TNF-alpha, phorbol myristate acetate (PMA) and opsonized zymosan particles (OZP) did not stimulate degranulation responses, while zymosan-activated serum (ZAS) did. Neither leptin nor TNF-alpha enhanced the ZAS-induced degranulation responses. TNF-alpha, PMA, OZP and ZAS increased superoxide production in different magnitudes, whereas leptin did not. TNF-alpha, but not leptin, enhanced OZP- and ZAS-induced superoxide production, possibly, in part due to facilitating translocation of p47(phox), a component of NADPH oxidase. These results indicate that, unlike in human PMN, leptin does not have any direct effect on degranulation and superoxide production in bovine PMN, although TNF-alpha influences superoxide production. 相似文献

The amount of heavy metals discharged from daily life was estimated in order to know the origin of the metals contained in the sewage sludge which is produced in the wastewater treatment plant treating only domestic wastewater. The amounts of the metals discharged from our daily life were estimated to be in the range of 0.2 to 0.3 for Cd, 1.6 to 1.9 for Ni, 3.5 to 6.8 for Pb, 0.8 to 1.4 for Cr, 8.2 to 19.3 for Mn 9.4 to 55.8 for Cu, 44.3 to 62.7 for Zn and 111 to 293 for Fe in mg.d–1 per person, Using these data, the cycle of the metals in our daily life was discussed in relation to land application of sewage sludge. 相似文献

The effects of the preceding crops, sunflower (mycorrhizal host) and mustard (nonhost), on arbuscular mycorrhizal (AM) colonization and growth of succeeding maize were examined in 17 soils in an attempt to clarify the influence of soil characteristics on the effects of preceding crops. Shoot weight and P uptake of maize planted after sunflower were much higher than those after mustard in 14 soils, although the preceding crop had little effect on soil-P availability. AM colonization of maize after sunflower was much higher than that after mustard. The effect of the preceding crop was eliminated by soil sterilization. These results suggested that the differences in maize growth were caused by differences in the AM colonization. Correlation analysis of the effect of the preceding crop and soil properties showed that the difference in the effects on maize growth could not be explained by soil chemical properties, but only by the AM colonization of the preceding sunflower crop. In one of the 17 soils, however, the effect was not evident despite the higher AM colonization of sunflower. This soil was sterilized, and the effect of inoculation by AM fungi (AMF) on maize was examined. However, it was found that the inoculation increased AM colonization but did not improve maize growth at any P level, suggesting that the effect of AMF was unusually inhibited in this soil by unknown soil physicochemical properties. In most soils, however, the preceding mycorrhizal host crop, sunflower, improved the growth and AM colonization of maize depending on the AM colonization of sunflower. 相似文献

The effects of B and Ca treatments on root growth, nutrient localization and cell wall properties in wheat ( Triticum aestivum L.) plants with and without Al stress were investigated. Seedlings were grown hydroponically in a complete nutrient solution for 7 d and then treated with B (0, 40 μM), Ca (0, 2,500 μM), and Al (0, 100 μM) in a 500 μM CaCl2 solution for 8 d. The cell wall materials (CWM) were extracted with a phenol: acetic acid: water (2:1:1 w/v/v) solution and used for subsequent pectin extraction with trans -1,2-diami-nocyclohexane- N,N,N,N -tetraacetic acid (CDTA) and Na2 CO3 solutions. Boron, Ca, and B + Ca treatments enhanced root growth by 19.5, 15.2, and 27.2%, respectively, compared to the control (pH 4.5). Calcium and B+Ca treatments enhanced root growth with Al stress by 43 and 54%, respectively, while B did not exert any effect. The amounts of CWM and pectin per unit of root fresh weight increased by Al treatment, whereas the Ca and B+Ca treatments slightly reduced the contents of these components. Seventy-four percent of total B, 69% of total Ca, and 85% of total Al were located in the cell wall in the B, Ca, and Al treatments, respectively and 32% of total B, 33% of total Ca, and 33% of total Al were located in the CDTA-soluble and Na2 CO3 -soluble pectin fractions. A more conspicuous localization of B was observed in the presence of Al. Aluminum treatment markedly decreased the Ca content in the cell wall as well as pectin fractions, mainly in the case of the CDTA-soluble pectin fraction. Boron + Ca treatment decreased the Al content in the cell wall and pectin fractions compared to the Ca treatment alone in the presence of Al. It is concluded that the B+Ca treatment enhanced root growth and, B and Ca uptake, and helped to maintain a normal B and Ca metabolism in the cell walls even in the presence of Al. 相似文献

This case report describes a case of spontaneous pancreatic islet cell carcinoma with vascular invasion in a 110-week-old male F344 rat. Histologically, a pancreatic nodule consisting of tumor cells and many blood-rich vessels, and covered with a fibrous capsule showed local invasion in the capsule and adjacent acinar tissues, encircling a large duct-like structure (DS). The tumor was composed of well-differentiated tumor cells resembling normal pancreatic islet cells, which had small round nuclei and eosinophilic cytoplasm. Mitotic figures were rare. Immunohistochemistry revealed that the tumor cells were positive for insulin. Although endothelial cells were not detected, the DS wall showed cells positive for α-smooth muscle actin and elastic fibers, suggesting that the DS is the pancreatic artery. This is a rare case of islet cell carcinoma consisting of well-differentiated tumor cells with invasion of the pancreatic artery in a rat. 相似文献

We have previously established the method for isolation of ceramide aminoethylphosphonate (CAEP) from jumbo flying squid Dosidicus gigas. In this study, we performed a MTT assay to evaluate the safety of CAEP to the cell lines for the application to health food and supplements. The CAEP did not show any cytotoxicity to various HEK293-transfectant cells. Next, we elucidated the positive function of CAEP to the somatic cells. Recently, we have reported that hepatotoxin microcystin-LR was taken up into the hepatocytes mediated by hepatocellular uptake transporters OATP1B1 and OATP1B3, and the cells were induced cytotoxicity subsequently. Cytotoxicity of microcystin-LR to permanently OATP1B3-expressing HEK293-OATP1B3 cells rather than to HEK293-OATP1B1 cells was preferentially attenuated by CAEP in a concentration-dependent manner. In addition, the enzyme activity of serine/threonine phosphatase, which was inhibited by microcystin-LR, was recuperated by co-exposure to CAEP. Furthermore, microcystin-LR-induced cellular protein phosphorylation were disrupted by CAEP exposure. These results suggested that CAEP is a promising remedy and/or preventive medicine for liver damage with microcystin-LR. 相似文献

To search for edible herbal extracts with potent antioxidant activity, we conducted a large scale screening based on the superoxide scavenging activity. That is, scavenging activity against superoxide anions were extensively screened from ethanol extracts of approximately 1,000 kinds of herbs by applying an electron spin resonance (ESR)-spin trapping method. Among them we chose four edible herbal extracts with prominently potent ability to reduce the signal intensity of 5,5-dimethyl-1-pyrroline-N-oxide (DMPO)-OOH, a spin adduct formed by DMPO and superoxide anion. They are the extracts from Punica granatum (Peel), Syzygium aromaticum (Bud), Mangifera indica (Kernel), and Phyllanthus emblica (Fruit), and are allowed to be used as foodstuffs according to the Japanese legal regulation. The ESR-spin trapping method coupled with steady state kinetic analysis showed that all of the four extracts directly scavenge superoxide anions, and that the superoxide scavenging potential of any of the extracts was comparable to that of L-ascorbic acid. Furthermore, polyphenol determination indicates that the activity is at least in part attributable to polyphenols. These results with such large scale screening might give useful information when choosing a potent antioxidant as a foodstuff. 相似文献

Interannual variations in abundance, timing of outmigration from rivers, growth rate and condition of juvenile chum salmon
(Oncorhynchus keta) were studied in the Nemuro Strait (eastern Hokkaido, Japan) during 1999–2002 to establish a possible relationship to zooplankton
abundance. The otolith microstructure of juveniles was examined each year in late June to determine their time and size at
sea entry (i.e., outmigration), and to estimate the early marine growth rates. Salmon outmigration peaked in mid- or late
May, which coincided, in three of the four study years, with the peak release of juveniles into rivers within the study area.
Abundance, growth rate and condition of fish were higher in 2001, when—compared to other years—smaller fish experienced higher
growth rates, coinciding with greater zooplankton abundance for that year. Our results suggest that high zooplankton abundance
positively influenced juvenile chum salmon growth and the condition of the fish during their early marine life despite their

In order to determine the effects of ghost fishing by lost gill nets, the relationship was examined between soak time and
number of enmeshed animals in experimentally lost gill nets by using diving observations. Two experimental gill nets were
set at 13 m depth in Tateyama Bay, Chiba Prefecture, Japan for approximately 200 days. One gill net was deployed in a small
trough surrounded by artificial reefs, and the other was deployed on an adjacent open sandy bed. Twelve species of crustaceans,
six species of gastropods, and five species of bony fish were enmeshed in the experimental gill nets. The number of enmeshed
animals in the artificial reef gill net was substantially larger than that in the sandy bed gill net. The number of enmeshed
animals in the experimental gill nets increased rapidly within one month after deployment, and then declined gradually showing
fluctuations caused by the decrease in newly enmeshed animals, and the drop off from gill nets caused by the decomposition
of dead animals. The decrease in the number of enmeshed animals was expressed by logarithmic equations, and based on these
equations, the duration of capture function for the lost gill nets was calculated to be 284–561 days in the artificial reef
gill net and 200 days in the sandy bed gill net. The duration of capture function for the lost gill nets for non-commercial
by-catch species such as small crustaceans and gastropods was longer than for commercial species such as Japanese spiny lobster
