Nilaparvata lugens (Stål) is one of the major pests of rice throughout tropical and temperate Asia. Indiscriminate use of insecticides for suppressing N. lugens has resulted in the development of resistance to multiple insecticide classes, causing frequent control failures in the field. Analysis of gut bacterial diversity within an insect host is the initial step towards understanding the ecological roles of the symbionts. Present study aimed to survey the bacterial diversity associated with laboratory-reared (insecticide-susceptible) and field-collected (insecticide-resistant) populations of N. lugens by culture-dependent and PCR-Denaturing Gradient Gel Electrophoresis (DGGE) methods. Seventeen bacterial isolates were obtained by the culture-dependent method. Molecular characterization using the 16S rRNA gene and phylogenetic analysis revealed that the isolates belonged to Firmicutes and Proteobacteria. Taxonomic assignment placed these isolates into seven families representing 10 genera. Enterobacteriaceae was the most dominant family with its occurrence in four out of the five populations studied. The DGGE profiles indicated a low complex gut bacteria associated with N. lugens with limited number of bands. The Shannon-Wiener index ranged from 0.898 in insecticide-susceptible population to 0.946–1.035 in resistant populations. Sequencing and phylogenetic analysis revealed that the DGGE bands belonged to Firmicutes, Proteobacteria and Bacteriodetes. Results of this study illustrated that gut bacterial community associated with N. lugens is dominated by Proteobacteria and Firmicutes. Present findings could provide the basis for future work on the possible role of the bacterial symbionts in insecticide resistance and to formulate potential resistance management strategies.  相似文献

The formation of biogenic amines in Indian oil sardines (Sardinella longiceps) collected from the Tuticorin coast of South India and treated with delayed salt-curing was investigated. Sardines were wet salt-cured in whole and gutted forms and examined in fresh and after 6, 9, 12, and 15 h delayed conditions at ambient temperature (32 ± 2°C) at four stages of salt-curing process for quality parameters and formation of biogenic amines. Moisture content decreased from 76% to 11.3% in salting followed by drying. Similarly, water activity reduced from 0.96 to 0.74 in salt-cured sardines. In contrast, total volatile base nitrogen content increased from 79 to 3,590 mg kg–1 in salt-cured sardines. Halophilic count of sardines was higher (8 log cfu g–1) after salting and decreased in drying stages. The 15 h delayed salt-cured whole sardines had higher histamine contents (1,568.27 mg kg–1), cadaverine (4,059.84 mg kg–1), putrescine (1,604.95 mg kg–1), and tyramine (862.94 mg kg–1) on the final day of drying. Whole sardines had higher biogenic amines and halophilic bacterial counts than the gutted sardines. The whole and gutted sardines salt-cured up to a delay of 6 and 12 h, respectively, at ambient temperature yielded the acceptable quality products with respect to biogenic amines.  相似文献

We isolated the essential oils from Boswellia ovalifoliolata N.P.Balakr and A.N.Henry using hydrodistillation, identified the obtained compounds using gas chromatography–mass spectrometry (GC–MS) and Fourier transform-infrared spectroscopy (FT-IR), and studied the photophysical and electrochemical properties using UV–visible and fluorescence spectroscopy and cyclic voltammetry. On the basis of GC–MS spectra, 38 compounds were identified in the essential oil from leaves and 26 in the oil from bark. The bark oil contained 13.44% β-Farnesene (sesquiterpene), 10.45% caryophyllene oxide (sesquiterpene) and 6.6% spathulenol, (2Z,6E)-(sesquiterpene alcohol); the leaf oil contained 11.1% spathulenol (sesquiterpene alcohol), 9.0% caryophyllene oxide (sesquiterpenoids) and 6.3% decyl acetate (ester). FT-IR spectra confirmed the presence of aliphatic aldehydes and ketones, carboxylic acid, alcohols, esters and ethers in both oil types. UV–visible absorption spectra showed maximum absorbance at 245, 290 and 402 nm for bark oil, and 250, 285 and 325 nm for leaf. Bark oil showed strong emission with maximum emission wavelength at 456 nm was higher than that of leaf oil at 414 nm. The essential oil had significant anti-inflammatory activity, and the reduction potential of the leaf oil was ?0.44 and ?0.56 eV for bark.  相似文献

Black-headed caterpillar, Opisina arenosella Walker (Lepidoptera: Crytophasidae) is an endemic, frequently outbreaking pest of coconut in India and Sri Lanka. It also occurs in Bangladesh and Myanmar. Two larval parasitoid wasps Goniozus nephantidis (Muesebeck) (Bethylidae) and Bracon brevicornis Wesmael (Braconidae), are widely employed, either singly or in combination, for the biological control of O. arenosella in India. The co-occurrence of G. nephantidis and B. brevicornis in the field indicates the potential for competitive interactions which may interfere with pest suppression. In order to study the outcome of competitive interactions between these parasitoids, host larvae were exposed to the parasitoid species either alone or in combination. Females of G. nephantidis and B. brevicornis did not discriminate between unparasitised hosts and hosts parasitised by the other species. Survival of G. nephantidis and B. brevicornis from eggs to adult eclosion was 89.6 and 81.8%, respectively, when larvae of O. arenosella were exposed to these parasitoids individually. When both species were released simultaneously, the survival of G. nephantidis was 86.8% and females of that species chased B. brevicornis females, attempting to bite and sting them. The mean percentage of progeny obtained was significantly higher for G. nephantidis than for B. brevicornis. G. nephantidis dominated due to its parental care behaviour producing 87.0% of progeny and a sex ratio (proportion of females) of 0.79, whereas B. brevicornis produced 84.0% progeny and a sex ratio of 0.36. Although parental care by G. nephantidis females serves to protect the progeny from intruders, it may be disadvantageous from the standpoint of pest suppression under field conditions especially, when the pest population density is high.  相似文献

Candidatus Liberibacter solanacearum”, a phloem-limited and Gram-negative bacterium that is spread from infected to healthy plants by psyllid insect vectors, is an economically important pathogen of solanaceous and carrot crops in the Americas, New Zealand and Europe. Three haplotypes of “Ca. L. solanacearum” have previously been described, two (LsoA and LsoB) in relation to solanaceous crops in the Americas and New Zealand and the third (LsoC) to carrots in Finland. Herein, we describe a fourth haplotype of this ‘Candidatus Liberibacter’ species (LsoD), also associated with carrots, but from Spain and the Canary Islands and vectored by the psyllid Bactericera trigonica. In addition, LsoC was confirmed in carrot and psyllid samples recently collected from Sweden and Norway. Phylogenetic analysis of the 16S rRNA gene suggests that two of the haplotypes, one in the Americas and the other in northern Europe are closer to each other in spite of a large geographic separation and host differences. Furthermore, during this study, potatoes with symptoms of zebra chip disease recently observed in potato crops in Idaho, Oregon and Washington states were analyzed for haplotype and were found to be positive for LsoA. This liberibacter haplotype was found in psyllids associated with the diseased potato crops as well. This finding contrasts with an earlier report of LsoB from psyllids in Washington which came from a laboratory colony originally collected in Texas.  相似文献

Asian cultivated rice shows allelic variation in sodium transporter, OsHKT1;5, correlating with shoot sodium exclusion (salinity tolerance). These changes map to intra/extracellularly-oriented loops that occur between four transmembrane-P loop-transmembrane (MPM) motifs in OsHKT1;5. HKT1;5 sequences from more recently evolved Oryza species (O. sativa/O. officinalis complex species) contain two expansions that involve two intracellularly oriented loops/helical regions between MPM domains, potentially governing transport characteristics, while more ancestral HKT1;5 sequences have shorter intracellular loops. We compared homology models for homoeologous OcHKT1;5-K and OcHKT1;5-L from halophytic O. coarctata to identify complementary amino acid residues in OcHKT1;5-L that potentially enhance affinity for Na+. Using haplotyping, we showed that Asian cultivated rice accessions only have a fraction of HKT1;5 diversity available in progenitor wild rice species (O. nivara and O. rufipogon). Progenitor HKT1;5 haplotypes can thus be used as novel potential donors for enhancing cultivated rice salinity tolerance. Within Asian rice accessions, 10 non-synonymous HKT1;5 haplotypic groups occur. More HKT1;5 haplotypic diversities occur in cultivated indica gene pool compared to japonica. Predominant Haplotypes 2 and 10 occur in mutually exclusive japonica and indica groups, corresponding to haplotypes in O. sativa salt-sensitive and salt-tolerant landraces, respectively. This distinct haplotype partitioning may have originated in separate ancestral gene pools of indica and japonica, or from different haplotypes selected during domestication. Predominance of specific HKT1;5 haplotypes within the 3 000 rice dataset may relate to eco-physiological fitness in specific geo-climatic and/or edaphic contexts.  相似文献

The defect structure of in situ pulsed, laser-deposited, thin films of the high-transition temperature superconductor Y-Ba-Cu-O has been observed directly by atomic resolution electron microscopy. In a thin film with the nominal composition YBa(2)Cu(3)O(7) (123), stacking defects corresponding to the cationic stoichiometry of the 248, 247, and 224 compounds have been observed. Other defects observed include edge dislocations and antiphase boundaries. These defects, which are related to the nonequilibrium processing conditions, are likely to be responsible for the higher critical currents observed in these films as compared to single crystals.  相似文献
