Runs of homozygosity (ROH) are contiguous homozygous regions of the genome. These regions can be used to identify genes associated with traits of economic interest, as well as inbreeding levels. The aim of the present study was to analyse the length and distribution of ROH islands in Gyr cattle and to identify genes within these regions. A population of 173 animals selected for beef production and a population of 291 animals selected for dairy production were used. Differences in the number of short ROH (ROH1-2 Mb) were observed between the two populations, while the number of long ROH (ROH>16 Mb) was similar. ROH islands with the highest incidences (>0.50) overlapped in several segments of the genome in the two populations. The genes identified were associated with milk production, growth, reproduction, immune response and resistance traits. Our results contribute to the understanding of how selection can shape the distribution of ROH and ROH islands within the same breed when animals are selected for different purposes such as dairy or beef production. 相似文献

Many microorganisms from various sources may be present in ejaculates of bulls. This study identified and isolated bacteria from bull sperm samples in a commercial stud and evaluated their resistance to antibiotics. The number of colony-forming units was determined in semen samples collected at distinct steps during freezing and thawing. The minimum inhibitory concentration and the minimum bactericidal concentration were determined for four antibiotics commonly used in commercial studs. A total of 135 microorganisms from 25 genera were isolated. After a sensitivity test, all evaluated microorganisms (n = 55) were resistant to penicillin and most of them were resistant to tylosin and lincomycin (n = 54). Resistance to all tested antibiotics was observed in 22% of all isolates, whereas only 3.9% of the isolates were inhibited by the tested antibiotics at the concentrations recommended by the international legislation. As the isolated microorganisms presented high resistance to frequently used antibiotics, sensitivity tests should be periodically conducted in commercial bull semen studs to prevent the use of contaminated semen in artificial insemination. 相似文献

The aim of this study was to estimate the (co)variance components and breeding values for birthweight (BW) in Nellore cattle by considering or not identical weights that exhibit a high frequency within the contemporary group (CG). A total of 175,258 BW records of Nellore cattle born between 2002 and 2018 were used. The CG was formed by farm, year of birth, sex and feeding regime at birth. CGs with more than 16% of identical BW values were eliminated, generating a data file called BWd. Another file was created without removing these animals (BWt). A mixed linear model was used for statistical analysis, which included fixed and random effects. In both data files analysed, single-trait analysis was performed by Bayesian inference. The mean direct and maternal heritability for BW and the correlation between direct and maternal effects were 0.27, 0.07 and −0.07 for BWt, respectively, and 0.30, 0.093 and −0.07 for BWd. This method should affect the estimation of genetic merits of animals for BW, providing greater safety in the choice of sires. 相似文献

This study aimed to evaluate the correlation of seminal plasma IGF-I and total proteins from peripubertal bulls with semen characteristics and onset of puberty. Sixteen dairy Gyr bulls were separated into early (n = 8) and regular (n = 8) groups, according to the onset of puberty. Semen was monthly collected by electroejaculation from 14 to 26 months of age, and the onset of puberty was retrospectively determined (17.0 ± 1.6 and 19.2 ± 1.5 months, to early and regular group, respectively). Five time points were evaluated (day −60, day −30, day 0, day 30 and day 60 days of puberty), being day 0 considered as beginning of puberty. Scrotal circumference and body weight were also assessed. Semen characteristics were evaluated, the seminal plasma was separated by centrifugation and total protein and IGF-I concentrations were determined. There was no difference between groups to concentration of the seminal plasma total proteins, but we found an interaction between group and age. Seminal plasma IGF-I concentrations were not different between early and regular groups; thus, the data were combined for analysis. Combined IGF-I concentrations were positively correlated with sperm motility and concentration, age, body weight and scrotal circumference. Negative correlation was found between IGF-I concentration and total sperm defects. Increased IGF-I was observed in day −30 and day 0 of puberty in early and regular groups, respectively. Seminal plasma total proteins are involved in peripubertal modifications and IGF-I from Zebus dairy bulls can influence the seminal characteristics and the growth factor increase occurs concomitantly with body growth but cannot be used to define puberty bulls earlier. 相似文献

BackgroundStanding surgery in horses combining intravenous sedatives, analgesics and local anaesthesia is becoming more popular. Ultrasound guided (USG) peribulbar nerve block (PB) has been described in dogs and humans for facial and ocular surgery, reducing the risk of complications versus retrobulbar nerve block (RB).ObjectiveTo describe a technique for USG PB in horse cadavers.MethodsLandmarks and PB technique were described in two equine cadaver heads (Phase 1), with computed tomography (CT) imaging confirming contrast location and spread. In Phase 2, ten equine cadaver heads were randomised to two operators naïve to the USG PB, with moderate experience with ultrasonography and conventional “blind” RB. Both techniques were demonstrated once. Subsequently, operators performed five USG PB and five RB each, unassisted. Contrast location and spread were evaluated by CT. Injection site success was defined for USG PB as extraconal contrast, and for RB intraconal contrast.ResultsSuccess was 10/10 for USG PB and 0/10 for RB (p < 0.001). Of the RB injections, eight resulted in extraconal contrast and two in the masseter muscle (p = 0.47).ConclusionsThe USG PB had a high injection site success rate compared with the RB technique; however, we cannot comment on clinical effect. The USG technique was easily learnt, and no potential complications were seen. The USG PB nerve block could have a wide application for use in horses for ocular surgeries (enucleations, eyelid, corneal, cataract surgeries, and ocular analgesia) due to reduced risk of iatrogenic damage. This study addressed fire‐induced changes in topsoil organic matter (SOM) from a eucalypt plantation in Portugal over 2 years by using three complementary biogeochemical techniques: elemental analysis, analysis of biomarkers from the total extractable lipids (TLE) and solid state 13C nuclear magnetic resonance (NMR) spectroscopy. Direct wildfire effects included a marked decrease in soil total organic carbon (TOC) and total nitrogen (TN) content. However, both contents seem to have recovered during the 2 years. Fire also substantially reduced the TLE, producing noticeable changes in its composition. These included the thermal breakdown and cracking of n‐alkyl compounds. Ratios of short‐to‐long n‐alkanes and n‐fatty acid methyl esters (FAMEs) increased and typical carbon number predominance indices for n‐alkanes (odd‐to‐even) and n‐FAMEs (even‐to‐odd) were altered. Furthermore, the relative abundances of certain markers that are plant‐species specific were modified, especially by decreasing terpenoids such as epiglobulol, ledol and globulol, which are characteristic of Eucalyptus globulus. Other differences observed in the burnt soil were the appearance of levoglucosan, a typical marker for the thermal alteration of polysaccharides, larger relative abundances of lignin‐derived compounds (vanillin and methoxyphenols) and the presence of N‐heteroaromatic structures, which suggested the accumulation of ‘black nitrogen’. The 13C NMR spectra indicated that the wildfire produced a considerable increase in the aromaticity and aromatic condensation of the topsoil SOM. This was reflected in a broadening of the signal of aromatic compounds at the expense of O‐alkyl and alkyl‐C compounds. The continuation of these differences in SOM quality during the 2‐year study suggested a slow recovery of soil properties, possibly influenced by a limited recovery of the vegetation after the fire combined with the fire‐enhanced losses of soil. 相似文献
