This study's objectives were to determine intestinal antimicrobial concentrations in calves administered enrofloxacin or ceftiofur sodium subcutaneously, and their impact on representative enteric bacteria. Ultrafiltration devices were implanted in the ileum and colon of 12 steers, which received either enrofloxacin or ceftiofur sodium. Samples were collected over 48 h after drug administration for pharmacokinetic/pharmacodynamic analysis. Enterococcus faecalis or Salmonella enterica (5 × 105 CFU/mL of each) were exposed in vitro to peak and tail (48 h postadministration) concentrations of both drugs at each location for 24 h to determine inhibition of growth and change in MIC. Enrofloxacin had tissue penetration factors of 1.6 and 2.5 in the ileum and colon, while ciprofloxacin, an active metabolite of enrofloxacin, was less able to cross into the intestine (tissue penetration factors of 0.7 and 1.7). Ceftiofur was rapidly eliminated leading to tissue penetration factors of 0.39 and 0.25. All concentrations of enrofloxacin were bactericidal for S. enterica and significantly reduced E. faecalis. Peak ceftiofur concentration was bactericidal for S. enterica, and tail concentrations significantly reduced growth. E. faecalis experienced growth at all ceftiofur concentrations. The MICs for both organisms exposed to peak and tail concentrations of antimicrobials were unchanged at the end of the study. Enrofloxacin and ceftiofur achieved intestinal concentrations capable of reducing intestinal bacteria, yet the short exposure of ceftiofur in the intestine may select for resistant organisms.  相似文献

In 1988, 315 lambs from 4 sheep (Ovis aries) herds comprised of 1003 animals were marked with silent mortality transmitters in Gammeldalen in Hedmark County, southern Norway. The aim was to identify causes of death and the so-called "hidden loss" of lambs on summer range. A total of 22 animals, 19 lambs and 3 ewes were found dead. Of these, 18 lambs were marked with radio transmitters. A total of 10 animals died of disease (45.5%) and 12 were killed by predators (54.5%). Of the 18 radio-located lambs, 6 died of disease (33.3%) and 12 due to predators (66.7%). Various common lamb diseases were diagnosed, e.g. lung, thoracic and intestinal infections. Eleven lambs were killed by lynx (Lynx lynx), a 12th was found chased to exhaustion, presumably by a predator. Use of mortality transmitters is concluded to be useful in investigating sheep losses on Norwegian ranges.  相似文献

We investigated the requirements for a commercial vitamin premix in diets of Lytechinus variegatus. Small L. variegatus (ca. 28 mm diameter) were collected from Saint Joseph Bay, Florida, and acclimated to laboratory conditions (32 ppt salinity, 22 C, 12:12 photoperiod) for 3 wk, then placed individually into plastic mesh cages (n = 16 per treatment). Sixteen urchins were dissected to obtain initial weights of organs (gut, gonad, test). Each treatment was fed one of five semipurified diets supplemented with levels of a commercial shrimp vitamin premix ranging from 0 to 0.9% dry weight. At 4‐wk intervals, wet weights and test diameters were measured. At 16 wk, urchins were dissected and organs weighed. Significant weight gain and 100% survival was observed with all diets. At 16 wk, urchins fed 0% vitamin premix exhibited no noticeable pathology but had significantly less weight gain than those fed the 0.6% vitamin premix. Those fed the 0.6% vitamin premix had significantly greater weight gain than all other diets (P < 0.0001). Gonad weights increased in all treatments, but did not vary significantly. Under the conditions of this study, L. variegatusexhibited a satisfactory nutritional response. Experiments are planned to determine specific dietary vitamin requirements for L. variegatus.  相似文献

The stable carbon isotope composition (delta(13)C) of foliage integrates signals resulting from environmental and hydraulic constraints on water movement and photosynthesis. We used branch length as a simple predictor of hydraulic constraints to water fluxes and determined the response of delta(13)C to varying water availability. Foliage up to 6 years old was taken from Pinus pinaster Ait. trees growing at four sites differing in precipitation (P; 414-984 mm year(-1)) and potential evaporation (ET; 1091-1750 mm year(-1)). Branch length was the principal determinant of temporal trends in delta(13)C. The strong relationship between delta(13)C and branch length was a function of hydraulic conductance, which was negatively correlated with branch length (r(2) = 0.84). Variation in P and ET among sites was reflected in delta(13)C, which was negatively correlated with P/ET (r(2) = 0.66). However, this analysis was confounded by differences in branch length. If the effects of branch length on delta(13)C were first removed, then the 'residual' delta(13)C was more closely related to P/ET (r(2) = 0.99), highlighting the importance of accounting for variation in hydraulic constraints to water flux between sites and years. For plant species that exhibit considerable phenotypic plasticity in response to changes in environment (e.g., variation in leaf area, branch length and number, or stem form), the environmental effects on delta(13)C in foliage can only be reliably assessed if deconvoluted from hydraulic constraints.  相似文献

Summary The wettability of four Asian meranti species (dark red, light red, white, and yellow) in various liquids was studied by a cone-immersion technique. Wettability was expressed in terms of the area under the force-immersion curve and referred to as the wettability index. Wettability was strongly influenced by the pH of the solution, generally increasing with increasing pH. Surfactant type affected wettability, with the cationic type yielding the greatest wettability followed by anionic and nonionic types. Among the liquids used, distilled water gave the lowest, and acetone-isopropyl alcohol solution gave the greatest wettability. Of the meranti species, yellow meranti showed the greatest wettability, followed by white, light red, and dark red meranti.A plot of the relative amount of liquid absorbed into the wood sample versus wettability index yielded two regions, one attributed mainly to liquid trapped in crevices on the wood surface, and another which may be more indicative of wood-liquid interactions. A highly significant linear correlation between wettability index and liquid surface tension was found for those species with low and medium wettability, while that with high wettability showed no correlation. A new empirical approach of determining the critical surface tension of wood is discussed. Limited data for the present method indicated close agreement with result from the literature obtained by the use of the well-known Zisman's contact angle approach.  相似文献

ABSTRACT Analyses of multiple field experiments indicated that the incidence and relative abundance of root-colonizing phlD+ Pseudomonas spp. were influenced by crop rotation, tillage, organic amendments, and chemical seed treatments in subtle but reproducible ways. In no-till corn plots, 2-year rotations with soybean resulted in plants with approximately twofold fewer phlD+ pseudomonads per gram of root, but 3-year rotations with oat and hay led to population increases of the same magnitude. Interestingly, tillage inverted these observed effects of cropping sequence in two consecutive growing seasons, indicating a complex but reproducible interaction between rotation and tillage on the rhizosphere abundance of 2,4-diacetlyphloroglucinol (DAPG) producers. Amending conventionally managed sweet corn plots with dairy manure compost improved plant health and also increased the incidence of root colonization when compared with nonamended plots. Soil pH was negatively correlated to rhizosphere abundance of phlD+ pseudomonads in no-till and nonamended soils, with the exception of the continuous corn treatments. Chemical seed treatments intended to control fungal pathogens and insect pests on corn also led to more abundant populations of phlD in different tilled soils. However, increased root disease severity generally was associated with elevated levels of root colonization by phlD+ pseudomonads in no-till plots. Interestingly, within a cropping sequence treatment, correlations between the relative abundance of phlD and crop stand or yield were generally positive on corn, and the strength of those correlations was greater in plots experiencing more root disease pressure. In contrast, such correlations were generally negative in soybean, a difference that may be partially explained by difference in application of N fertilizers and soil pH. Our findings indicate that farming practices can alter the relative abundance and incidence of phlD+ pseudomonads in the rhizosphere and that practices that reduce root disease severity (i.e., rotation, tillage, and chemical seed treatment) are not universally linked to increased root colonization by DAPG-producers.  相似文献

Modern felid species descend from relatively recent (<11 million years ago) divergence and speciation events that produced successful predatory carnivores worldwide but that have confounded taxonomic classifications. A highly resolved molecular phylogeny with divergence dates for all living cat species, derived from autosomal, X-linked, Y-linked, and mitochondrial gene segments (22,789 base pairs) and 16 fossil calibrations define eight principal lineages produced through at least 10 intercontinental migrations facilitated by sea-level fluctuations. A ghost lineage analysis indicates that available felid fossils underestimate (i.e., unrepresented basal branch length) first occurrence by an average of 76%, revealing a low representation of felid lineages in paleontological remains. The phylogenetic performance of distinct gene classes showed that Y-chromosome segments are appreciably more informative than mitochondrial DNA, X-linked, or autosomal genes in resolving the rapid Felidae species radiation.  相似文献

Because exercise fatigue has been associated with the accumulation of lactic acid, factors that influence lactate metabolism during exercise can potentially enhance performance. The objective of this study was to examine the effects of supplemental betaine on eight mature Thoroughbred horses before and after 8 wk of conditioning. The effects of betaine were tested in two cross-over design experiments, allowing each horse to receive both the control and betaine treatments at each fitness level. Ingestion of 80 mg of betaine/kg of BW for 14 d before exercise testing did not alter plasma lactate, glucose, free fatty acids (FFA), or triglyceride concentrations during exercise in the untrained or trained horses. A time x treatment interaction (P < .05) was observed for plasma lactate in untrained horses during recovery from exercise, and plasma lactate concentrations were lower (P < .05) at 60 min after exercise when untrained horses received betaine. Plasma FFA concentrations were lower (P < .05) before exercise and at 720 min after exercise when untrained horses received betaine. These data indicate that betaine may influence lactate metabolism following exercise in untrained horses; however, betaine does not seem beneficial for trained horses.  相似文献

The Chlamydiales are a unique order of intracellular bacterial pathogens that cause significant disease of birds and animals, including humans. The recent development of a Chlamydiales-specific 16S rDNA polymerase chain reaction (PCR) assay has enabled the identification of Chlamydiales DNA from an increasing range of hosts and environmental sources. Whereas the Australian marsupial, the koala, has previously been shown to harbour several Chlamydiales types, no other Australian marsupials have been analysed. We therefore used a 16S rDNA PCR assay combined with direct sequencing to determine the presence and genotype of Chlamydiales in five wild Australian mammals (gliders, possums, bilbies, bandicoots, potoroos). We detected eight previously observed Chlamydiales genotypes as well as 10 new Chlamydiales sequences from these five Australian mammals. In addition to PCR analysis we used antigen specific staining and in vitro culture in HEp-2 cell monolayers to confirm some of the identifications. A strong association between ocular PCR positivity and the presence of clinical disease (conjunctivitis, proliferation of the eyelid) was observed in two of the species studied, gliders and bandicoots, whereas little clinical disease was observed in the other animals studied. These findings provide further evidence that novel Chlamydiales infections occur in a wide range of hosts and that, in some of these, the chlamydial infections may contribute to clinical disease.  相似文献
