A multiplex PCR protocol was established to simultaneously detect major bacterial pathogens in olive flounder (Paralichthys olivaceus) including Edwardsiella (E.) tarda, Streptococcus (S.) parauberis, and S. iniae. The PCR assay was able to detect 0.01 ng of E. tarda, 0.1 ng of S. parauberis, and 1 ng of S. iniae genomic DNA. Furthermore, this technique was found to have high specificity when tested with related bacterial species. This method represents a cheaper, faster, and reliable alternative for identifying major bacterial pathogens in olive flounder, the most important farmed fish in Korea.  相似文献

In this study, we determined how rosiglitazone (RSG) differentially affected hippocampal neurogenesis in mice fed a low-fat diet (LFD) or high-fat diet (HFD; 60% fat). LFD and HFD were given to the mice for 8 weeks. Four weeks after initiating the LFD and HFD feeding, vehicle or RSG was administered orally once a day to both groups of mice. We measured cell proliferation and neuroblast differentiation in the subgranular zone of the dentate gyrus using Ki67 and doublecortin (DCX), respectively, as markers. In addition, we monitored the effects of RSG on the levels of DCX and brain-derived neurotrophic factor (BDNF) in hippocampal homogenates. At 8 weeks after the LFD feeding, the numbers of Ki67- and DCX-positive cells as well as hippocampal levels of DCX and BDNF were significantly decreased in the RSG-treated group compared to the vehicle-treated animals. In contrast, the numbers of Ki67- and DCX-positive cells along with hippocampal levels of DCX and BDNF in the HFD fed mice were significantly increased in the RSG-treated mice compared to the vehicle-treated group. Our data demonstrate that RSG can modulate the levels of BDNF, which could play a pivotal role in cell proliferation and neuroblast differentiation in the hippocampal dentate gyrus.  相似文献

A 4‐week feeding trial was conducted to test the use of marine yeasts purified from seawater and sediments as a dietary source in cultivating a cladoceran, Moina macrocopa, a potential alternative live food for fish larvae. Optimal growth conditions of two yeast strains were obtained for NaCl concentration, pH and temperature. Moina macrocopa was cultivated using different diets: marine yeasts (Debaryomyces hansenii Yeast‐14 and Candida austromarina Yeast‐16) and a commercial diet (Erythrobacter sp. Sπ‐I). The essential amino acids of both the yeast strains were nearly as great as those in M. microcopa. Further, the yeast‐fed M. macrocopa had essential amino acid profiles similar to the documented values for rotifers and Artemia enriched in microalgae and commercial diets. Erythrobacter sp. Sπ‐I lacked n‐3 polyunsaturated fatty acids, 20:5n‐3 and 22:6n‐3, which were also low but detected in both yeasts. An increase in the 20:5n‐3 [eicosapentaenoic acid (EPA)] and 22:6n‐3 [docosahexaenoic acid (DHA)] levels, compared with the levels in yeast strains themselves, was more pronounced in the 22:6n‐3 level of Moina fed the C. austromarina, resulting in a high DHA:EPA ratio. When the Moina diets were switched, their δ13C values shifted gradually towards the values of the switched diets. Diet switch from Erythrobacter sp. Sπ‐I to C. austromarina Yeast‐16 resulted in a more rapid turnover of Moina tissue carbon than that in the inverse case. When fed a mixed diet, the δ13C values of Moina tissue approached the value of marine yeasts immediately. These temporal changes in the δ13C values of Moina tissue indicate the preferential ingestion of marine yeasts and a selective assimilation of the carbon originated from marine yeasts. These findings suggest that marine yeasts, particularly C. austromarina Yeast‐16, are highly available to mass cultures of M. macrocopa, providing better nutritional and dietary values than the commercial diet (Erythrobacter sp. A new inhibitor, placotylene A (1), of the receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation, and a regioisomer of placotylene A, placotylene B (2), were isolated from a Korean marine sponge Placospongia sp. The chemical structures of placotylenes A and B were elucidated on the basis of 1D and 2D NMR, along with MS spectral analysis and revealed as an iodinated polyacetylene class of natural products. Placotylene A (1) displayed inhibitory activity against RANKL-induced osteoclast differentiation at 10 μM while placotylene B (2) did not show any significant activity up to 100 μM, respectively.  相似文献

This study was performed to evaluate and optimize a small bowel contrast technique using barium and methylcellulose in dogs. Ten healthy dogs underwent both a conventional upper gastrointestinal study that used only barium and a modified study that used barium and methylcellulose. The control group received 10 mL/kg of 40% barium suspension. Experimental groups were divided into three subgroups given 15 mL/kg of different viscosities (low, moderate, and high viscosity) of 0.5% methylcellulose after 4 mL/kg of 40% barium suspension. Compared with the control group, dogs receiving methylcellulose had higher-quality upper gastrointestinal studies. Moderate viscosity of methylcellulose was superior to the other methylcellulose groups. In conclusion, the use of methylcellulose is a simple and effective method for improving the image quality in an upper GI examination.  相似文献

To evaluate the exposure of Koreans to ochratoxin A, we conducted a survey in 2003 for ochratoxin A in various domestic food commodities: 60 polished rices, 22 barleys, 35 wheat flours, 46 beers, and 14 unstrained rice wine (makkolli) samples. They were analyzed for ochratoxin A using immunoaffinity column and high-performance liquid chromatography (HPLC)-fluorescence detection, and the positive samples were confirmed using HPLC-tandem mass spectrometry. By combining results from different surveys on the levels of ochratoxin A in selected foods and the consumption patterns, we obtained the Korean probable daily intakes (PDI) of ochratoxin A. The polished rice commodity had the highest mean levels of ochratoxin A, which ranged from 0.2 (not detected, i.e., ND = 0) to 1.0 ng/g (ND = limit of detection, i.e., LOD). The estimated PDI for all Koreans fell into the range of 0.8-4.1 ng/kg bw/day, while for heavy consumers the estimates ranged from 1.7 to 9.1 ng/kg bw/day, which did not exceed the PTDI value (14 ng/kg bw/day). Staple rice is the major contributor (>90%) to the Korean dietary intake of ochratoxin A. On the basis of these estimates, it may be concluded that there is at present no considerable risk of ochratoxin A exposure for the average Korean consumer.  相似文献

Characteristic aroma components of water dropwort (Oenanthe javanica DC.) were evaluated by aroma extract dilution analysis and solid-phase microextraction-gas chromatography-olfactometry. Alpha-Terpinolene (plastic/cucumber-like) was the most intense aroma-active compound in water dropwort. Other potent aroma-active compounds included p-cymene (kerosene-like), alpha-terpinene (lemon), (E)-caryophyllene (woody), (Z,E)-alpha-farnesene (woody), hexanal (green), (Z)-3-hexenol (green), phenylacetaldehyde (honey), (E)-2-nonenal (cucumber), bornyl acetate (cooked vegetable), and gamma-terpinene (lemon). Of these, p-cymene was believed to be primarily responsible for the distinct kerosene-like aroma note of water dropwort. The aroma property of p-cymene was dependent on its concentration and was described as kerosene-like at relatively high concentrations but changed to citrus and green aroma notes at low concentrations.  相似文献

Single- and bi-solute sorption and desorption of 2,4-dichlorophenol (2,4-DCP) and 2,4,5-trichlorophenol (2,4,5-TCP) in montmorillonite modified with hexadecyltrimethylammonium (HDTMA) were investigated using multi-step sorption and desorption procedure. Effect of pH on the multi-step sorption and desorption was investigated. As expected by the magnitude of octanol-water partition coefficient, K ow , both sorption and desorption affinity of 2,4,5-TCP was higher than that of 2,4-DCP at pH 4.85 and 9.15. For both chlorophenols, the protonated speciation (at pH 4.85) exhibited a higher affinity in both sorption and desorption than the predominant deprotonated speciation (about 95% and 99% of 2,4-dichlorophenolate and 2,4,5-trichlophenolate anions at pH 9.15, respectively). Desorption of chlorinated phenols was strongly dependent on the current pH regardless of their speciation in the previous sorption stage. Freundlich model was used to analyze the single-solute sorption and desorption data. No appreciable desorption-resistant (or non-desorbing) fraction was observed in organoclays after several multi-step desorptions. This indicates that sorption of phenols in organoclay mainly occurs via partitioning into the core of the pseudo-organic medium, thereby causing desorption nearly reversible. In bisolute competitive systems, sorption (or desorption) affinity of both chlorophenols was reduced compared to that in its single-solute system due to the competition between the solutes. The ideal adsorbed solution theory (IAST) coupled to the single-solute Freundlich model successfully predicted bisolute multi-step competitive sorption and desorption equilibria.  相似文献
