Trypsin from the pyloric ceca of masu salmon (Oncorhynchus masou) cultured in fresh water was purified by a series of chromatographies including Sephacryl S-200, Sephadex G-50 and diethylaminoethyl cellulose to obtain a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS–PAGE) and native PAGE. The molecular mass of the purified trypsin was estimated to be approximately 24,000 Da by SDS–PAGE. The enzyme activity was strongly inhibited by phenylmethylsulfonyl fluoride, soybean trypsin inhibitor, and N α -p-tosyl-l-lysine chloromethyl ketone. Masu salmon trypsin was stabilized by calcium ion. The optimum pH of the masu salmon trypsin was around pH 8.5, and the trypsin was unstable below pH 5.0. The optimum temperature of the masu salmon trypsin was around 60°C, and the trypsin was stable below 50°C, like temperate-zone and tropical-zone fish trypsins. The N-terminal 20 amino acid sequence of the masu salmon trypsin was IVGGYECKAYSQPHQVSLNS, and its charged amino acid content was lower than those of trypsins from frigid-zone fish and similar to those of trypsins from temperate-zone and tropical-zone fish. In the phylogenetic tree, the masu salmon trypsin was classified into the group of the temperate-zone fish trypsin.  相似文献

Fifteen healthy castrated male dogs were separated into three treatment groups that were administered etodolac, aspirin and a placebo orally, respectively. All treatments were continued for 28 days. The animals were examined endoscopically on days 0, 3, 7, 10, 14, 17, 21, 24 and 28. There were no gastrointestinal mucosal lesions in either the etodolac or the placebo group, whereas some gastric lesions developed in the aspirin group after day 17. We considered that etodolac could be used for long-term treatments in dogs with fewer side-effects on the gastric mucosa than aspirin.  相似文献

As neglected zoonosis for many years, Q fever is now ubiquitous in Japan. Similarly to elsewhere in the world, domestic animals are considered to be important reservoirs of the causal agent, Coxiella burnetii, a resistant intracellular bacterium. Infected animals shed bacteria in milk, feces, urine, vaginal mucous and birth products. Inhalation of bacteria present in the environment is the main route of animal and human infection. Shedding of C. burnetii in milk by domestic ruminants has a very limited impact as raw milk is seldom ingested by the Japanese population. The clinical expression of Q fever in Japan is similar to its clinical expression elsewhere. However clinical cases in children are more frequently reported in this country. Moreover, C. burnetii is specified as one of the causative organisms of atypical pneumonia in the Japanese Respiratory Society Guideline for the management of community-acquired pneumonia. In Japan, C. burnetii isolates are associated with acute illness and are mainly of moderate to low virulence. Cats are considered a significant source of C. burnetii responsible for human outbreaks in association with the presence of infected parturient cats. Since its recognition as a reportable disease in 1999, 7-46 clinical cases of Q fever have been reported by year. The epidemiology of Q fever in Japan remains to be elucidated and the exact modes of transmission are still unproven. Important further research is necessary to improve knowledge of the disease itself, the endogenous hosts and reservoirs, and the epidemiological cycle of coxiellosis in Japan.  相似文献

Interspecific hybridization is a useful technique to introduce characteristics from wild species into crops. Garden asparagus (Asparagus officinalis, 2n = 2x = 20) is an economically important vegetable that is native to the Mediterranean region but widely cultivated in many countries. The genus Asparagus is comprised of over 100 species. Asparagus kiusianus (2n = 2x = 20) is a wild asparagus species endemic to Japan. This species occurs on the coast and is likely to be a salt-resistant species. Although the geographic distribution of these two species is not close, molecular phylogenetic analysis demonstrated that these two species are closely related. In this study, a reciprocal cross between A. officinalis and A. kiusianus was carried out by hand pollination, and progeny were obtained from both crossings. These progeny exhibited a morphologically intermediate phenotype in terms of flower shape, and restriction fragment length polymorphism analysis confirmed that these were indeed interspecific hybrids. The interspecific hybrids were fertile, and backcross progeny with garden asparagus was also generated. These interspecific hybrids are expected to supply novel traits to garden asparagus.  相似文献

With glass capillary pressure vessels it has been possible to study the effect of pressure on the temperature of maximum density (TMD) and on the "sharpness" of the density maximum in liquid H(2)O and D(2)O in the important but little-studied supercooled regime. A pressure of 1200 bars produces a 33 degrees C depression of the TMD in these liquids and a considerable reduction in sharpness. Comparison with the rather flat density maximum for liquid SiO(2) supports the notion that the presence or absence of density anomalies in "tetrahèdral" liquids depends on the average bridge-bond angle, which is evidently unusually large in water at normal pressure.  相似文献

Recently, micropedological investigations have become an important means to explain some pedogenetical processes. The micromorphological features of Brown Forest soils and Mountain Brown Forest soils have been reported by several investigators(12, 14–15), Braunerde can be distinguished from Braunlehm by the presence of a spongy structure, immobility of peptizable iron hydroxides, or by the absence of any fluidal structure in its (B) horizon. In his review on micropedology, osmond(16) has stated that Brown Earths do not show any evidence of illuviation of ferruginous compounds or clay formed by oxidative weathering. As a result of micromorphological investigations on soils derived from serpentine in Greece, Krause(12) has described that the dominant soil of the hillsides is rubefied Braunlehm (Rotlehm), whereas the soil developed on valley terraces is Braunerde which appears as a transformation product of Rotlehm and has a spongy fabric in the A horizon.  相似文献
