High throughput DNA isolation from plants is a major bottleneck for most studies requiring large sample sizes. A variety of protocols have been developed for DNA isolation from plants. However, many species, including conifers, have high contents of secondary metabolites that interfere with the extraction process or the subsequent analysis steps. Context

In the interior Northwest, debate over restoring mixed-conifer forests after a century of fire exclusion is hampered by poor understanding of the pattern and causes of spatial variation in historical fire regimes.

Objectives

To identify the roles of topography, landscape structure, and forest type in driving spatial variation in historical fire regimes in mixed-conifer forests of central Oregon.

Methods

We used tree rings to reconstruct multicentury fire and forest histories at 105 plots over 10,393 ha. We classified fire regimes into four types and assessed whether they varied with topography, the location of fuel-limited pumice basins that inhibit fire spread, and an updated classification of forest type.

Results

We identified four fire-regime types and six forest types. Although surface fires were frequent and often extensive, severe fires were rare in all four types. Fire regimes varied with some aspects of topography (elevation), but not others (slope or aspect) and with the distribution of pumice basins. Fire regimes did not strictly co-vary with mixed-conifer forest types.

Conclusions

Our work reveals the persistent influence of landscape structure on spatial variation in historical fire regimes and can help inform discussions about appropriate restoration of fire-excluded forests in the interior Northwest. Where the goal is to restore historical fire regimes at landscape scales, managers may want to consider the influence of topoedaphic and vegetation patch types that could affect fire spread and ignition frequency.

Context

The roosting habits of many temperate zone bats are well documented at microhabitat scales, but fewer studies have included multi-scale assessments of landscape patterns in bat roost site selection.

Objectives

To identify and assess at the landscape-scale the location of spring and early season maternity roosts of female northern long-eared bats (Myotis septentrionalis) from 2015 to 2016 at Mammoth Cave National Park (MACA), Kentucky, USA.

Methods

We used mist-nets and radiotelemetry to catch and track bats to roost trees across the landscape of MACA. Data on roosting sites were evaluated using spatial point pattern analysis to examine distributional trends of roosts. A variety of spatial covariates were used to model the effect of landscape pattern, including: forest type, elevation, and proximity to hibernacula, water, and road corridors.

Results

Data indicate that roost locations of female northern long-eared bats in MACA were typically situated within 2000 m of known winter hibernacula, occurring more often at higher elevations in mesic upland deciduous forests, and in close proximity to water sources and roads. We present hypotheses to account for the patterns observed in relation to landscape features and habitat resources in the Park.

Conclusions

Our data indicate that a more comprehensive understanding of habitat requirements which includes empirically-based, landscape-scale patterns, and not solely considerations at stand or local levels, could lead to better informed management policies targeting conservation of maternity habitat of forest-dwelling bats, including the northern long-eared bat, a species in decline throughout much of its distribution in North America.

Context

Landscape and habitat filters are major drivers of biodiversity of small habitat islands by influencing dispersal and extinction events in plant metapopulations.

Objectives

We assessed the effects of landscape and habitat filters on the species richness, abundance and trait composition of grassland specialist and generalist plants in small habitat islands. We studied traits related to functional spatial connectivity (dispersal ability by wind and animals) and temporal connectivity (clonality and seed bank persistence) using model selection.

Methods

We sampled herbaceous plants, landscape (local and regional isolation) and habitat filters (inclination, woody encroachment and disturbance) in 82 grassland islands in Hungary.

Results

Isolation decreased the abundance of good disperser specialist plants due to the lack of directional vectors transferring seeds between suitable habitat patches. Clonality was an effective strategy, but persistent seed bank did not support the survival of specialist plants in isolated habitats. Generalist plants were unaffected by landscape filters due to their wide habitat breadth and high propagule availability. Clonal specialist plants could cope with increasing woody encroachment due to their high resistance against environmental changes; however, they could not cope with intensive disturbance. Steep slopes providing environmental heterogeneity had an overall positive effect on species richness.

Conclusions

Specialist plants were influenced by the interplay of landscape filters influencing their abundance and habitat filters affecting species richness. Landscape filtering by isolation influenced the abundance of specialist plants by regulating seed dispersal. Habitat filters sorted species that could establish and persist at a site by influencing microsite availability and quality.

Context

While remote sensing imagery is effective for quantifying land cover changes across large areas, its utility for directly assessing the response of animals to disturbance is limited. Soundscapes approaches—the recording and analysis of sounds in a landscape—could address this shortcoming.

Objectives

In 2011, a massive wildfire named “the Horseshoe 2 Burn” occurred in the Chiricahua National Monument, Arizona, USA. We evaluated the impact of this wildfire on acoustic activity of animal communities.

Methods

In 2013, soundscape recordings were collected over 9 months in 12 burned and 12 non-burned sites in four ecological systems. The seasonal and diel biological acoustic activity were described using the “Bioacoustic Index”, a detailed aural analysis of sound sources, and a new tool called “Sonic Timelapse Builder” (STLB).

Results

Seasonal biophony phenology showed a diurnal peak in June and a nocturnal peak in October in all ecological systems. On June mornings, acoustic activity was lower at burned than at non-burned sites in three of four ecological systems, due to a decreased abundance of cicadas directly impacted by the death of trees. Aural analyses revealed that 55% of recordings from non-burned sites contained insect sounds compared to 18% from burned sites. On October nights, orthopteran activity was more prevalent at some burned sites, possibly due to post-fire emergence of herbaceous.

Conclusions

Soundscape approaches can help address long-term conservation issues involving the responses of animal communities to wildfire. Acoustic methods can serve as a valuable complement to remote sensing for disturbance-based landscape management.

Context

Freshwater ecosystems depend on surrounding terrestrial landscape for resources. Most important are terrestrial leaf litter subsidies, which differ depending on land use. We lack a good understanding of the variation of these inputs across spatial scales.

Objectives

We sought to determine: (1) the relative importance of local versus catchment-level forestation for benthic leaf litter biomass in streams, (2) how landscape configuration alters these relationships, and (3) how land use affects the quality and diversity of leaf litter subsidies.

Methods

We measured biomass and identity of benthic leaf litter in 121 reaches in 10 independent catchments seasonally over the course of a year. We assessed direct and indirect effects of forestation, reach position, and seasonality on leaf litter biomass using structural equation models, and assessed how leaf litter diversity varied with land use.

Results

In catchments with forested headwaters, the degree of forestation and reach position in the catchment influenced benthic leaf litter biomass indirectly through local reach-scale forestation. In catchments where forest was only located downstream, or with minimal forest, none of these factors influenced reach-level benthic leaf litter. Leaf litter diversity peaked in fall in all land use types, but was generally lowest in forested reaches.

Conclusions

Not only habitat amount, but its location relative to other habitats is important for ecosystem function in the context of cross-ecosystem material flows. Here, lack of upstream forest altered spatial patterns of leaf litter storage. Studies with high spatiotemporal resolution may further reveal effects of landscape configuration on other ecosystems.

Context

Common species important for ecosystem restoration stand to lose as much genetic diversity from anthropogenic habitat fragmentation and climate change as rare species, but are rarely studied. Salt marshes, valuable ecosystems in widespread decline due to human development, are dominated by the foundational plant species black needlerush (Juncus roemerianus Scheele) in the northeastern Gulf of Mexico.

Objectives

We assessed genetic patterns in J. roemerianus by measuring genetic and genotypic diversity, and characterizing population structure. We examined population connectivity by delineating possible dispersal corridors, and identified landscape factors influencing population connectivity.

Methods

A panel of 19 microsatellite markers was used to genotype 576 samples from ten sites across the northeastern Gulf of Mexico from the Grand Bay National Estuarine Research Reserve (NERR) to the Apalachicola NERR. Genetic distances (FST and Dch) were used in a least cost transect analysis (LCTA) within a hierarchical model selection framework.

Results

Genetic and genotypic diversity results were higher than expected based on life history literature, and samples structured into two large, admixed genetic clusters across the study area, indicating sexual reproduction may not be as rare as predicted in this clonal macrophyte. Digitized coastal transects buffered by 500 m may represent possible dispersal corridors, and developed land may significantly impede population connectivity in J. roemerianus.

Conclusions

Results have important implications for coastal restoration and management that seek to preserve adaptive potential by sustaining natural levels of genetic diversity and conserving population connectivity. Our methodology could be applied to other common, widespread and understudied species.
