We examined the relationship between the time elapsed after human chorionic gonadotropin (hCG) administration and developmental stage of porcine embryos after collection. Prepubertal gilts, 7 to 8 months old, were given 1500 IU equine chorionic gonadotropin (eCG) intramuscularly, followed by 500 IU hCG 72 h later. The treated gilts were inseminated artificially on Day 1 (Day 0=the day of hCG administration) and on Day 2. Embryos were collected surgically on Day 6 (140, 144, and 147 h after hCG administration) or on Day 7 (164, 168, and 171 h), and the developmental stages of the collected embryos were examined. From 75.2% (276/367) of the prepubertal gilts treated with hormones, we collected an average of 20.7 embryos per gilt with normal morphology. At 140 h after hCG administration, morulae (54.4%) could be collected. At 144 h, morulae and early blastocysts (57.7% and 28.9%, respectively) were collected. By 147 h, the proportion of embryos at the blastocyst to expanded blastocyst stages had increased (10.0%). From 164 h to 171 h, expanding or expanded blastocysts of more than 200 microm in diameter and hatched blastocysts could be collected. The proportion of hatched blastocysts increased from 3.2% (164 h) to 41.0% (171 h). These results suggests that although the number of ovulations differed among gilts, porcine embryos at the appropriate stages can be collected efficiently by controlling the time elapsed between hCG administration and embryo collection.  相似文献

To determine lignin content in triticale and wheat straws, calibration models were built using Fourier transform mid-infrared spectroscopy combined with partial least-squares regression. The best model for triticale and wheat straws was built using averaged spectra with raw spectrum in spectrum format and constant in path length as spectral pretreatments. The values of r(2), root-mean-square error of prediction (RMSEP), and residual predictive deviation (RPD) for the triticale straw model were 0.935, 0.305, and 3.89, respectively. The r(2), RMSEP, and RPD values for the wheat straw model were 0.985, 0.163, and 8.50, respectively. Both models showed good predictive ability. A model built using both triticale and wheat straws indicated that the values of r(2), RMSEP, and RPD were 0.952, 0.27, and 4.63, respectively. This model also showed good predictive ability and could predict lignin contents in triticale and wheat straws with the same high accuracy.  相似文献

Fine roots play a key role in carbon and nutrient dynamics in forested ecosystems. Fine-root dynamics can be significantly affected by forest management practices such as thinning, but research on this topic is limited. This study examined dynamics of fine roots <1 mm in diameter in a 10-year-old stand of hinoki cypress (Chamaecyparis obtusa) for 3 years following thinning (65% in basal area). Fine-root production and mortality rates were estimated using a minirhizotron technique in combination with soil coring. In both thinned and un-thinned control plots, fine-root elongation occurred from early spring to winter (March to December) and fluctuated seasonally. In the thinned and the control plots, the annual fine-root production rates were estimated to be 101 and 120 g m−2 year−1, respectively, whereas the estimated annual fine-root mortality rates were 77 and 69 g m−2 year−1, respectively. At 3 years after thinning, live fine-root biomass was significantly smaller in the thinned plot (143 g m−2) than in the control plot (218 g m−2), whereas dead fine-root biomass was not (147 and 103 g m−2, respectively). Morphological and physiological indices of fine roots such as diameter, specific root length, and root tissue density of the live fine roots was similar in both plots. These results suggested that thinning tended to decrease biomass and production of fine roots, but the effects on characteristics of fine roots would be less evident.  相似文献

Chlamydophila psittaci is the causative agent of human psittacosis and avian chlamydiosis. This zoonotic pathogen is frequently transmitted from infected birds to humans. Therefore proper and rapid detection of C. psittaci in birds is important to control this disease. We developed a method for detecting C. psittaci by using SYBR Green Real-time PCR based on targeting the cysteine-rich protein gene (envB) of C. psittaci. This one step procedure was highly sensitive and rapid for detection and quantification of C. psittaci from fecal samples. This assay was also able to detect other zoonotic Chlamydophila species such as C. abortus and C. felis. The assay is well suited for use as a routine detection method in veterinary medicine.  相似文献

Objective— To determine the effect of humeral wedge and humeral slide osteotomies on force distribution between the articular surfaces of the humerus and the radius and ulna in normal canine thoracic limbs.
Study Design— In vitro mechanical testing.
Sample population— Cadaveric canine right thoracic limbs (n=12).
Methods— Transarticular elbow force maps were measured using a tactile array pressure sensor in elbow joints of axially aligned limbs under 200 N axial load before and after humeral wedge and humeral slide osteotomies.
Results— Loading induced 2 distinct areas of high forces that corresponded with the proximal articular surfaces of the radius and ulna. Mean force on the proximal articular surface of the ulna was reduced by 25% and 28% after 4 and 8 mm sliding osteotomies, respectively. Statistically significant differences were not observed for the wedge osteotomies.
Conclusion— Humeral slide osteotomy significantly decreases force on the proximal articular surface of the ulna.
Clinical Relevance— The proximal articular surface of the ulna contributes significantly to load transfer through the canine elbow joint. Abnormalities that significantly increase this force might contribute to canine elbow dysplasia, specifically fragmentation of the medial coronoid process and osteochondritis dissecans of the medial aspect of the humeral condyle. Under the conditions studied, the overall reduction in mean joint surface force across the proximal articular surface of the ulna after humeral slide osteotomy indicates that this technique merits further investigation for potential use in medial compartmental osteoarthritis of the canine elbow joint.  相似文献

Damage caused by Orobanchaceae root parasitic weeds is a substantial agricultural problem for global food security. Many studies have been conducted to establish practical methods of control, but efforts are still required for successful management. Seed germination of root parasitic weeds requires host-derived germination stimulants including strigolactones (SLs). Studies on SLs have revealed that a butenolide ring is the essential moiety for SL activity as a germination stimulant. Interestingly, recent studies have revealed that butenolide hormones regulate the biosynthesis of secondary metabolites and mediate communication in actinomycete bacteria. Because of the structural similarity between SLs and the bacterial butenolides, we evaluated the germination stimulatory activity of butenolides isolated from Streptomyces albus J1074 on root parasitic weeds. These butenolides were found to specifically induce seed germination of Orobanche minor. Our findings contribute to understanding the molecular mechanisms of germination stimulant perception and to the development of a method for their biological control.  相似文献

Oilseed rape (Brassica napus L.) response to root temperature regimes (20/20, 16/8 and 12/12°C day/night) at constant 20°C air temperature was studied. At each regime, three NO3 :NH4 + ratios (10:0, 8:2, or 6:4), at constant 10 mM N, in the irrigation solution were tested. Plant growth, transpiration, ionic composition and level of cytokinins and gibberellins in the xylem exudate were monitored. The two low root temperature regimes, 12/12 and 16/8°C, reduced rape shoot growth by 28 and 22%, and increased the accumulation of soluble carbohydrates by 42 and 26% in the roots, respectively, as compared to the 20/20°C regime. Low root temperatures reduced plants transpiration. The NO3 :NH4 + ratios had no effect on rape growth. At low root temperatures NO3 contents increased in the shoot and decreased in the roots. The sum of cations and that of anions at 12/12 and 16/8°C root temperatures decreased significantly as compared to 20/20°C. The presence of NH4 + in the irrigation solution decreased the concentrations of Ca2+ and Mg2+ in the shoots and roots and increased that of Cl in the shoots and of H2PO4 in the roots at all root temperatures. Cytokinins and gibberellins contents in the xylem exúdate decreased at the low root temperature regimes. Low root temperature reduced total upward transport of the mineral nutrients and phytohormones, most probably because of reduced water flow through the plant.  相似文献

Physical, chemical, and biological properties of S-allylcysteine (SAC) were investigated. SAC showed stable properties under tested conditions, and its acute/subacute toxicity was very minor in mice and rats (LD(50) value >54.7 mM/kg po; >20 mM/kg ip). The pharmacokinetics of SAC was investigated after oral administration of garlic supplement containing SAC to human volunteers. SAC from garlic consumption was rapidly absorbed from the gastrointestinal tract, however, the half-life and excretion time were more than 10 h and 30 h, respectively.  相似文献

We measured seasonal variation in area-based nitrogen concentration (N), maximum rate of carboxylation (Vcmax) and maximum rate of electron transport (Jmax) in 1-year-old needles along four first-order branches within a Pinus densiflora Sieb. & Zucc. crown, and analyzed their relationships to growth irradiance and temperature. Each leaf light environment was expressed as a ratio of the monthly mean of daily integrated photosynthetically active irradiance (Iint) for the particular needle to Iint above the canopy (Irel). Needle N decreased in the upper crown during the development of new needles, whereas it remained fairly constant in the lower crown, reflecting differences between upper and lower crown needles in their contribution to the nitrogen of new needles. Gradients of N within the crown were correlated with Irel in all seasons (r2 = 0.40-0.78). Seasonal variation in N was weakly correlated with mean daily air minimum temperatures. Both Vcmax and Jmax showed seasonal variation in all first-order branches, and decreased to their lowest values in winter. The gradients of Vcmax and Jmax within the crown were not correlated with Irel in some seasons, but were correlated with changes in N in most months (r2 = 0.33-0.75), except in the winter. Furthermore, the regression slope of the relationship between N and Vcmax and the temperature response of Vcmax and Jmax exhibited seasonal variation.  相似文献
