The aqueous extract of Rhizophora mangle bark and its polyphenolic fractions showed remarkable in vitro antiinflammatory activity in a preliminary study. The low molecular weight fraction exhibited cyclooxygenase-2 inhibitory activity while the total aqueous extract and the low molecular weight fraction showed secretory phospholipase A(2) inhibitory activity.  相似文献

ABSTRACT In the past 10 years, the winegrape industry in the southeastern United States has experienced rapid growth; however, further expansion may be inhibited by Pierce's disease (PD). Epidemiological studies were conducted to identify the primary vectors of Xylella fastidiosa, the cause of PD of grape, by surveying sharpshooter population dynamics in the eastern Piedmont and Coastal Plain regions of North Carolina. Sharpshooter species were assessed for the presence of X. fastidiosa in the field. Leafhoppers were trapped in three vineyards in the eastern Piedmont and one vineyard in the northeastern Coastal Plain in 2004 and 2005. Four insects were identified as most abundant: Oncometopia orbona, Graphocephala versuta, Paraphlepsius irroratus, and Agalliota constricta. Adult specimens of O. orbona, G. versuta, and P. irroratus were tested for the presence of X. fastidiosa by nested polymerase chain reaction. In all, 27% of O. orbona, 28% of G. versuta, and 33% of P. irroratus trapped were positive for X. fastidiosa over the two seasons. Transmission experiments demonstrated that both O. orbona and G. versuta have the ability to transmit X. fastidiosa to grape. These vectors are likely to be important in all winegrowing regions of the Southeast, because their presence has been documented throughout the southern states. In DNA analyses, X. fastidiosa strains from insects trapped in North Carolina were genetically similar to one another and to the known "PD strain" from California. This is the first report of these two leafhopper species transmitting X. fastidiosa to grapevines in the Southeast.  相似文献

In extremely acidic mining sediments of the Lusatian mining district, the alkalinisation process relies on organic C, which can serve as electron donor for microbially induced sulfate reduction. Plant material of the pioneer plant Juncus bulbosus is an important organic matter source in lake sediments. Therefore, decomposition of the plant tissue was assessed during the exposure of litterbags for 30 months in the 0-5 cm layer of waterlogged mining sediments, which have a pH between 2.5 and 3. The ash free dry weight (AFDW) and elemental content of the plant tissue were recorded several times during the exposure. Changes in chemical structure were analyzed by solid-state 13C cross polarization magic angle spinning nuclear magnetic resonance (CPMAS NMR) spectroscopy and the lignin component characterized by wet-chemical CuO oxidation. The AFDW accounted for about 34% of initial biomass after field exposure for 30 months. Mass loss of biomass occurred in two phases with decomposition rates varying between 30 and 430 mg AFDW d−1. The mass loss increased considerably after 5-7 months when litterbags were invaded by fresh J. bulbosus plants. With respect to higher mass loss, 13C CPMAS NMR spectroscopy, showed slight changes of the bulk chemical composition after 11 months, indicating that microorganisms present in the sediments or in the rhizosphere degrade plant material as a whole, rather than selectively. During the second phase from about 11 months until the end of the exposure period, contribution of O-alkyl C most probably assignable to easily degradable polysaccharides decreased. In contrast, the contribution of alkyl, aromatic and carboxyl C increased. CuO oxidation showed that the lignin component of J. bulbosus is degraded oxidatively during field exposure. Our results indicate that the exposed plant material is decomposed in the sediment due to changes in sediment conditions that followed plant invasion of the litterbags. It is suggested that the rhizosphere of J. bulbosus by its influence on the redox potential, pH and the microbial component plays a crucial role in organic matter degradation in acidic mining sediments.  相似文献

The polychlorinated dibenzo-p-dioxin (PCDD), dibenzofuran (PCDF), and polychlorinated biphenyl (PCB) contents of 123 Spanish commercial salmon, tuna fish, sardine, oyster, mussel, and clam samples from 1995 to 2003 were investigated. A significant decrease of dioxin and non-ortho PCB concentrations in the studied species was found over the years. The decrease was greater in the case of dioxins than in that of non-ortho PCBs, especially during the early years of the study. PCB and PCDD/F concentrations in the years 2001-2003 were comparable to those reported in the literature for similar species collected after 1999. Mean PCB concentrations ranged from 3.46 ng/g of fresh weight (fw) in clams to 100 ng/g of fw in tuna fish. PCDD/F mean current levels ranged from 0.62 pg/g of fw in clams to 2.89 pg/g of fw in oysters. Toxic equivalent quantities (WHO-TEQ) ranged from 0.05 pg of WHO-TEQ(PCDD/Fs)/g of fw in clams to 0.5 pg of WHO-TEQ(PCDD/Fs)/g of fw in salmon (in the upper bound determination levels). When coplanar PCBs were included, the WHO-TEQ(PCDD/Fs+cop) (PCBs) values increased by a range of 1.7 times in oysters to 14.1 times in tuna fish. The decrease in dioxin concentrations suggests that efforts to control dioxin emissions and to reduce human exposure through foodstuffs are succeeding. The high contribution of PCBs to total WHO-TEQs in the fish and shellfish species investigated suggests that it is important to determine PCBs in foodstuffs, and especially in fish products, and they should be included in further research and future legislation.  相似文献

To produce monoclonal antibodies (MAbs) to the pesticide carbofuran, three compounds with carboxylic spacer arms of different lengths introduced at the carbamate group of the analyte structure were synthesized, conjugated to proteins, and used as immunizing haptens in mice. MAbs were subsequently characterized for affinity and specificity in the conjugate-coated format and in the antibody-coated format using newly synthesized compounds as heterologous assay haptens. Depending on the immunoreagent combination and assay format, competitive assays with I(50) values in the 1.2-10.2 nM (0.27-2.27 ng/mL) range were obtained. LIB-BFNB67 MAb in combination with the hapten BFNH, coupled either to horseradish peroxidase or to ovalbumin, was used to develop a direct and an indirect enzyme-linked immunosorbent assay, respectively. Optimized immunoassays displayed very similar analytical characteristics, with an I(50) value around 0.7 ng/mL and a limit of detection around 0.08 ng/mL. Both immunoassays were able to tolerate the presence of methanol up to a 15% concentration. Compounds very similar in structure to carbofuran (benfuracarb, furathiocarb, bendiocarb, and carbofuran-hydroxy) exhibited cross-reactivity values in the 18-37% range, but major N-methylcarbamate pesticides were not recognized by the MAb. These immunoassays should reasonably allow the rapid, low-cost, and sensitive determination of carbofuran in food, in soils, and in the environment at levels of regulatory and practical importance.  相似文献

The N-methylcarbamate pesticide carbaryl is one of the most important insecticides used worldwide. In the present work, the validation of a monoclonal antibody-based enzyme immunoassay (ELISA) for the determination of this compound in fruits and vegetables is described. The immunoassay is a competitive heterologous ELISA in the antibody-coated format, with an I(50) value for standards in buffer of 101.0 +/- 26.9 ng/L and with a dynamic range between 31.6 and 364.0 ng/L. For recovery studies, peppers, cucumbers, strawberries, tomatoes, potatoes, oranges, and apples were spiked with carbaryl at 10, 50, and 200 ppb. After liquid extraction, analyses were performed by ELISA on both extracts purified on solid-phase extraction (SPE) columns and crude, nonpurified extracts. Depending on the crop and the fortification level, recoveries in the 59.0--120.0% range were obtained for purified samples and in the 70.0--137.7% range for crude extracts. The carbaryl immunoassay performance was further validated with respect to high-performance liquid chromatography (HPLC) with postcolumn derivatization and fluorescence detection (EPA Method 531.1). Samples were spiked with carbaryl at several concentrations and analyzed as blind samples by ELISA and HPLC after SPE cleanup. The correlation between methods was excellent (y = 1.04x + 0.71, r(2) = 0.992, n = 33), with HPLC being more precise than ELISA (mean coefficients of variation of 5.2 and 12.0%, respectively). The immunoassay was then applied to the analysis of nonpurified extracts of the same samples. Results also compared very well with those obtained by HPLC on purified samples (y = 1.28x - 0.59, r(2) = 0.987, n = 33) while maintaining similar precision. Therefore, the developed immunoassay is a suitable method for the quantitative and reliable determination of carbaryl in fruits and vegetables even without sample cleanup, which saves time and money and considerably increases sample throughput.  相似文献

ABSTRACT: An experimental infection with highly pathogenic avian influenza virus (HPAIV) and low pathogenic avian influenza virus (LPAIV) was carried out in red-legged partridges (Alectoris rufa) in order to study clinical signs, gross and microscopic lesions, and viral distribution in tissues and viral shedding. Birds were infected with a HPAIV subtype H7N1 (A/Chicken/Italy/5093/1999) and a LPAIV subtype H7N9 (A/Anas crecca/Spain/1460/2008). Uninoculated birds were included as contacts in both groups. In HPAIV infected birds, the first clinical signs were observed at 3 dpi, and mortality started at 4 dpi, reaching 100% at 8 dpi. The presence of viral antigen in tissues and viral shedding were confirmed by immunohistochemistry and quantitative real time RT-PCR (qRRT-PCR), respectively, in all birds infected with HPAIV. However, neither clinical signs nor histopathological findings were observed in LPAIV infected partridges. In addition, only short-term viral shedding together with seroconversion was detected in some LPAIV inoculated animals. The present study demonstrates that the red-legged partridge is highly susceptible to the H7N1 HPAIV strain, causing severe disease, mortality and abundant viral shedding and thus contributing to the spread of a potential local outbreak of this virus. In contrast, our results concerning H7N9 LPAIV suggest that the red-legged partridge is not a reservoir species for this virus.  相似文献

A calcrete profile developed on the top of a calcareous consolidated dune located in a coastal area of NE Tunisia (semi-arid climate) was studied with the aim to investigate the behavior of the chemical elements (rare earth elements—REE—and other trace and major elements) during the processes associated with calcrete formation, particularly dissolution and precipitation of carbonates in the vadose zone. The profile shows a vertical sequence, with clear zonations from the surface downwards: a reddish soil at the land surface, a nodular horizon, a laminar-structured level, and the consolidated old dune. Chemical and mineralogical analyses of samples from all levels were performed by neutron activation analysis and X-ray diffraction (bulk samples and < 2 μm fraction). Detailed studies of iron speciation and iron minerals were done by Mössbauer spectroscopy. Microfauna, mineralogical and chemical variations in the Slimene weathering profile point to a long term aerial exposure of the old dune in a semi-arid carbonated environment leading to the development of a pedogenic calcrete profile. The mineralogical variations with depth indicate carbonate dissolution at the surface and downward leaching of calcium. Secondary carbonates overgrow primary ones and precipitate as coating or concretions below. Minerals found in the upper levels correspond to original materials of the old dune and also weathering phases and atmospheric inputs. Phyllosilicates decrease while calcite and K-feldspars increase with depth. Kaolinite was the only clay mineral found in the old dune. Illite and chlorite were also found in all samples with calcrete. Authigenic smectite formation occurs above the laminar-structured calcrete due to restricted drainage conditions. The most significant chemical variations associated with the calcrete formation are (i) enrichment of Co, U, Br, and REE in calcrete; (ii) depletion of middle REE in the upper levels, particularly Eu, and enrichment of middle REE and heavy REE in calcrete; and (iii) Hf, Zr, Cr, Th, Cs, Ta, Ga, Rb, and K appear to be retained in the upper levels, where calcite has been dissolved. Here iron is more reduced. Fe3+ occurs in iron oxides (goethite and hematite), and clay minerals. The ratio goethite/hematite appears to increase down the profile; and Fe2+ is mainly present in clay minerals.  相似文献

Response of five citrus rootstocks to iron deficiency   总被引:1,自引:1,他引:0  
Citrus established in calcareous soils can be affected by iron (Fe)‐deficiency chlorosis which limits yield and the farmers' income. The degree of deficiency depends on the rootstock, but the resistance to Fe chlorosis still requires further investigation. To study physiological parameters of citrus rootstocks that could be used to evaluate resistance to Fe deficiency, plants of Troyer citrange (Citrus sinensis L. Osb. × Poncitrus trifoliata L. Raf.), Carrizo citrange, Volkamer lemon (Citrus volkameriana Ten. & Pasq.), alemow (Citrus macrophylla Wester), and sour orange (Citrus aurantium L.) were grown in nutrient solutions with 0, 5, 10, 15, or 20 μM Fe. For each rootstock, plant height, root and shoot dry weights, and concentration of Fe in the shoots and roots were measured at the end of the experiment. Chlorophyll (CHL) concentration was estimated throughout the experimental period using a portable CHL meter (SPAD‐502) calibrated for each rootstock. At the end of the experiment, CHL fluorescence parameters were measured in each rootstock with a portable fluorimeter. Maximal and variable fluorescence values indicated that the photochemistry of Troyer was more affected by a low concentration of Fe in the nutrient solution than that of other rootstocks. To compare rootstocks, the absolute CHL concentration was converted into relative yield by employing a scaling divisor based on the maximum value of total CHL in plants without Fe‐deficiency symptoms. Exponential models were developed to determine the minimum Fe concentration in nutrient solution required to maintain leaf CHL at 50% of the maximum CHL concentration (IC50). Models were also developed to assess the period of time the rootstocks were able to grow under Fe‐stress conditions before they reached IC50. Volkamer lemon and sour orange needed the lowest Fe concentration (between 4 and 5 μM Fe) to maintain IC50, and Troyer citrange had the highest Fe requirement (14 μM Fe). Citrus macrophylla and Carrizo citrange required 7 and 9 μM of Fe, respectively. Similarly, Volkamer lemon and sour orange rootstocks withstood more days under total Fe depletion or with a low concentration of Fe (5 μM Fe in nutrient solution) until they reached IC50, compared to the other rootstocks. The approach used led to a classification of the rootstocks into three categories, regarding their internal tolerance to Fe chlorosis: resistance (sour orange and Volkamer lemon), intermediate resistance (C. macrophylla and Carrizo citrange), and reduced resistance (Troyer citrange).  相似文献

The effect of Fe chlorosis on the mineral composition of field grown peach tree leaves was studied in two different areas. No significant differences in total Fe content were found, whereas 2,2’ bipyridyl extractable Fe, K and the K/Ca ratio were significantly affected in both experiments.
