以日本落叶松(Larix kaempferi)不同杂交组合为材料,依据转录组测序信息,成功获得了11种日本落叶松材料的4 CL基因的编码区序列,该序列长1 537 bp,包含完整ORF片段1 368 bp,编码455个氨基酸。qRT-PCR结果分析表明,除日本落叶松Larix16×Larix61组合外,Larix82×Larix13、Larix40×Larix10和Larix82×Larix61子代的表达均高于双亲,推测4 CL基因差异表达与落叶松杂种的木质素生物合成有关。进一步采用SNP标记方法和DNAMAN软件对不同日本落叶松杂种和亲本组合的单核苷酸多态性进行分析。共检测到66个SNP多态性位点,表明日本落叶松4CL 具有丰富的遗传多样性。66个SNP变异中,属于三突变1个;转换类型45个,占总变异的68.18%;20个属于颠换类型,占总变异的30.30%,转换:颠换=9:4。在转换类型中,A/G和C/T转换分别占28.79%和39.39%;颠换类型中A/C、A/T、G/C、G/T分别占4.55%、9.09%、9.09%和7.58%。采用NJ 聚类法对克隆片段的氨基酸序列进行了遗传多样性分析。  相似文献

An experiment was conducted to evaluate the effects of five attractants on feed intake, growth performance and appetite‐regulating genes expression for juvenile turbot (Scophthalmus maximus L.). Fish were fed with seven diets: fish‐meal‐based diet (FM), fermented soybean meal replacing 45% fish meal (FSBM) and other diets with 1% compound attractant A (CA‐A) consisting of amino acids, compound attractant B (CA‐B) consisting of quaternary ammonium bases, compound attractant C (CA‐C) consisting of nucleotides, compound attractant G (CA‐G) consisting of the combination of CA‐A, CA‐B and CA‐C and blue mussel protein hydrolysate (BMH) on the basis of FSBM diet respectively. Daily feed intake in CA‐A group was significantly higher, but that in CA‐B group was significantly lower than FSBM group. Similarly, specific growth rate was significantly enhanced in CA‐A and CA‐G groups and were significantly decreased in CA‐B group. Protein efficiency ratio and protein retention in CA‐B group were significantly lower than FSBM group. Furthermore, BMH supplement increased the mRNA expression of Ghrelin in the intestine of fish. These results showed that CA‐A and CA‐G supplement could enhance growth in plant‐based diets, but CA‐B supplement had the opposite effect, which might be associated with lower feed intake and lower feed utilization.  相似文献

To examine the effects of dietary lipids on the growth and feed utilization of jade perch juveniles, Scortum barcoo, diets containing 36.3% crude protein supplemented with increasing lipid levels (6, 9, 12, and 15% of the dry matter) were used to feed triplicate groups of 30 fish for 60 d. At the end of the experiment, more than 95% fish survived well from all diet groups (P > 0.05). Measurements on the weight gains and the daily specific growth rates indicated that fish fed with diets of 12 and 15% lipids exhibited higher growth rates (P < 0.05); evaluations for the feed conversion ratio and the protein efficiency ratio indicated that fish fed with 12 and 15% lipid diets used their feed and dietary proteins more efficiently (P < 0.05). The muscle lipid and dry matter contents increased dramatically in fish fed with higher dietary lipid levels (P < 0.05). The highest lipid contents were obtained from fish in the 15% lipid diet group and the highest amount of dry matters from the 12% lipid diet group. On the other hand, protein contents in fish muscles declined with increasing dietary lipid levels (P < 0.05), and the lowest values were shown in the 15% lipid diet group. Ash contents showed no significant differences from muscles of fish fed with four different diets (P > 0.05). Together, increasing lipid levels in fish diets was effective to improve fish growth, feed efficiency, and protein utilization.  相似文献

Intramammary infusion of Bifidobacterium breve (B. breve)-induced somatic cell (SC) counts, chemiluminescent response (CL), lactoferrin (LF) concentrations and mastitis-causing pathogens from quarters with subclinical mastitis were measured to evaluate innate immune response of mammary glands in dairy cows at 3 to 4 weeks before drying off. SC counts in 7 quarters of 7 control cows and 5 quarters of 6 cows with mastitis increased markedly on day 1 and SC values in control cows were significantly (P<0.05) increased and returned to pre-infusion levels on day 5 after B. breve-infusion. CL values in both groups increased markedly on day 1 and then decreased after B. breve-infusion; however, CL values in cows with mastitis did not return to normal levels on day 5 and at postpartum. The CL values were highly correlated with their SC counts in milk from both groups. LF concentrations increased toward day 3 after B. breve-infusion and were higher in cows with mastitis. B. breve-infusion eliminated 16.6% (1/6) of pathogens from 6 quarters with chronic subclinical mastitis. B. breve-induced SC responses in quarters from 3 cows with mastitis showed characteristic patterns of recovery, persistent and new infections. B. breve-induced SC counts in quarters from the cows in the pre-drying off were lower (25.7–70.6%) than those of the cows in mid-lactation. The intrinsic innate immune response in cows on pre-drying off may be decreased and appears to be insufficient to eliminate pathogens from mammary gland in the pre-drying off.  相似文献

The purpose of this study was to explore the mechanism of by which docosahexaenoic acid (DHA) inhibit the accumulation of adipose tissue lipid in grass carp (Ctenopharyngodon idella). We therefore designed two semi-purified diets, namely DHA-free (control) and DHA-supplemented, and fed them to grass carp (22.19 ± 1.76 g) for 3 and 6 weeks. DHA supplementation led to a significantly lower intraperitoneal fat index (IPFI) than that in the control group by reducing the number of adipocytes but significantly higher adipocyte size (P < 0.05). In the intraperitoneal adipose tissue, the DHA-fed group showed significantly higher peroxisome proliferator-activated receptor (PPAR)γ, CCAAT enhancer-binding protein (C/EBP)α, and sterol regulatory element-binding protein (SREBP)1c mRNA expression levels at both 3 and 6 weeks (P < 0.05). However, the ratio of the expression levels of B cell leukemia 2 (Bcl-2) and Bcl-2-associated X protein (Bax) was significantly lower in the DHA-fed group than in the control group (P < 0.05), and the protein expression levels of the apoptosis-related proteins caspase 3, caspase 8, and caspase 9 were also significantly higher (P < 0.05). Overall, although DHA promotes lipid synthesis, it is more likely that DHA could suppress the lipid accumulation in adipocytes of grass carp by inducing adipocyte apoptosis.  相似文献

Elaeocarpus yellows” (ELY) is a widely reported phytoplasma disease of Elaeocarpus zollingeri trees in Japan. The phytoplasma associated with ELY (ELY phytoplasma) had not been identified at the species level because its 16S rRNA sequence had yet to be reported. Here, we report the results of a sequence analysis based on 16S rRNA and secA gene sequences, which showed that the ELY phytoplasma is related to ‘Candidatus Phytoplasma malaysianum’. To our knowledge, this is the first report showing the occurrence of ‘Ca. P. malaysianum’ outside Malaysia and the infection of E. zollingeri by the phytoplasma.  相似文献
