Cucumber is one of the most important vegetable crops worldwide, which makes it a good candidate to produce doubled haploid (DH) lines to accelerate plant breeding. Traditionally, these approaches involved induction of gynogenesis or parthenogenesis with irradiated pollen, which carries some disadvantages compared to androgenesis. Despite this, studies on anther/microspore cultures in cucumber are surprisingly scarce. Furthermore, most of them failed to unambiguously demonstrate the haploid origin of the individuals obtained. In this work we focused on anther cultures using two cucumber genotypes, different previously published protocols for anther culture, different in vitro culture variants to make it more efficient, and most importantly, a combination of flow cytometry and microsatellite molecular markers to evaluate the real androgenic potential and the impact of anther wall tissue proliferation. We developed a method to produce DH plants involving a bud pretreatment at 4 °C, a 35 °C treatment to anthers, culture with BAP and 2,4-D, and induction of callus morphogenesis by an additional 35 °C treatment and sequential culture first in liquid medium in darkness and second in solid medium with light. We also found that factors such as genotype, proliferation of anther wall tissues, orientation of anthers in the culture medium and growth regulator composition of the initial anther culture medium have a remarkable impact. Our rate of chromosome doubling (81%) was high enough to exclude additional chromosome doubling steps. Together, our results present androgenesis as an improvable but yet more convenient alternative to traditional gynogenesis and parthenogenesis-based approaches. 相似文献

The effects of vermicompost (VC) (0% and 1% w/w) on treated calcareous clay soil with 0 and 50 mg phosphorus (P) kg?1 as calcium phosphate [Ca(H2PO4)2.H2O] was investigated. The soil samples were incubated for 7, 30, 60, 120, and 150 d at 25 ± 1°C and Olsen-P was measured after each incubation time. Results showed that Olsen-P increased 36% and 38% after VC addition in treated soil with 0 and 50 mg P kg?1, respectively. Recovery of Olsen-P in treated soils with VC, combined fertilizer VC + P, and fertilizer P was 42%, 42%, and 17%, respectively. The rate coefficient in treated soils with fertilizer, VC, and combined fertilizer VC + P was 0.033, 0.026, and 0.023 mg kg?1 d?1/2, respectively. It seems that the process that leads to the decrease in available P in amended soils, is controlled by P diffusion into sorption sites in micropores of aggregates. 相似文献

In this study, we selected 180 accessions from different wild relatives of wheat (Aegilops–Triticum species) and tested them in the presence of a tolerant and a sensitive control variety under well-watered and drought-stressed conditions. The results of analysis of variance for dry masses and all measured root traits showed that water regimes, accession and species main effects were highly significant. Drought stress significantly declined shoot dry mass (59.42%), root length (37.85%) and the total number of branch roots (36.25%), but increased the root-to-shoot ratio (75.00%), specific root length (64.19%) and root tissue density (29.46%). Principal component analysis for 182 individuals and 12 species groups identified two components that explained 75.67 and 82.39% of the total variation in dry mass and root traits under drought-stressed conditions, respectively. Taking together, our results identified 12 accessions with superior tolerance to drought stress. Remarkably, four species of wild relatives – Ae. cylindrica (DC genome), Ae. neglecta (UM genome), Ae. speltoides (B genome) and Ae. tauschii (D genome) – responded well to drought stress. The potential of these species could be used for molecular analysis such as marker assisted selection and gene mapping, ultimately aimed at breeding for root traits with improved adaptation to drought environments. 相似文献

In order to study the effects of desert dust on yield and yield components of cowpea, two field experiments were conducted in the factorial layout based on randomized complete block design with three replicates in Dezful and Mashhad, Iran in 2015. The experimental treatments were combinations of desert dust in three levels (0, 500 and 1500 µg m?3), numbers of desert dust application (once, twice and thrice) and two types of desert dust. The desert-dust particles were collected during dust storms occurred in Dezful and Zabol, which are the main sources of dust in Iran. The results showed that the biological yield and the grain yield were significantly decreased in both studied sites (Dezful and Mashhad) when the plants exposed to 500 µg m?3 desert dust treatment compared to normal conditions. Total soluble sugar and proline contents in plant leaf tissues increased significantly when they affected by 1500 µg m?3 of desert dust, while the plant height was significantly decreased by increasing the desert-dust concentrations at both sites. Biological yield, grain yield, and 100-seed weight were also significantly affected by the desert dust. Overall, the desert dust has adverse effects on yield of cowpea. 相似文献

A study was conducted to investigate the effects of dietary oligofructose (1, 2 and 3%) on the blood profiles of beluga (Huso huso) juveniles (18.77 ± 0.76 g) compared to fish fed an un-supplemented diet. After 7 weeks of feeding on the experimental diets,
haematological parameters, metabolic products (cholesterol, glucose and total protein) and serum enzymes (lactate dehydrogenase,
alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase) were measured. Compared to the control group
(0% oligofructose), dietary oligofructose had no effect on red blood cell counts (RBC), mean corpuscular volume (MCV), mean
cellular haemoglobin (MCH) or mean cell haemoglobin concentration (MCHC) (P > 0.05). However, haemoglobin (Hb) concentration, leucocyte (WBC) levels and the proportion of lymphocytes were significantly
higher (P > 0.05) in the 2% oligofructose fed fish than in the 3% oligofructose fed fish. Additionally, haematocrit (Hct) values (P = 0.049) and the proportion of lymphocytes (P ≤ 0.01) were significantly higher in the 2% oligofructose group than in the control group. Although serum glucose and total
protein remained unaffected, serum cholesterol was significantly lower in the 2% oligofructose group than in the control and
3% oligofructose group (P < 0.05). The results of the present study showed that oligofructose had no significant effects on serum lactate dehydrogenase,
alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase. These results indicate that fish blood profiles

This study was conducted to evaluate the effects of dietary sodium diformate (NaDF) on growth performance, gut microflora, digestive enzyme activities and immune response parameters of Asian sea bass (Lates calcarifer) juveniles. Fish with initial weight of 12.5 ± 0.4 g were fed with five experimental diets contained 0.0 (control), 5, 10.0, 15.0 and 20 g NaDF kg?1 in triplicate for 6 weeks. Fish fed diet containing 5 g NaDF kg?1 had significantly the highest final body weight and feed intake among different treatments. The gut total viable bacterial counts gradually decreased with increasing dietary NaDF level. Specific activity of chymotrypsin improved in fish fed diets administered with NaDF compared to the control group. Fish fed 5 g NaDF kg?1 diet showed the highest serum lysozyme level among different treatments. The serum classical pathway activity of complement showed higher level in fish fed diets contained 5 and 10 g NaDF kg?1 than other groups. According to break‐point regression method analysis, the optimum inclusion of dietary NaDF in L. calcarifer juveniles was estimated between 4.6 and 5.1 g/kg, when specific growth rate and feed conversion ratio were plotted against dietary NaDF levels. 相似文献

Background:Glioblastoma multiforme is the most invasive and lethal form of brain cancer with unclear etiology. Our study aimed to investigate the molecular prevalence of HCMV and EBV infections in patients with GBM. Methods:This case-control study was conducted on 42 FFPE brain tumor samples from GBM patients and 42 brain autopsies from subjects without neurological disorders. The presence of EBV and HCMV DNA was determined, using PCR and nested-PCR assays, respectively. Results:HCMV DNA was detected in 3 out of 42 (7.1%) of GBM samples and was absent from the control group (p = 0.07). Importantly, EBV DNA was detected in 9 out of 42 (21.4%) brain tissue specimens of GBM subjects, but again in none of the control group (p = 0.001).
