The aim of this study was to evaluate whether an additional intramuscular (im) injection of pFSH would increase the embryo production in zebu cows superovulated with a single subcutaneous (sc) pFSH injection. Twenty‐one Nelore cows were treated with a progesterone vaginal implant (Controlled Internal Drug Relased – CIDR B®) and injected im with 2.5 mg estradiol benzoate. Four days later, cows were assigned randomly into three groups and superovulated with pFSH. Groups A and B received single sc injections of 400 and 320 IU, respectively; Group C received multiple im injections of 400 IU in decreasing doses at 12‐h intervals over 4 days. In the morning (07:00 am) of day 3 after starting superovulation, cows received im 150 mcg cloprostenol and Group B was additionally injected im with 80 IU of pFSH. CIDR‐B was withdrawn in the afternoon (07:00 pm). Cows were inseminated 48 and 62 h after the cloprostenol injection. Embryo collection and corpora lutea (CL) estimation were done 7 days after insemination. Alternation of treatments (crossover design) occurred at a 60‐day interval. There was no significant difference (p > 0.05) of CL counts among treatments. The total (transferable and no transferable) number of recovered embryos from Group A (6.9 ± 1.5) was not different from Group C (9.8 ± 1.2), whereas Group B (5.7 ± 1.5) was lower than Group C (p < 0.05). The number of transferable embryos from Groups A (2.4 ± 0.7) and B (1.7 ± 0.6) was lower (p < 0.05) than Group C (4.6 ± 1.2). Lesser (p < 0.05) embryo production from Group B was related to lower recovery rate (46.4%), compared with Groups A (65.1%) and C (81.7%). It was concluded that an additional im subdose of pFSH, injected 48 h after a single subcutaneous (sc) dose of pFSH, does not improve the embryo production in zebu cows.  相似文献

Fulvic acids (FA) were immobilized on alumina particles in order to evaluate their catalytic effect as solid-phase redox mediator (RM) during the reductive dechlorination of carbon tetrachloride (CT) by anaerobic sludge. FA were extracted from three different soil samples and two commercial composts. Electron carrying capacity (ECC) was determined in all FA samples in order to select the appropriate source of redox-mediating compounds for CT dechlorination. TiO2, Al(OH)3, and γ-Al2O3 particles were tested as immobilizing materials for the extracted FA. FA extracted from a temperate pine forest soil showed the highest ECC (291.72 μmol g−1). The highest adsorption capacity of FA, measured as total organic carbon (TOC), was achieved by alumina (γ-Al2O3) particles (12 mg TOC-FA g−1). Results suggest that the transfer of electrons rather than their microbial generation through glucose fermentation was the rate-limiting factor during dechlorination of CT. Immobilized FA increase up to 10.4-fold the rate of CT dechlorination as compared with the control lacking FA. Immobilization of FA on alumina particles was very stable, and spectrophotometric screening did not detect any detachment of FA during dechlorination of CT, thus confirming that the enhanced dechlorination achieved could exclusively be linked to the redox-mediating capacity of immobilized FA. The present study constitutes the first demonstration that immobilized FA on alumina particles could serve as a solid-phase RM in dechlorination reactions.  相似文献

In commercial dairy production, the risk of drug residues and environmental pollutants in milk from ruminants has become an outstanding problem. One of the main determinants of active drug secretion into milk is the ATP-binding cassette transporter G2/breast cancer resistance protein (ABCG2/BCRP). It is located in several organs associated with drug absorption, metabolism, and excretion, and its expression is highly induced during lactation in the mammary gland of ruminants, mice, and humans. As a consequence, potential contamination of milk could expose suckling infants to xenotoxins. In cows, a SNP for this protein affecting quality and quantity of milk production has been described previously (Y581S). In this study, our main purpose was to determine whether this polymorphism has an effect on transcellular transport of veterinary drugs because this could alter substrate pharmacokinetics and milk residues. We stably expressed the wild-type bovine ABCG2 and the Y581S variant in Madin-Darby canine kidney epithelial cells (MDCKII) and MEF3.8 cell lines generating cell models in which the functionality of the bovine transporter could be addressed. Functional studies confirmed the greater functional activity in mitoxantrone accumulation assays for the Y581S variant with a greater relative V(MAX) value (P = 0.040) and showed for the first time that the Y581S variant presents greater transcellular transport of the model ABCG2 substrate nitrofurantoin (P = 0.024) and of 3 veterinary antibiotics, the fluoroquinolone agents enrofloxacin (P = 0.035), danofloxacin (P = 0.001), and difloxacin (P = 0.008), identified as new substrates of the bovine ABCG2. In addition, the inhibitory effect of the macrocyclic lactone ivermectin on the activity of wild-type bovine ABCG2 and the Y581S variant was also confirmed, showing a greater inhibitory potency on the wild-type protein at all the concentrations tested (5 μM, P = 0.017; 10 μM, P = 0.001; 25 μM, P = 0.008; and 50 μM, P = 0.003). Differential transport activity depending on the genotype together with the differential inhibition pattern might have clinical consequences, including changes in substrate pharmacokinetics (and subsequently pharmacodynamics) and more specifically, changes in secretion of ABCG2 substrates into milk, potentially implying important consequences to veterinary therapeutics.  相似文献

Danofloxacin, a veterinary fluoroquinolone antimicrobial drug, is actively secreted into milk by an as yet unknown mechanism. One of the main determinants of active drug secretion into milk is the transporter (BCRP/ABCG2). The main purpose was to determine whether danofloxacin is an in vitro substrate for Bcrp1/BCRP and to assess its involvement in danofloxacin secretion into milk. In addition, the role of potential drug-drug interactions in this process was assessed using ivermectin. Danofloxacin was transported in vitro by Bcrp1/BCRP, and ivermectin efficiently blocked this transport. Experiments with Bcrp1(-/-) mice showed no evidence of the involvement of Bcrp1 in plasma pharmacokinetics of danofloxacin. However, the milk concentration and milk-to-plasma ratio of danofloxacin were almost twofold higher in wild-type compared with Bcrp1(-/-) mice. The in vivo interaction with ivermectin was studied in sheep after co-administration of danofloxacin (1.25 mg/kg, i.m.) and ivermectin (0.2 mg/kg, s.c.). Ivermectin had no significant effect on the plasma levels of danofloxacin but significantly decreased danofloxacin concentrations in milk by almost 40%. Concomitant administration of multiple drugs, often used in veterinary therapy, may not only affect their pharmacological activity but also their secretion into milk, because of potential drug-drug interactions mediated by BCRP.  相似文献

In this study we explored the spatial variation of Bovine tuberculosis (BTB) risk of being positive, new positive or persistently positive, as well as the risk of eliminating BTB in positive herds throughout Spain from 2006 to 2009 by means of hierarchical Bayesian models. The results of the models showed that the risk of infection (positive or new positive herds), persistence and elimination was lower in counties located in north and north-eastern of Spain, and in the Balearic and Canary islands than in the rest of the country. In some counties the risk of positivity was high during the four years of study, whereas there were others where the risk of positivity was high only in some of the years. With regard to the risk of persistence of BTB positive herds, counties located in the central, western and south-western part of the country had a higher risk in the three studied periods. This study has identified some specific areas of increased BTB risk in Spain, information that is useful for disease management.  相似文献

The optimum culture system for in vitro matured and fertilised oocytes still remains to be clarified. Culture media (CM) for mammalian embryos are routinely prepared fresh for use and preserved under refrigeration during one or two weeks. The purposes of this work were (1) to compare the efficiency of a synthetic oviduct fluid (SOF) with two different bovine serum albumin (BSA) concentrations (3 and 8 g/L) for the in vitro production of bovine blastocysts, (2) to test the effect of timing on adding fetal calf serum (FCS) to the SOF, and (3) to evaluate the effects on bovine embryo development of freezing and lyophilisation as procedures for preserving the SOF. Supplementation of SOF with 3 g/L BSA increased Day-7 blastocyst expansion rates (18.3 ± 1.6 vs. 14.4 ± 0.7; P < 0.05), although no differences in hatching rates were found. Addition of FCS to SOFaa (SOF with amino acids) medium supplemented with sodium citrate (SOFaaci) at 48 and at 72 h post-insemination (PI) allowed obtaining higher Day-6 embryo development rates than when FCS was added at 18 or 96 h PI (Day-6 morulae + blastocyst rate: 30.0 ± 1.1, 40.8 ± 1.1, 43.9 ± 2.3 and 39.3 ± 0.5 for FCS addition at 18, 48, 72 and 96 h, respectively). Hatching rates were significantly improved when serum was added at 72 h PI. Finally, both refrigeration and lyophilisation appeared as useful cryopreservation procedures for SOFaaci, although a significant loss of its ability to support embryo development, compared to the control fresh culture medium, was observed.  相似文献

In recent years, elevated arsenic concentrations in groundwater used for drinking water supplies have been recognised in the Madrid Tertiary detrital aquifer. Although only natural causes have been suggested as the source of arsenic, this study aims to highlight that the anthropogenic contribution cannot be disregarded. During the sub-catchment??s areas sampling, we found many geographical sites where natural arsenopyrite [FeAsS] originally encapsulated in pegmatite bodies and quartz veins, was artificially outcropped and dumped out, since mining wastes were scattered and exposed to weathering. Several mineral and ground specimens were collected to analyse its mineralogical and chemical composition by X-Ray Diffraction (XRD) and X-Ray Fluorescence (XRF) spectrometry and by Environmental Scanning Electron Microscope (ESEM). Both, the abundant existence of secondary phases, such as scorodite [FeAsO4?2H2O] and jarosite [KFe3(SO4)2(OH)6], much more soluble than arsenopyrite, and the lixiviation experiments of arsenopyrite in acidic media to simulate acid mine drainage (AMD) conditions, usually found in old mining districts, point to a potential risk of arsenic contamination of surface water bodies, which operate as recharged waters of the aquifer in the studied area. The elemental determination of heavy metals present in ground samples by XRF analyses, reaching up to 1,173 mg kg?C1 of copper, 347 mg kg?C1 of lead and 113,702 mg kg?C1 of arsenic; and the physicochemical and arsenic fractionation studies of soil samples, led us to classify the soil as Spolic Technosol (Toxic). The contamination of the area due to old mining activities could release arsenic to Madrid water supplies; accordingly, additional decontamination studies should be performed.  相似文献

Cataiase, peroxidase and IAA‐oxidase activities, so as odiphenolics content in sunflower leaves grown in hydroponics with 4 B levels (0.05; 0.25; 0.5 and 2.0 ppm) were determined. The biochemical assays were made when visual symptons were clearly developed.

Cataiase activity significantly increased in B deficient leaves. Peroxidase, IAA‐oxidase activities and o‐diphenolics content did not significantly change with B levels. The lag period for IAA‐oxidase activity was significantly reduced in B deficient and toxic leaves with respect to the normal ones. These results are in accordance with the hypothesis that one of the B action could be to enhance cataiase activity, the “in vivo” decrease of H2O2 levels beeing the cause of the variation in other enzymatic activities related with that substrate.  相似文献
