Purified lipopolysaccharides (LPS) from 16 serotypes of Pasteurella multocida were complexed with Aspergillus fumigatus ribosomes. The complexes were used as inocula to prepare antisera, in chickens, for somatic antigen typing by the gel diffusion precipitin test (GDPT). Antisera made against 15 of 16 LPS reacted with their respective specific heat-stable antigens in the GDPT and homologous LPS in the passive hemagglutination test. Antisera could not be made against serotype 15 LPS. Correlation was not observed between intensity of the precipitin reaction in the GDPT and titer to homologous LPS in the passive hemagglutination test. Most antisera cross-reacted with other heat-stable antigens of other serotypes in the GDPT. Many of these cross-reactions were eliminated by dilution. Cross-reactions that occurred in the GDPT with antisera made against LPS of serotypes 2, 5, 7, and 8 could not be eliminated by dilution. 相似文献

Changes in tissue and plasma isoenzymes of alkaline phosphatase (ALP) were qualitatively and quantitatively determined for male and female Atlantic salmon parr, silvery parr, smolt, immature grilse, prespawning grilse and postspawning grilse using cellulose acetate electrophoresis, densitometry and spectrophotometry. Tissue ALP isoenzymes were isolated from intestine, kidney, bone, liver, and gonad and compared to plasma isoenzymes. Parr plasma displayed three isoenzymes from bone and liver (slow and fast). During smoltification, ALP activity increased in tissue extracts from liver, gonad, and kidney of males and females. Total plasma ALP activity also increased and was due to slow and fast liver isoenzymes. During ovarian development, total ALP plasma activity increased in females and was due mostly to liver isoenzymes and an incompletely identified isoenzyme or isoenzyme mixture (band 2). However, in males total ALP plasma activity did not increase during maturation and no band 2 was evident. In male and female maturing adult grilse, bone ALP activity declined and the isoenzyme band evident in parr plasma could not be detected. ALP activity declined in the plasma of postspawning males and females. In females this was due partly to the total clearance of band 2 from the plasma, together with lowered levels of liver isoenzymes. Treatment of postspawned grilse in February and March with triiodothyronine and thyroxine elevated plasma thyroid hormone levels and increased plasma ALP levels. In conclusion, plasma ALP isoenzyme activities change with physiological state, and knowledge of the conditions governing these changes is important when using these enzymes as a diagnostic tool. 相似文献

- Local population characteristics and habitat connectivity both have important influences on metapopulation persistence; however, the relative importance of each can vary depending on the ecological context, making it difficult to apply general ‘rules-of-thumb’ for conservation actions. This is particularly true in dendritic networks, where habitat connections are constrained.
- By constructing a metapopulation model that specifically accounts for the unique characteristics and ecological system of a target species, the relative influence of local population characteristics and habitat connectivity on metapopulation persistence can be assessed more accurately.
- This modelling approach was used to improve conservation outcomes for the Canterbury mudfish (kо̄waro Neochanna burrowsius), a critically endangered and poorly understood species that is endemic to the Canterbury Plains of Aotearoa/New Zealand. A spatially structured metapopulation model of N. burrowsius was used to investigate the relationship between habitat patch connectivity, rate of population growth as limited by habitat quality (rconditional), and metapopulation persistence.
- N. burrowsius metapopulation persistence increased most strongly under increases in rconditional, but increases in habitat connectivity also had positive effects (94% metapopulation persistence for highly connected networks vs. 39% metapopulation persistence for poorly connected networks where rconditional = 1.3).
- Although rates of population growth were more influential than increases in habitat connectivity in this case study, both will be important to consider for effectively allocating conservation resources for metapopulations in dendritic networks, depending on what managers could effectively manipulate to enhance metapopulation persistence.

The transport of auxin controls developmental events in plants. Here, we report that in addition to maintaining vacuolar pH, the H+-pyrophosphatase, AVP1, controls auxin transport and consequently auxin-dependent development. AVP1 overexpression results in increased cell division at the onset of organ formation, hyperplasia, and increased auxin transport. In contrast, avp1-1 null mutants have severely disrupted root and shoot development and reduced auxin transport. Changes in the expression of AVP1 affect the distribution and abundance of the P-adenosine triphosphatase and Pinformed 1 auxin efflux facilitator, two proteins implicated in auxin distribution. The number of vesicles released at excitatory synapses and the number of release sites per synaptic connection are key determinants of information processing in the cortex, yet they remain uncertain. Here we show that the number of functional release sites and the number of anatomically identified synaptic contacts are equal at connections between spiny stellate and pyramidal cells in rat barrel cortex. Moreover, our results indicate that the amount of transmitter released per synaptic contact is independent of release probability and the intrinsic release probability is high. These properties suggest that connections between layer 4 and layer 2/3 are tuned for reliable transmission of spatially distributed, timing-based signals. 相似文献
