Artificial insemination is widely used in the cattle industry and a major challenge is to ensure that semen is free of infectious agents. A healthy donor bull was tested for freedom from infectious agents. A bovine herpesvirus was isolated in testis cells and identified as bovine herpesvirus type 5 (BoHV-5) by polymerase chain reaction and by direct amplicon sequencing. The amplicon sequence shared 100% similarity with the published sequence of BoHV-5. This is the first report in Australia of BoHV-5 in semen. The implications of this finding are discussed.  相似文献

Dihydroergosine (DHES) is the principal toxic alkaloid produced by sorghum ergot (Claviceps africana). It has recently been shown that DHES levels as low as 1 mg/kg in animal feed can cause significant production losses. Quantitative immunoassays for detecting the related rye ergot alkaloid, ergotamine, are described in the literature, but those assays are relatively insensitive for DHES. This paper describes competitive enzyme-linked immunosorbent assays (ELISA) for measuring the DHES concentration in grains and mixed animal feed. The assays were developed using a DHES specific mouse monoclonal antibody and rabbit polyclonal antibodies raised against DHES conjugated to bovine serum albumin. Recoveries of between 77 and 103% were obtained from spiked grain using a simple, one step extraction with 70% methanol. Both the monoclonal and the polyclonal assays are capable of detecting DHES concentrations above 0.01 mg/kg, but quantification is most reliable at concentrations of 0.1 mg/kg or higher.  相似文献

A supplement system for the control of equine nutritional secondary hyperparathyroidism (NSH) was evaluated on 4 farms in the Arcadia Valley of the Queensland brigalow region. Thirty-three local stock horses (of which 13 had clinical NSH and 7 were recent introductions) were grazed on buffel grass (Cenchrus ciliaris) for the 6 months from September 1979 to February 1980. Each horse was fed 1.0 kg of a mixture of ground limestone plus dicalcium phosphate (1:2) in 1.5 kg molasses each week. The pasture was hazardous during this time (total oxalate content above 0.5% and calcium: oxalate ratio below 0.5), but no new NSH cases occurred and those horses with clinical NSH improved, most becoming normal. After the first 6 months of supplementation, 3 new cases occurred on one farm. Doubling of the supplement dose and substituting rock phosphate in molasses for the previous mixture corrected the problem. The breakdown was thought to be partly because the supplement dose was slightly inadequate and partly because of behavioural factors.  相似文献

A novel fungal infection of wheat by Botryosphaeria zeae was identified on the Darling Downs of Queensland, Australia and called ‘white grain’ because of its bleached appearance. The only nutritional changes in the wheat grain infected with B. zeae were decreases in nitrogen and total amino acids (~5%), and slight increases in the lysine, fibre and fat content, with starch unaffected. Nutrient digestibility and potential toxicity were assessed in weaner pigs housed in metabolism crates and fed this grain over a 4‐week period, as they grew from 15 to 35 kg. Digestibility of energy and nitrogen in white grain was not different from that of normal wheat. The piglets were then bled for biochemical and haematological testing, slaughtered and the entire viscera subjected to gross pathological inspection, followed by histological examination of the liver, kidney, pancreas, spleen, heart, lung, muscle and intestine. White grain did not affect palatability of diets, pig growth rates; no abnormalities were detected in tissues, while biochemical and haematological parameters did not suggest any toxic effect. Hence, wheat with white grain appears suitable for use in pig diets, but the use of this wheat for human food should be restricted until additional longer term studies are conducted.  相似文献

Abstract

AIMS: To determine the concentration of Campylobacter spp. as well as faecal indicator bacteria; faecal coliforms, Escherichia coli and enterococci in the faeces of healthy adult horses in a sample of properties in the Canterbury region of New Zealand.

METHODS: The faeces of healthy adult horses (n=59), including ponies, pleasure horses and Thoroughbreds, were collected from eight properties around Christchurch, New Zealand. The faeces were analysed for concentrations of Campylobacter spp and faecal indicator bacteria; faecal coliforms, Escherichia coli and enterococci. The presence of other animals on the properties sampled as well as the age, feed and health of the horses at the time of sampling was recorded.

RESULTS: Enterococci and faecal coliforms were isolated from all samples, and E. coli was isolated from 58/59 samples. Mean concentrations of faecal coliforms and E. coli did not differ between properties, but there was a significant difference in mean concentration of enterococci between properties. Campylobacter spp. were detected in two faecal samples with one isolate being determined by PCR analysis to be a thermotolerant Campylobacter species, the other C. jejuni.

CONCLUSIONS: This is the first known report quantifying the concentration of Campylobacter spp. present in healthy adult horses in New Zealand. The presence of equine faecal material in water could elevate concentrations of faecal bacteria and therefore needs to be considered as a source of water contamination. The access of horses to waterways and coastal environments may also need to be restricted to prevent transmission of faecal indicator bacteria and potentially zoonotic agents.  相似文献
