An enzyme-linked immunosorbent assay (ELISA) was developed to measure total antibody to Toxoplasma gondii in serum samples from macropods. The validity of the assay was established by comparing parasite isolation in mice for 17 Tasmanian pademelons (Thylogale billardierii) and 17 Bennett's wallabies (Macropus rufogriseus rufogriseus). The ELISA was then used to detect antibody against T. gondii in serum from 236 macropods, collected from 21 locations in Tasmania, including Flinders Island. Antibody against T. gondii was detected in 20 animals (15 T. billardierii and 5 M. rufogriseus). There was a significant (p less than 0.01) difference in possession of T. gondii antibodies between adult (greater than or equal to 1 year of age) Tasmanian pademelons and Bennett's wallabies.  相似文献

Eight gnotobiotic lambs deprived of colostrum were infected by mouth when two hours old with nonenterotoxigenic strains of Escherichia coli. All developed clinical signs of watery mouth disease and seven died within 24 hours. The mean concentrations of several blood constituents were determined in samples taken at intervals until 24 hours after infection in infected lambs and in four control lambs. The biochemical and haematological changes observed in the lambs developing watery mouth disease were those characteristic of endotoxic shock.  相似文献

Nineteen does (female goats) were dosed with 500,000 oocytes of Hammondia hammondi prior to breeding. At about 90 days of gestation these, and 18 uninoculated does were challenged with 25,000 Toxoplasma gondii oocysts. The 19 H. hammondi--inoculated does produced 26 live and one dead kid (newborn goat). The 18 does not given H. hammondi produced 12 live and 19 dead kids. However, examination of all of the kids by isolation of T. gondii in mice, serology and histology revealed that they were all infected with T. gondii. Thus, while H. hammondi "vaccination" is protective against the deleterious effects of T. gondii on pregnant does, perhaps by reducing the severity of placental lesions, it does not prevent foetal infection.  相似文献

AIMS: To investigate the seroprevalence of antibodies to Toxoplasma gondii in New Zealand sea lions (Phocarctos hookeri), as a potential contributor to reproductive failure.

METHODS: Archived sera were sourced from New Zealand sea lions from two recolonising mainland populations in the Otago Peninsula (n=15) and Stewart Island (n=12), as well as a declining population at Enderby Island (n=28) in the New Zealand sub-Antarctic. Sera were tested for antibodies to T. gondii using a commercially available ELISA (with samples considered positive if the sample to positive ratio was?>30%), and latex agglutination test (LAT; with titres ≥1:32 considered positive). Western blot analysis was used to validate the results of a subset of 14 samples.

RESULTS: Five samples from sea lions in mainland locations were confirmed positive for antibodies to T. gondii. Two adult females exhibited high LAT antibody titres (min 1:2048, max 1:4096) on both occasions when sampled 1 and 2 years apart, respectively. No animals from Enderby Island were seropositive.

CONCLUSIONS: Toxoplasma gondii infection is unlikely to be a major contributor to poor reproductive success in New Zealand sea lions. However, continued surveillance is pertinent to assess subclinical and clinical impacts of the parasite on these threatened populations. The commercial tests evaluated here, with further species-specific threshold refinement could provide a fast, inexpensive and reliable indicator of T. gondii exposure in New Zealand sea lions.  相似文献

OBJECTIVE: To determine if composite connecting rods confer nonlinear stiffness characteristics on unilateral and bilateral external skeletal fixators (ESF) in cranial-caudal bending and axial loading. STUDY DESIGN: Mechanical testing performed on models. SAMPLE POPULATION: Six models of 6-pin ESF constructs composed of birch dowels, a commercial ESF system, and composite connecting rods. METHODS: Unilateral and bilateral ESF configurations were assembled using either specially designed composite titanium and silicone (composite group) or solid titanium (solid group) connecting rods. Mechanical testing was performed in axial loading and 4-point cranial-caudal bending. Stiffness was determined at a low and high-load range, and was considered increasing and nonlinear if the stiffness at high loads was greater than at low loads. RESULTS: The stiffness of the solid group was linear in all testing modes and configurations. Bilateral composite fixators had a nonlinear increasing stiffness in axial loading and cranial-caudal bending. Unilateral composite fixators had a nonlinear increasing stiffness in axial loading, but not cranial-caudal bending. Solid connecting rods conferred a higher stiffness in all testing modes and configurations. CONCLUSIONS: Composite connecting rods resulted in nonlinear increasing axial and bending stiffness in bilateral fixators, and in axial load in unilateral fixators. CLINICAL RELEVANCE: Conventional ESF can be constructed so that the stiffness increases as load increases. This provides the surgeon with additional options to control the local mechanical environment of a healing fracture, which may be used to enhance fracture healing.  相似文献

OBJECTIVE: To compare results (ie, return to racing and earnings per race start) of surgical versus nonsurgical management of sagittal slab fractures of the third carpal bone in racehorses. DESIGN: Retrospective study. ANIMALS: 32 racehorses (19 Thoroughbreds, 11 Standardbreds, and 2 Arabians). PROCEDURE: Medical records and radiographs were reviewed to obtain information regarding signalment and treatment. Follow-up information was obtained from race records. Robust regression analysis was performed to evaluate earnings per start in horses that raced at least once before and after injury. RESULTS: 22 (69%) horses raced at least once after treatment of the fracture. All 7 horses treated by means of interfragmentary compression raced after treatment, and horses that underwent interfragmentary compression had significantly higher earnings per start after the injury than did horses treated without surgery. Eight of 9 horses treated by means of arthroscopic debridement of the damaged cartilage and bone raced after treatment, but only 7 of 16 horses treated without surgery (ie, stall rest) were able to return to racing after treatment. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that racehorses with sagittal slab fractures of the third carpal bone have a favorable prognosis for return to racing after treatment. Horses treated surgically were more likely to race after treatment than were horses treated without surgery.  相似文献

Downy mildew, caused by Peronospora arborescens, has become the major disease affecting oilseed poppy (Papaver somniferum) since its first record in Tasmania in 1996. Two field trials conducted in 2000 and 2001 studied the progression and spatial distribution of downy mildew epiphytotics. The logistic and exponential models best described the progression of disease incidence and severity, respectively. Incidence and severity increased rapidly following canopy closure. In 2001, incidence increased from 0.16%, prior to canopy closure, to 100% at late flowering (40 days). Spatial analyses of epiphytotics were conducted by fitting the beta-binomial and binomial distributions, median runs analysis, and the spatial analysis by distance indices. All analyses demonstrated that the distribution of incidence and severity was strongly spatially aggregated from canopy closure until at least late flowering. These results suggest that secondary spread from a few primary infections is the major factor in epiphytotics.  相似文献
