A homogenous enzyme immunoassay (EIA) for measurement of serum thyroxine (T4) concentration was evaluated for use with canine and feline serum. The EIA method was linear from 0 to 150 nmol T4/L for human serum, 0 to 94 nmol T4/L for feline serum and 10 to 60 nmol T4/L for canine serum. Intra- and interassay precision studies yielded coefficients of variation = 8% using single point measurements. Method comparison studies gave close agreement between radioimmunoassay (RIA) and EIA results. Correlation coefficients (r values) were 0.88 and 0.97 for canine and feline samples respectively, after application of species-specific factors to correct the calibrator values assigned for human serum samples. The EIA showed no interference from hemolysis at hemoglobin concentrations = 20 g/L or from moderate lipemia. Highly lipemic specimens could be tested after centrifugation to clarify the sample. The EIA showed less interference from autoantibodies to T4 than the RIA method. The EIA method allows automation of T4 testing in a veterinary hospital or laboratory, and can be integrated with the routine clinical chemistry panel. 相似文献

A 2-year longitudinal survey was carried out to investigate factors affecting milk yield in crossbred cows on smallholder farms in and around an urban centre. Sixty farms were visited at approximately 2-week intervals and details of milk yield, body condition score (BCS) and heart girth measurements were collected. Fifteen farms were within the town (U), 23 farms were approximately 5 km from town (SU), and 22 farms approximately 10 km from town (PU). Sources of variation in milk yield were investigated using a general linear model by a stepwise forward selection and backward elimination approach to judge important independent variables. Factors considered for the first step of formulation of the model included location (PU, SU and U), calving season, BCS at calving, at 3 months postpartum and at 6 months postpartum, calving year, herd size category, source of labour (hired and family labour), calf rearing method (bucket and partial suckling) and parity number of the cow. Daily milk yield (including milk sucked by calves) was determined by calving year (p < 0.0001), calf rearing method (p = 0.044) and BCS at calving (p < 0.0001). Only BCS at calving contributed to variation in volume of milk sucked by the calf, lactation length and lactation milk yield. BCS at 3 months after calving was improved on farms where labour was hired (p = 0.041) and BCS change from calving to 6 months was more than twice as likely to be negative on U than SU and PU farms. It was concluded that milk production was predominantly associated with BCS at calving, lactation milk yield increasing quadratically from score 1 to 3. Although the megamitochondria (MM) were localized in various pathological conditions, normal retina of some mammalian species was reported to include MM for various physiological roles. However, it was not clearly confirmed whether the MM is present in the retina of lower vertebrate as well. In this study, we tried to show the presence of the MM in the zebrafish retina using electron microscopic technique. In all the photoreceptors including rods, cones and double cones of the zebrafish retina, MM were observed in the ellipsoid of inner segment. In the photoreceptor epllipsoid of the zebrafish retina, the mitochondria located in the central portion of the ellipsoid had a highly electron-dense matrix, which were accompanied by the mitochondria with electron-lucent matrix in the apical portion of the ellipsoid. The presence of MM was more clearly discernable in the rods, which were localized under the double cones. This finding is somewhat different from those observed in the previous studies because MM were localized in the inner segment of cones, but were not in those of rods in the case of mammalian retina. Although the exact physiological meaning for the presence of MM in some vertebrate species should be further studied, the present study could show that the MM in the ellipsoid of the retinal photoreceptors was not only restricted in some mammalian species. 相似文献

Effects of long-term growth hormone-releasing factor (GRF) on growth performance, feed conversion efficiency (FCE) and dry matter intake (DMI) were studied in growing buffaloes. Twelve female Murrah buffaloes of 6-8 months of age were divided into two groups of six each on the basis of their body weights so that the average body weights of the groups did not differ (p > 0.05). Animals of each group were administered intravenously with either sterile distilled water (control group) or equal volume of GRF solution containing 10 mug GRF (1-44)-NH2/100 kg body weight (treatment group) at fortnight interval from week 6 (5-week pre-treatment period) for 36 weeks (weeks 6-42 treatment period). Thereafter a 10-week post-treatment period was added. All the animals were weighed consecutively 2 days in a week and the average body weight of the two observations in the week was thus considered for further calculation. Dry matter intake was recorded daily. Average daily gain, FCE and DMI/100 kg body weight were also calculated. Plasma progesterone was estimated in the samples collected twice a week at 3-4-day intervals to assess whether either group had begun ovarian cycles. It was found that ADG and FCE were higher (p < 0.01) in GRF-treated animals during treatment and even 10-week post-treatment period. Interestingly, total DMI was not different (p > 0.05) between the groups during treatment period but found to be lower in GRF treated animals during post-treatment period. The DMI/100 kg body weight was lower (p < 0.01) in GRF-treated animals during treatment and even after cessation of treatment for 10 weeks. The GRF administration for long-term increased (p < 0.05) plasma progesterone. Plasma progesterone concentrations suggest that no animal from either group reached puberty till the end of the experiment. In conclusion, repeated GRF administration for longer term decreased (p < 0.01) DMI/100 kg body weight and increased (p < 0.01) FCE and enabled the animals to grow faster. 相似文献

The repellency of fennel (Foeniculum vulgare Miller)-containing products (5% aerosol and 8% cream) against mosquitoes was compared with those of citronella oil, geranium oil and deet, as well as three commercial repellents, Baby Keeper cream containing IR3535, MeiMei cream containing citronella and geranium oils, and Repellan S aerosol containing 19% N,N-diethyl-m-toluamide (deet) under laboratory and field conditions. In a laboratory study with female Aedes aegypti (L), fennel oil exhibited good repellency in a release-in-cage test and repellency in skin and patch tests of the oil was comparable with those of citronella and geranium oils. In paddy field tests with five human volunteers, 5% and 8% fennel oil-containing aerosol and cream produced 84% and 70% repellency, respectively, at 90 min after exposure, whereas Baby Keeper cream and MeiMei cream gave 71% and 57% repellency at 90 min after exposure, respectively, and Repellan S aerosol gave 89% repellency at 210 min. The species and ratio of mosquitoes collected were the genera Culex (44.1%), Anopheles (42.2%), Aedes (7.8%) and Armigeres (5.9%). The performance of the standard agar gel immunodiffusion (AGID) test and the competitive enzyme-linked immunosorbent assay (cELISA) for the detection of serum antibody against bluetongue virus (BTV) in clinically healthy and diseased camels in Gujarat state were compared. Out of 176 sera tested, 22 (12.5%) and 34 (19.3%) were positive for group-specific bluetongue antibodies by AGID and cELISA, respectively. Maximum seropositivities of 18.0% by AGID and 25.8% by cELISA were recorded in the Kutchhi breed, and of 6.9% and 12.6%, respectively, in the Marwari breed. The seroprevalence detected by AGID and cELISA in clinically healthy and diseased camels did not differ significantly with regard to bluetongue disease in these breeds. 相似文献
