The centre of origin of the globally distributed wheat pathogen Parastagonospora nodorum has remained uncertain because only a small number of isolates from the Fertile Crescent were included in earlier population genetic and phylogeographic studies. We isolated and genetically analysed 193 P. nodorum strains from three naturally infected wheat fields distributed across Iran using 11 neutral microsatellite loci. Compared to previous studies that included populations from North America, Europe, Africa, Australia, and China, the populations from Iran had the highest genetic diversity globally and also exhibited greater population structure over smaller spatial scales, patterns typically associated with the centre of origin of a species. Genes encoding the necrotrophic effectors SnToxA, SnTox1, and SnTox3 were found at a high frequency in the Iranian population. By sequencing 96 randomly chosen Iranian strains, we detected new alleles for all three effector genes. Analysis of allele diversity showed that all three effector genes had higher diversity in Iran than in any population included in previous studies, with Iran acting as a hub for the effector diversity that was found in other global populations. Taken together, these findings support the hypothesis that P. nodorum originated either within or nearby the Fertile Crescent with a genome that already encoded all three necrotrophic effectors during its emergence as a pathogen on wheat. Our findings also suggest that P. nodorum was the original source of the ToxA genes discovered in the wheat pathogens Phaeosphaeria avenaria f. sp. tritici 1, Pyrenophora tritici-repentis, and Bipolaris sorokiniana.  相似文献

The influence of stocking density (1, 2, 4, 6 and 8 kg m?2) was investigated on haematological parameters, growth and fin erosion of great sturgeon (Huso huso) juveniles for a period of 8 weeks. The mean weight of fish at the start of trial was 93.13±1.04 g. After 8 weeks of rearing, the mean weight was 362.4, 319.7, 267, 242.1 and 211.1 in densities 1–8 kg m?2 respectively. The results of this study showed that growth parameters, including condition factor, weight, weight gain, feed conversion ratio, specific growth rate and body weight increase, had a statistically significant difference among treatments (P<0.05). A significant difference (P<0.05) was observed in haematocrit, but the other haematological parameters, including red blood cells, white blood cells (WBC), haemoglobin concentration, mean corpuscular haemoglobin, mean corpuscular volume, mean corpuscular haemoglobin concentration and differential WBC count, showed no significant effect with stocking density (P>0.05). At the end of the experiment, stocking density had no significant effect on plasma cortisol and glucose concentration. The fin length (dorsal, anal, ventral, pectoral and caudal) of fish was measured to calculate the fin index. According to this index, dorsal, anal, ventral and pectoral fins showed no significant difference among treatments (P>0.05), but the erosion of the caudal fin was significantly different between fish held on 6 and 8 kg m?2 (P<0.05). These results showed that rearing density has a major effect on the growth indices of H. huso. Unlike many other fish, great sturgeon exhibited lower stress responses to high stocking density. This indicates that they are more tolerable to rearing conditions in high stocking densities. With respect to the various effects that density causes on growth, fin erosion and physiological and haematological parameters, better understanding of these phenomena considering different levels of density could have a beneficial impact on many rearing steps of this species.  相似文献

The objective was to evaluate interactions between starter protein (180 vs. 200 g/kg, DM basis) and a mixture of essential oils (EOs; containing thymol, eugenol, vanillin, limonene and guaiacol) on growth, metabolic and ruminal functions of Holstein dairy calves. In a completely randomized 2 × 2 factorial design, 48 calves, 3 days old (averaging BW 42.7 ± 1.9 kg), were allocated into groups fed the following diets: (i) 180 g/kg CP with no EO (180P‐NEO); (ii) 180 g/kg CP with EO (180P‐EO); (iii) 200 g/kg CP with no EO (200P‐NEO); and (iv) 200 g/kg CP with EO (200P‐EO). The EO was supplemented as 1 g/kg of starter DM. Calves were fed ad libitum starter diet and were weaned at day 59 of age, but diets continued until day 80. There were no interactive effects of CP and EO on intake and growth. Pre‐weaning feed efficiency tended to be increased for 200P‐EO (p = .09). Average daily gain and feed efficiency during pre‐weaning period as well as weaning weight were increased (p < .05) by EO, whereas wither height was increased by EO (p = .03) and tended to be increased for 200P vs. 180P (p = .06). Post‐weaning blood urea nitrogen concentration tended to be lower in 180P vs. 200P (p = .08). Ruminal short‐chain fatty acids concentration was greatest in 200P‐EO. The EO increased both butyrate (p = .02) and propionate proportions (p = .01) and reduced acetate proportional ratio (p < .01). Ruminal ammonia‐N was tended to be lower in calves‐fed EO (p = .05) and was lower in those fed 180P vs. 200P (p < .01). In conclusion, supplementation of the starter diet with essential oil improved weight gain, growth and feed efficiency of dairy calves, irrespective of dietary protein content.  相似文献

The effect of five different light regimes on growth, stress and hematological indices was studied in Persian sturgeon, Acipenser persicus. Fish with average weight of 645.3 ± 11.2 g were subjected to different photoperiods (24 L, 12 L:12 D, 16 L:8 D, 8 L:16 D and 24 D) for 8 wk. Blood samples were collected at the end of the experiment for measuring cortisol, glucose and hematological features. The whole growth parameters showed no significant difference. Plasma cortisol concentration was significantly higher in 12 L:12 D, while the lowest level was observed in fish exposed to 24 D. No significant changes were observed among the treatments for glucose concentration. Lactate concentration varied significantly among the treatments. Some hematological indices including hematocrit and number of white blood cells were affected by different light regimes, but the others (hemoglobin and number of red blood cells) were not affected significantly. The results showed that photoperiod manipulation can alters some stress‐related metabolites and may enhance growth rate in fish exposing to continuous darkness.  相似文献

Brown rust or leaf rust is one of the most important diseases of wheat occurring almost in all wheat-producing regions and reduces crop yield. In order to produce resistant cultivars, it is necessary to identify resistance genes in different germplasms and combine them in (a) suitable stock(s). To identify the presence of the leaf rust resistance genes using STS and SCAR markers, 83 Iranian wheat genotypes, Lr near-isogenic lines in Thatcher (positive controls), and the cultivar Thatcher (negative control) were used. After growing plants in the greenhouse, DNA was extracted by SDS method. Following that, polymerse chain reaction was performed for the markers of the resistance genes Lr9, Lr26, Lr28, Lr34, and Lr35 which amplified 1,100, 1,100, 378, 150, and 900 bp bands, respectively. Based on the results, the resistance genes Lr9 and Lr35 were only present in the positive controls. The resistance gene Lr26 was only detected in four cultivars; Arta, Pishtaz, Shiroodi, and Falat, and the gene Lr34 was present in six cultivars (Akbari, Bam, Tajan, Khazar 1, Sistan and Niknezhad). The Lr28 primer amplified a band of the same size in all genotypes even the negative control and therefore the presence/absence of this gene could not be validated. These results indicate the necessity for designing a specific primer for Lr28. In general, only the genes Lr26 and Lr34 were present in some genotypes. The genes Lr9 and Lr35 were not present in this collection and as based on rust surveys, no virulence has been detected for Lr9 and Lr28, so they could be transferred to suitable lines from donor sources.  相似文献

Necrotic enteritis (NE) is one of the most important enteric diseases in poultry and is a high cost to the industry worldwide. It is caused by avian-specific, Necrotic Enteritis Beta toxin (NetB)-producing, strains of Clostridium perfringens that also possess in common other virulence-associated genes. In Europe the disease incidence has increased since the ban on in-feed “growth promoting” antibiotics. Because of this, many recent studies of NE have focused on finding different ways to control the disease, and on understanding its pathogenesis. Frustratingly, reproduction of the disease has proven impossible for some researchers. This review describes and discusses factors known to be important in reproducing the disease experimentally, as well as other considerations in reproducing the disease. The critical bacterial factor is the use of virulent, netB-positive, strains; virulence can be enhanced by using tpeL- positive strains and by the use of young rather than old broth cultures to increase toxin expression. Intestinal damaging factors, notably the use of concurrent or preceding coccidial infection, or administration of coccidial vaccines, combined with netB-positive C. perfringens administration, can also be used to induce NE. Nutritional factors, particularly feeding high percentage of cereals containing non-starch polysaccharides (NSP) (wheat, rye, and barley) enhance disease by increasing digesta viscosity, mucus production and bacterial growth. Animal proteins, especially fish meal, enhance C. perfringens proliferation and toxin production. Other factors are discussed that may affect outcome but for which evidence of their importance is lacking. The review compares the different challenge approaches; depending on the aim of particular studies, the different critical factors can be adjusted to affect the severity of the lesions induced. A standardized scoring system is proposed for international adoption based on gross rather than histopathological lesions; if universally adopted this will allow better comparison between studies done by different researchers. Also a scoring system is provided to assist decisions on humane euthanasia of sick birds.  相似文献

The aim of this study was to determine the prevalence, variability with host age, and the genotypes of species of Cryptosporidium in cattle from 15 dairy farms in Qazvin province, Iran. Fecal samples, collected from 272 cattle during May 2006 to December 2007, were characterized microscopically. Oocysts from 51 positive samples were analyzed using PCR assay of 18S SSU rRNA, restriction fragment length polymorphism (RFLP) and sequencing. We identified 72.6% of the positive samples as Cryptosporidium parvum, 17.7% as Cryptosporidium andersoni, 7.8% as Cryptosporidium bovis and 1.9% as a novel genotype of C. parvum possessing a single mutation on MboII restriction. An infection rate of 19.5% of C. parvum among 174 pre-weaned calves was significantly higher than the 3.1% among 98 post-weaned calves (P < 0.0006). This is the first report of C. bovis and the new subgenotype of C. parvum in Iranian cattle.  相似文献

Studies have shown the detection of emerging contaminants (ECs), of which pharmaceuticals are a subset, in surface waters across the United States. The objective of this study was to develop methods, and apply them, to evaluate the potential for food chain transfer when EC-containing waters are used for crop irrigation. Greenhouse experiments were performed in which select food crops were irrigated with water spiked with three antibiotics. Field experiments, at two different sites, were conducted. Select crops were irrigated with wastewater effluent known to contain ECs, EC-free well water, and Colorado River water containing trace-level ECs. The results of the greenhouse studies show the potential for uptake of one or more of the antibiotics evaluated, albeit at very low levels. In those food crops watered with wastewater effluent, only an industrial flavoring agent, N,N'-dimethylphenethylamine (DMPEA), was consistently found. None of the evaluated contaminants were found in crops irrigated with Colorado River water.  相似文献

1. A 6-week study was conducted to investigate the effects of phytase and hydroalcoholic extract of Withania somnifera root (WS) on productive performance and bone mineralisation of laying hens in the late phase of production.

2. Diets were arranged factorially (3?×?2?×?2) and consisted of a positive control with adequate Ca (4·37%) and nonphytate P (NPP; 0·39%) and a negative control diet with Ca (4·06%) and NPP (0·36 %); three concentrations of Withania somnifera (0, 65 and 130?mg/kg diet); and two concentrations of microbial phytase (0 and 300 U/kg diet).

3. A total of 144 72-week-old Hy-Line W36 laying hens were randomly assigned to the 12 treatment groups. Each treatment was replicated 4 times (4 x 3 hens). Egg production and egg weight were recorded daily, while feed intake and egg quality traits were recorded every two weeks. Bone quality traits were evaluated at the end of experiment.

4. Withania somnifera supplementation increased egg production and lowered egg weight only in the second two weeks of the experiment. Addition of phytase significantly depressed specific gravity of the eggs for the entire experiment period. No dietary treatment effects were observed on egg shell thickness and yolk weight.

5. Withania somnifera at 130?mg/kg did not affect feed intake. The hens fed on the positive control diet had higher albumen weight than the negative control diet in the second two-week period. Supplementation of the positive control diet with 65?mg/kg Withania somnifera in the absence of phytase significantly improved shell weight compared with the negative control (5·779 vs. 5·273?g respectively).

6. Supplementing Withania somnifera significantly improved Ca and P retention in tibia bone. In addition, an increase in tibia bone P was observed with phytase supplementation. There were significant interactions between Withania somnifera content and phytase for tibia bone Ca and P.

7. The results of this experiment indicated that dietary Withania somnifera has beneficial effects on tibia bone Ca and P content, and phytase improved tibia bone P retention without adverse effects on productive performance.  相似文献
