The Puccinia striiformis f. sp. tritici (Pst) pathotype, 134 E16A+, detected in 2002 in Australia, produced relatively lower and higher adult plant stripe rust responses, respectively, on cultivars Kukri and Janz in comparison to the pre-2002 Pst pathotype 110 E143A+. Molecular mapping of adult plant stripe rust response variation among 180 Kukri/Janz-derived doubled haploid lines over 4 years, two each with Pst pathotypes 110 E143A+ and 134 E16A+, was performed. QYr.sun-7B and QYr.sun-7D were consistently contributed by Kukri and Janz, respectively. QYr.sun-7D corresponded to the genomic location of Yr18 and QYr.sun-7B remains to be formally named. QYr.sun-1B, QYr.sun-5B, and QYr.sun-6B were detected during more than one season irrespective of the Pst pathotypes used, whereas QYr.sun-3B was identified only during the 2003 crop season. QYr.sun-1A contributed by Janz, and QYr.sun-2A from Kukri, were detected only against Pst pathotypes 110 E143A+ and 134 E16A+, respectively. The DH lines showing better resistance than the either parent carried combinations of 4 to 6 QTL. These lines are currently being used as stripe rust resistance donors in wheat breeding programs.  相似文献

Given the importance of potassium (K) and phosphorus (P) contents to wheat yield and grain quality, and the very little experience that has been gained on nutritional monitoring of other than nitrogen using remotely sensed technologies, a study was undertaken to explore the possibility of identifying these mineral stresses using spectral data. Canopy spectra and biophysical data were collected from commercial and experimental fields in India and Israel. Traditional and newly developed vegetation indices, together with Partial Least Squares (PLS) regression models, were calculated in order to predict potassium and phosphorus contents from the wheat canopy spectral data. Results show that the application of PLS and specific narrow bands vegetation indices reached significant levels of accuracy in the retrieval of K and P levels, in comparison to traditional broad band indices. Additionally, it was observed that a significant improvement is obtained when the mineral total content is considered instead of the relative content. Therefore it was suggested that the biomass should also be retrieved from the spectral data. Finally, as very different crop conditions were included in this study, it was possible to confirm that the level of accuracy in the retrieval of K and P levels is related to the quality and variability of the data used for calibrating the models.  相似文献

A field study was conducted at the ICAR-Indian Agricultural Research Institute, New Delhi, to study the response of spring wheat to boron applied as boron-coated urea (BCU) and the effect of boron (B) coating on the use efficiency of applied fertilizer urea nitrogen. The BCU (0.5% B) significantlyrecorded the highest increase in leaf area index (30.2%), spike length (12.5%), number of spikes (10.9%), filled grains (15.7%) and grain weight (16.3%) per spike in spring wheat than uncoated prilled urea (PU). Furthermore, the application of 0.5% BCU enhanced the grain and straw yields of spring wheat by 11% and 10.6% over uncoated PU. The BCU (0.5% B) also significantly increased the N concentration and N uptake both in wheat grain and straw. The BCU (0.5% B) recorded 24.8% higher recovery efficiency of applied nitrogen and an increase of 3.6 kg grain kg?1 N applied in agronomic efficiency compared to uncoated PU. The net returns (USD915.1 ha?1) and benefit:cost ratio (1.40) werealso the highest with 0.5% BCU, significantly higher than uncoated PU. Therefore, 0.5% boron coating onto PU is recommended for increased productivity and enhanced efficiency of applied fertilizer nitrogen for spring wheat in Indo-Gangetic plains of India.  相似文献

In the case of high valuable individuals with very precious genetic material, widening the genetic pool including gametes with poor morphological characteristics, as cumulus‐denuded oocytes (CDOs), could be an option. To improve the in vitro culture of low‐competence feline CDOs, an enriched three‐dimensional (3D) system in association with competent cumulus–oocyte complexes (COCs) was developed. For this purpose, domestic cat CDOs were cultured with or without companion COCs in the 3D barium alginate microcapsules. The overall viability and the meiotic progression of feline CDOs cocultured with COCs or cultured separately in 3D or in 2D (traditional microdrops) system were compared. The 3D system was able to support viability and meiotic resumption of the feline oocytes, as well as the 2D microdrops. In 3D microcapsules, the presence of COCs resulted in a higher viability of CDOs (91.1%, < .05), than that obtained without COCs or in 2D microdrops (71.2% and 67.3%, respectively), but the percentages of meiotic resumption were similar of those of CDOs cultured separately (55.4% vs. 40.4%, p > .05). It is notable that the presence of CDOs seemed to enhance the meiotic progression of the associated COCs. In conclusion, the 3D barium alginate microcapsules are a suitable system for feline oocytes in vitro culture, but more specific enriched conditions should be developed to improve the CDOs full competence in vitro.  相似文献

Humoral and cell-mediated immune responses to the capsular polysaccharide (CPS) of M. dispar and polygalacturonic acid (pGaIU—a structurally similar polysaccharide) were investigated in calves experimentally infected with Mycoplasma dispar and in mice immunized with CPS or pGaIU. Sera, tracheobronchial lavage and nasal fluids, collected before and after infection in calves, were checked for the presence of anti-CPS and anti-pGaIU antibodies. The sera from mice injected with CPS or pGaIU were checked for different classes of anti-CPS and anti-pGaIU antibodies. Peripheral blood lymphocytes from calves and splenic lymphocytes from mice were monitored for specific proliferative responses to CPS and pGaIU. At about 2 weeks post-infection, anti-CPS IgM response in serum, anti-CPS and anti-pGaIU IgM and IgA response in lavage fluid and lymphocyte proliferative response was seen in the calves. Mice immunized with CPS and pGaIU gave exclusively IgM responses. No secondary response was seen in mice immunized with CPS in contrast to mice immunized with pGaIU. Antibodies cross-reactive with pGaIU were present in the sera of CPS-immunized mice but antibodies cross-reactive with CPS were not found in pGaIU-immunized mice. No significant blastogenic response was shown by mouse splenocytes to CPS or pGaIU.  相似文献

The chemoprophylactic efficacy of long-acting oxytetracycline was determined in 13 susceptible ewes infected with Anaplasma ovis. The drug was administered intramuscularly at a dose rate of 20 mg kg-1 body weight during the prepatent period. When the ewes were exposed to an equivalent homologous challenge on Day 45 post-infection, each showed a mild or inapparent reaction. The host reactions, i.e., body temperature, parasitaemia, packed-cell volume (PCV), haemoglobin, humoral and cell mediated immunity (CMI) were also studied. The results indicate that the use of oxytetracycline during the incubation period would minimise clinical signs of an Anaplasma infection; this may be due to an increasing CMI response.  相似文献

A 1‐year‐old female Labrador retriever was referred with a few days history of haematic‐like vulvar discharge. Physical examination, vaginal inspection and palpation did not reveal any remarkable finding. Transabdominal ultrasound showed echogenic fluid accumulation in the vagina suggesting haemato‐pyocolpos. An exploratory laparotomy was performed: a well‐delimited ectasic vagina was identified. Ovariohysterectomy and partial vaginectomy and vaginoplasty were performed to spay the bitch and to remove the ectasic vagina. Post‐operative recovery and 12‐month follow‐up were uneventful. Clinical, morphological and histological findings were consistent with a congenital abnormality of the muscular layer of the vagina complicated by haemato‐pyocolpos. The disorganization of the vaginal tunica muscularis may have acted as locus minoris resistentiae in the vaginal wall. The organ was dilated and atonic due to the gradual accumulation of physiological fluids complicated by an overgrowth of genital bacteria. This congenital disorder has to be taken into account as differential diagnosis of haemato‐pyocolpos with vaginal discharge in young bitches.  相似文献

The pharmacokinetics of erythromycin was studied in five lactating dairy cows following single intramammary infusion of 300 mg erythromycin in each of two quarters per cow with specific mastitis. Levels of erythromycin in plasma and quarter milk samples were measured by agar plate diffusion assay using Micrococcus luteus (ATCC 9341) as the test organism. Erythromycin level in plasma reached a peak concentration value (C max) of 0.07 ± 0.01 μg/ml at 30 min; thereafter, levels declined gradually to reach 0.05 ± 0.00 μg/ml 12 h post drug administration. The pharmacokinetic profile of the drug revealed mean absorption half life (t 1/2ka) as 0.26 ± 0.05 h. The drug was eliminated slowly with elimination half-life (t 1/2β) of 13.75 ± 0.35 h and elimination rate constant (k el) of 0.04 ± 0.00 h−1. The volume of distribution based on the zero-time plasma concentration intercept of the least-squares regression line of the elimination phase (V d(B)) was 0.032 L/kg. The drug crossed to untreated quarters also; mean drug levels of 0.20 ± 0.07, 0.23 ± 0.07, 0.17 ± 0.04, and 0.17 ± 0.04 μg/ml were found at 3, 6, 8 and 12 h, respectively. The mean drug concentration for treated quarters was measured as 22.97 ± 2.31 μg/ml milk at first milking (12 h) following drug infusion. No apparent adverse reaction was seen in cows administered erythromycin. It is concluded that following intramammary infusion erythromycin diffuses readily and extensively in various body fluids and tissues and adequate concentration is maintained in udder tissues for at least 12 h post intramammary administration. Thus, erythromycin may be recommended for local therapy of acute mastitis caused by Gram-positive bacteria in lactating dairy cows.  相似文献
