Streptococcus suis is an important pathogen in the swine industry. This article is the first to report the occurrence, risk factors, serotype distribution, and antimicrobial susceptibility of S. suis recovered from employees and environmental samples of swine slaughterhouses in Brazil. Tonsillar swabs from all 139 pig-slaughtering employees and 261 environmental swabs were collected for detection of S. suis and serotyping by monoplex and multiplex polymerase chain reaction, respectively. Antimicrobial susceptibility was determined by the disk-diffusion method. Although S. suis was not detected in any of the tested employees, it was isolated from 25% of the environmental samples. Significant differences (P < 0.05) in the occurrence of S. suis were observed between slaughterhouses and between areas of low, medium, and high risk. The most frequent serotypes were 4 and 29, each accounting for 12% of the isolates, followed by 5, 12, 21, and 31, each accounting for 6%. High rates of susceptibility to the antimicrobials doxycycline (100%), ceftiofur (94%), ampicillin (81%), and cephalexin (75%) were observed. However, multidrug resistance was observed in all the isolates. Because S. suis is present in the environment of swine slaughterhouses, on carcasses and knives, as well as on the hands of employees in all areas, all employees are at risk of infection. 相似文献

Lectins are a structurally heterogeneous group of proteins that have specific binding sites for carbohydrates and glycoconjugates. Because of their biotechnological potential, lectins are widely used in biomedical research. The present study aimed to evaluate the healing potential of the lectin isolated from the marine red alga Bryothamnion seaforthii (BSL). The lectin was purified using ion exchange chromatography with DEAE cellulose and characterized using tandem mass spectrometry. For healing tests, skin wounds were induced in the dorsal thoracic region of mice. These animals were randomly divided into three groups and subjected to topical treatment for 12 days with BSL, bovine serum albumin and 150 mM NaCl. To evaluate the potential of each treatment, the animals were anesthetized and sacrificed on days 2, 7 and 12, respectively. The parameters evaluated included the wound area, the proportion of wound closure and the histological diagnosis. The wound closure was more effective with BSL (Postoperative Day 7 and 12) than controls. The luminal epithelium was completely restructured; the presence of collagen in the dermis and the strongly active presence of young skin annexes demonstrate the potential of treatment with BSL compared with controls. Our findings suggest that BSL has pro-healing properties and can be a potential medical process in the treatment of acute wounds. 相似文献

Pittosporum undulatum from the Azores archipelago was characterized by both molecular and volatiles analyses. Inter simple sequence repeat (ISSR) evaluation was performed based on a random sampling of 77 individuals from a total of 123 samples collected on all the Azorean islands. Molecular studies grouped the samples mainly according to the geographical collection site, with some exceptions. Leaf volatiles were isolated by distillation-extraction and analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) from all the 123 samples. Cluster analysis based on the leaf volatiles chemical composition defined three main clusters, not related to sample site collection, and was based mainly on the relative amounts of limonene (3-89%), sabinene (0.1-64%) and terpinen-4-ol (traces-43%). Clusters obtained from both molecular studies and volatiles composition were not superimposable and no correlation between ISSR markers and volatile oils could be drawn. The results suggest that there is a high genetic variability among individuals of P. undulatum that can explain the invasive ability of this species in the Azores archipelago. The detailed characterization of this alien weed species may assist finding potential commercial uses as well as management strategies to help its eradication and/or control. 相似文献

Weissella ceti is an emerging bacterial pathogen that affects rainbow trout, Oncorhynchus mykiss (Walbaum), farms. The aims of this study were to genotype W. ceti strains isolated from distinct geographical origins and to determine the efficacy of an oil-adjuvanted vaccine against the disease. Between 2010 and 2012, outbreaks were recorded in five Brazilian farms, and 34 W. ceti isolates were genetically characterized by repetitive extragenic palindromic PCR, enterobacterial repetitive intergenic consensus sequences PCR and pulsed-field gel electrophoresis. Two different W. ceti vaccines were tested: an aqueous-based whole-cell inactivated vaccine (bacterin) and oil-adjuvanted vaccine. Their efficacy was evaluated in rainbow trout at 30 and 60 days post-vaccination (d.p.v.). W. ceti was found to be a highly homogeneous population in Brazil, with clonally related genotypes. Oil-adjuvanted vaccine exhibited the best (P < 0.05) protection against disease, reaching relative percentage survival (RPS)values of 92% at 30 and 60 d.p.v. Bacterin resulted in RPS values of 67% and 58% at day 30 and 60, respectively. The oil-adjuvanted vaccine provided effective protection against W. ceti infection in rainbow trout. 相似文献

Portable accumulation chambers (PACs) enable gaseous emissions from small ruminants to be measured over a 50-min period; to date, however, the repeatability of consecutive days of measurement in the PAC has not been investigated. The objectives of this study were 1) to investigate the repeatability of consecutive days of gaseous measurements in the PAC, 2) to determine the number of days required to achieve precise gaseous measurements, and 3) to develop a prediction equation for gaseous emissions in sheep. A total of 48 ewe lambs (c. 10 to 11 mo of age) were randomly divided into four measurement groups each day, for 17 consecutive days. Gaseous measurements were conducted between 0800 and 1200 hours daily. Animals were removed from perennial ryegrass silage for at least 1 h before measurements in the PAC, and animals were assigned randomly to each of the 12 chambers. Methane (CH4; ppm) concentration, oxygen (O2; %), and carbon dioxide (CO2; %) were measured at three time points (0, 25, and 50 min after entry of the first animal into the first chamber). To quantify the effect of animal and day variation on gaseous emissions, between-animal, between-day, and error variances were calculated for each gaseous measurement using a linear mixed model. The number of days required to gain a certain precision (defined as the 95% confidence interval range) for each gaseous measurement was also calculated. For all three gases, the between-day variance (39% to 40%) accounted for a larger proportion of total variance compared with between-animal variance, while the repeatability of 17 consecutive days of measurement was 0.36, 0.31, and 0.23 for CH4, CO2, and O2, respectively. Correlations between consecutive days of measurement were strong for all three gases; the strongest correlation between day 1 and the remaining days for CH4, CO2, and O2 was 0.71 (days 1 and 6), 0.77 (days 1 and 2), and 0.83 (days 1 and 5), respectively. A high level of precision was achieved when gaseous measurements from PAC were taken over three consecutive days. The prediction equation overestimated gaseous production for all three gases: the correlations between actual and predicted gaseous output ranged from 0.67 to 0.71, with the r2 ranging from 0.45 to 0.71. The results from this study will aid the refinement of the protocol for the measurement of gaseous emissions in sheep using the PAC. 相似文献

A growth trial was conducted to examine the effect of dietary digestible energy (DE) content on methionine (Met) utilization and requirement in juvenile Nile tilapia (Oreochromis niloticus). Ten iso‐nitrogenous (288 g kg?1 protein) practical diets, with two DE levels (10.9 MJ kg?1; 12.4 MJ kg?1) and five methionine supplementation levels (0, 1, 2, 4 and 6 g kg?1), were hand‐fed twice daily to triplicate groups of Nile tilapia (initial body weight 8.95 ± 0.06 g) for 8 weeks. Weight gain (WG) and specific growth rate (SGR) increased significantly with increasing dietary methionine concentration at the same DE content (P < 0.001). At the same dietary methionine level, WG and SGR of fish fed high‐DE diets were significantly higher than that of fish fed low‐DE diets (P = 0.0001), although no interaction was found between dietary DE and methionine supplementation. Based on quadratic regression analysis between dietary methionine concentration and weight gain, optimal methionine requirement for maximum growth, expressed as g Met required kg?1 diet (low‐ versus high‐DE diets), increased as diet DE concentration increased (7.34 versus 9.90 g kg?1 diet, respectively; with cysteine 4.70 g kg?1 diet). The results indicated that diet DE content affects methionine utilization and requirement in juvenile Nile tilapia, fish fed high‐DE diets required more methionine for maximum growth. 相似文献

The undulate ray Raja undulata Lacepède is a coastal species common along the north‐eastern Atlantic Ocean and Mediterranean Sea and is highly accessible to coastal fisheries. Between 2009 and 2015, the species was under a European Union (EU) fisheries moratorium that hampered the collection of data to assess its stock status in Portuguese waters. After that period, a small experimental EU fishing quota was set for Portugal enabling collection of fishery data under a fishermen self‐sampling scenario. Based on the data collected, R. undulata abundance was estimated along the Portuguese continental coast through the application of a N‐mixture model and incorporating environmental factors. The results support the species coastal and patchy nature across the study area with higher abundances estimated in areas associated with shallow sandy bottoms as the Southwest region. The present work constitutes an important step for the management of this fishery resource, in particular concerns about its abundance trends over time and its spatial distribution and habitat requirements. 相似文献
