Isoamylases in clinically normal dogs   总被引:1,自引:0,他引:1  
Isoenzymes of canine serum amylase were evaluated by electrophoresis on cellulose acetate membranes in a discontinuous buffer system. Two isoamylase groups were identified in the serum of clinically normal dogs. Most of the serum amylase activity was the more cathodal isoamylase. The anodal isoamylase was rarely observed in serum from normal dogs and, when present, accounted for little of the enzymatic activity. Amylase activities of various tissues were determined and isoenzymes were identified. The anodal isoenzyme was found in the pancreas and uterus. The cathodal isoamylase had its origin mainly from the intestinal tract. Activities of other tissues were not greater than was serum amylase activity. Effects of feeding upon total serum amylase activity and isoenzyme composition also were examined over a period of 5 minutes to 7 hours. Feeding did not markedly alter the serum amylase activity.  相似文献

The expansion of the aquaculture production is restricted due to the pressure it causes on the environment by the discharge of waste products in the water bodies and by its dependence on fish oil and fishmeal. Aquaculture using bio-flocs technology (BFT) offers a solution to both problems. It combines the removal of nutrients from the water with the production of microbial biomass, which can in situ be used by the culture species as additional food source. Understanding the basics of bio-flocculation is essential for optimal practice. Cells in the flocs can profit from advective flow and as a result, exhibit faster substrate uptake than the planktonic cells. The latter mechanisms appear to be valid for low to moderate mixing intensities as those occurring in most aquaculture systems (0.1–10 W m− 3). Yet, other factors such as dissolved oxygen concentration, choice of organic carbon source and organic loading rate also influence the floc growth. These are all strongly interrelated. It is generally assumed that both ionic binding in accordance with the DLVO theory and Velcro-like molecular binding by means of cellular produced extracellular extensions are playing a role in the aggregation process. Other aggregation factors, such as changing the cell surface charge by extracellular polymers or quorum sensing are also at hand. Physicochemical measurements such as the level of protein, poly-β-hydroxybutyrate and fatty acids can be used to characterize microbial flocs. Molecular methods such as FISH, (real-time) PCR and DGGE allow detecting specific species, evaluating the maturity and stability of the cooperative microbial community and quantifying specific functional genes. Finally, from the practical point of view for aquaculture, it is of interest to have microbial bio-flocs that have a high added value and thus are rich in nutrients. In this respect, the strategy to have a predominance of bacteria which can easily be digested by the aquaculture animals or which contain energy rich storage products such as the poly-β-hydroxybutyrate, appears to be of particular interest.  相似文献

Two methods are described for the assay of the enzyme that transfers a methyl group from methyl parathion (O,O-dimethyl-O, p-nitrophenyl phosphorothionate) to glutathione. A colorimetric assay depends on the measurement of alkali-soluble, chloroform-insoluble aromatic nitro compounds in the reaction mixture and a more rapid and sensitive direct spectrophotometric assay is described that uses the double wavelength Perkin-Elmer 356 Spectrophotometer. The utility of both methods has been demonstrated by the measurement of some kinetic constants and the distribution of the enzyme among some insects and vertebrates.  相似文献

Summary To give an impression of the usefulness of indirect haemagglutination (IHA) in the diagnosis of lungworm infections in cattle under practical conditions, five calves vaccinated against Dictyocaulus viviparus and five unvaccinated calves were periodically subjected to clinical, parasitological, and serological examinations over a period of seven months. All calves grazed on a lungworm-infected plot. 82% of the observations in unvaccinated calves, which were positive with respect to one or more of the used parameters, concerned IHA-positive animals which, however, showed negative results with the parasitic parameters. The titre variation of the serological examination was a further indication of the fact that the IHA detected antibodies against lungworm antigens. No indications of false positive reactions were obtained. An investigation carried out on 46 farms on the correlation between serological and clinical findings on lungworm infections revealed a positive correlation in 80% of the groups between results obtained with both methods. The authors consider that IHA offers good prospects for the diagnosis of lung-worm infections.  相似文献

Four new iodobenzene-containing dipeptides (1–4), a related bromotryptophan-containing dipeptide (5), and an iodophenethylamine (6) were isolated from the ascidian Aplidium sp. collected off the coast of Chuja-do, Korea. The structures of these novel compounds, designated as apliamides A–E (1–5) and apliamine A (6) were determined via combined spectroscopic analyses. The absolute configuration of the amino acid residue in 1 was determined by advanced Marfey’s analysis. Several of these compounds exhibited moderate cytotoxicity and significant inhibition against Na+/K+-ATPase (4).  相似文献

Evidence that dietary lycopene decreases the risk for a number of health conditions has generated new opportunities for the addition of lycopene to functional foods. This work examined the potential of oil-in-water emulsions as a lycopene delivery system for foods. Oil-in-water emulsions containing lycopene were prepared using different kinds of surfactant (cationic, anionic, and nonionic) and oil types (corn oil, stripped corn oil, and hexadecane). The formation of fatty acid oxidation products and the degradation of lycopene and tocopherol were then monitored. Fatty acids and lycopene had greater stability in oil-in-water emulsions stabilized by cationic dodecyltrimethylammonium bromide (DTAB) or nonionic polyoxyethylene (23) lauryl ether than by anionic sodium dodecyl sulfate (SDS). Oxidative stability in the corn oil-in-water emulsions stabilized by SDS was in the following order: tocopherolhexadecane>tocopherol-stripped corn oil. Lycopene degradation rates were similar in emulsions with and without fatty acids, suggesting that lycopene loss was independent of the presence of fatty acids. These results suggest that the stability of lycopene in oil-in-water emulsions could be inhibited by altering the emulsion droplet interface and by the presence of tocopherols.  相似文献
