AIM:To explore the effects of PMA(phorbol-12-myristate-13-acetate, a tumor promoter, mimicking the action of diacylglycerol on PKC)and laminin on the adhesion and the proliferation of human hepatocellular carcinoma cells, and provide a new clue to liver cancer treatment.METHODS:Human hepatocellular carcinoma cell line(BEL-7402)was used to identify the endogenous laminin and protein kinase C-α(PKC-α) expression, and the effects of laminin and PMA on the adhesion and the proliferation were also investigatedin vitro.RESULTS:By the effect of exogenous laminin, human hepatocellular carcinoma cell (BEL-7402) possessed endogenous laminin expression and increased the adhesion and the proliferation, which was showed the synergistic action by the effect of PMA in combination. By the action of PMA alone, the proliferation and the PKC-α expression increased by exogenous laminin were decreased, and the adhesion and the endogenous laminin expression were increased.CONCLUSIONS:The finding suggested that the adhesion and the proliferation of human hepatocellular carcinoma cell were closely related to the effects of endogenous or exogenous laminin, which were associated with cPKC-α activity. Therefore, the application of anti-laminin antibody in combination with PKC antagonist might be a new clue to find out the therapy for liver cancer.  相似文献

通过对渭河阶地全新世土壤的野外考察和粒度成分的高分辨率分析研究，揭示了该区域褐色土的成壤作用过程与季风气候变化的关系。研究表明渭河第二级阶地褐色土形成在全新世气候最适宜期（8500－3100a B.P.），从8500a B.P.开始该土壤开始发育，老官台文化兴起。从3100a B.P.开始的气候干旱化和沙尘暴堆积形成了黄土L。层。西周时期对应于该干旱期的初期。  相似文献

本研究通过PCR扩增和六种限制性内切酶（AluⅠ，HinfⅠ，MboⅠ，RsaⅠ，HaeⅢ和PvuⅡ）酶切，对国内害虫防治上常用的几种昆虫病原线虫，包括斯氏属S.car-pocapsae,S.feltiae和S.glaseri以及异小杆属H.bacteriophora,H.zealandica,H.indicus和H.megidis等8个品系rDNA-ITS进行分析。建立起可以区分各线虫种的标准RFLP图谱。该方法快速简便，稳定可靠，需要的样品量少。可以用于新鲜的，或冻存的样品，甚至分析单条的线虫，不仅可进行昆虫病原线虫的快速分类鉴定。而且进一步可以应用于线虫田间释放的辅助监测。实际田间感染率的测定和线虫毒力的比较。  相似文献

对山西菜田捕食性食蚜蝇优势种及对菜蚜的控制效果进行了研究，菜田常见捕食性食蚜蝇有黑带食蚜蝇，大灰食蚜蝇，斜斑鼓额食蚜蝇，野食蚜蝇，四条小食蚜蝇，短翅细腹食蚜蝇6种，以黑带食蚜蝇种群数量最大，平均占全年发生总量的36.3%，通过对食蚜蝇产卵量，卵化率，幼虫期捕食量和羽化率测试，筛选出的黑带食蚜蝇，大灰食蚜蝇，斜斑鼓额食蚜蝇幼虫对菜蚜均有较强的捕食能力，其中黑带食蚜蝇为山西菜田菜蚜捕食性食蚜蝇优势种，用黑带食蚜蝇三龄幼虫对菜蚜进行防治试验，结果以1：180防效最佳，72h后控制效果达93.5%。  相似文献

 于温室盆栽条件下研究了丛枝菌根(AM) 真菌Gigaspora rosea、Glomus mosseae 和Glomus versi-forme 对芋(Colocasia esculenta) 组织培养苗移栽成活率、矿质营养、光合速率及生长的影响。结果表明, 接种AM真菌能提高芋组织培养幼苗移栽成活率和叶片光合速率, 降低气孔阻力; 其叶片和根内氮、磷、钾含量和生长量显著高于不接种对照。认为接种有效AM真菌是促进组织培养苗健康生长的重要技术。  相似文献

本试验对 6个不同滑菇品种进行了菌丝体生长和制种观察 ;在栽培试验过程中进行了多点小试和中试 ,初选出Ph0 8号和Ph0 6号两个品种。Ph0 8的单盘产量比CK1高出 2 5 8% ,比CK2 高出 35 3% ;Ph0 6的单盘产量比CK1高出 2 1 5 % ,比CK2 高出 30 6 %。它们均属早生品种 ,适合辽宁地区的气候特点 ,在幼菇期菌盖呈半球形、桔红色、附着的粘液多 ,菌柄比对照短 ,菇体健壮 ,符合商品收购和产品加工的要求  相似文献   

Metallothionein inhibits homocysteine-induced proliferation of rat vascular smooth muscle cells

AIM:To investigate the effect of metallothionein(MT) on proliferation of rat vascular smooth muscle cells (VSMCs) stimulated by homocysteine and its mechanism. METHODS:VSMCs proliferation was measured by [³-H]-TdR incorporation, mitogen-activated protein kinase(MAPK)activity were determined by immunoprecipitation method, the intracellular contents of MT and malondialdehyde (MDA)were assayed by -hemoglobin saturation method and TBA reaction, respectively, and lactate dehydrogenase (LDH) leakage was measured by NADH oxidation. RESULTS:Hcy(10^-6-10^-4 mmol/L) stimulated [³-H]-TdR incorporation by the VSMCs in a concentration-dependent manner. Compared with control, [³-H]-TdR incorporation in VSMCs treated with 0.1 mmol/L Hcy was increased by 4.2 fold (P＜0.01). Meanwhile, Hcy enhanced MAPK activity, MDA formation and LDH release (P＜0.01)in a concentration-dependent manner. Treatment of VSMCs with MT alone did not change above parameters, compared with control. However, MT (10^-6-10^-4 mol/L)attenuated significantly Hcy-stimulated proliferation of VSMCs (P＜0.01)in a concentration-dependent manner. And MT inhibited obviously Hcy-induced activation of MAPK activity, MDA formation and LDH release. Preincubation of VSMCs with 0.5 mmol/L ZnCl₂ for 6 h induced an increase cellular MT content by 5.7-fold (P＜0.01). The MT-overexpressed VSMCs resisted Hcy-stimulating action on MAPK activity, MDA formation and LDH leakage (P＜0.01). CONCLUSION:These results show that MT has an inhibitory effect on Hcy-induced VSMCs proliferation, and that MT could inhibit Hcy-stimulated MAPK activity and lipid peroxidation.  相似文献

AIM: To study the electrophysiological characteristics of ion channels of stem cell derived cardiomyocytes(SCDC) of mouse. METHODS: Embryonic stem cells of D₃ line(ES-D₃) were cultured on the MEF feeder layer with BRL conditioned medium, and fetal mouse heart cells(FMHC)were cultured in vitro. Then ES-D₃ cells were induced to differentiate into many kinds of cells. SCDC were harvested on day 12 after differentiation initiating and identified by electro-microscope and immunocytochemistry. SCDC and FMHC were prepared for the patch-clamp research. Sodium and calcium currents together were elicited and compared between SCDC and FMHC. RESULTS: The current characteristics of sodium and calcium channels of SCDC were very similar to FMHC. CONCLUSION: The functional expression of ion channels occurred during ES-D₃ cells differentiation and the electrophysiological characteristics of sodium and calcium channels of SCDC are very similar to FMHC.  相似文献

研究表明，金耳(Tremella aurantialba)液体发酵最适培养基组成为玉米粉2％，蔗糖3％，酵母膏0．1％，KH2PO40．1％，MgSO40．05％；培养基最佳初始pH、250mL三角瓶装量、接种量、种子菌龄分别是3．5、80mL、10％和120h；用10L发酵罐发酵时，最佳培养条件是种子种龄144h、28℃、接种量10％、通风量2．5L／min。  相似文献
