Efficient enzymatic conversion of crystalline polysaccharides is crucial for an economically and environmentally sustainable bioeconomy but remains unfavorably inefficient. We describe an enzyme that acts on the surface of crystalline chitin, where it introduces chain breaks and generates oxidized chain ends, thus promoting further degradation by chitinases. This enzymatic activity was discovered and further characterized by using mass spectrometry and chromatographic separation methods to detect oxidized products generated in the absence or presence of H(2)(18)O or (18)O(2). There are strong indications that similar enzymes exist that work on cellulose. Our findings not only demonstrate the existence of a hitherto unknown enzyme activity but also provide new avenues toward more efficient enzymatic conversion of biomass. 相似文献

Parasites can, in theory, have large impacts on the survival of fish populations. One method to evaluate such impacts on anadromous species is to apply manipulative field experiments in which parallel groups of antiparasitically treated and non‐treated fish are simultaneously released and then subsequently recaptured as returning adults. A systematic review and meta‐analysis on all such Norwegian studies on Salmo salar provided a data set for the time period 1996 to 2011 on 118 release groups comprising 657 624 fish released and 3989 recaptured. The overall risk ratio (RR) was estimated to be 1.18 (95% CI: 1.07–1.30). The effect varied strongly between groups, (Higgins I2 = 40.1%). Over 70% of this heterogeneity could be explained by the release location, time period and baseline survival. The most important predictor variable was baseline survival. In groups with low recapture in the control group (low baseline survival), the effect of treatment was high (RR = 1.7), while in groups with high recapture in the control group (high baseline survival), there was no effect of treatment (RR ~ 1.00). The most prevalent parasite in the region affected by the drugs administered was Lepeophtheirus salmonis. Hence, the meta‐analysis supports the hypothesis that L. salmonis contributes to the mortality of S. salar during outward migration. However, the effect of treatment was not consistent, but was evidently strongly modulated by other risk factors. The results suggest that the population‐level effects of parasites cannot be estimated independently of other factors affecting the marine survival of S. salar. 相似文献

Greywater is a potential resource of water that can be improved to meet the quality needed for irrigation. This study evaluated the performance of bark, activated charcoal, polyurethane foam and sand filters in removing biochemical oxygen demand (BOD5), surfactants, phosphorus, nitrogen and microbial indicators from greywater during start-up and steady state. In column experiments, 0.6?m high filters (diameter 20?cm) were fed for 113?days with artificial greywater at a hydraulic loading rate of 0.032?m3?m?2?day?1 and an organic loading rate of 0.014?kg BOD5 m?2?day?1. Bark and activated charcoal efficiently reduced the concentrations of organics (BOD5), surfactants (methylene blue active substances??MBAS), total phosphorus (Tot-P) and total thermotolerant coliform numbers, while sand and foam were less efficient. Bark, activated charcoal, foam and sand reduced influent BOD5 by 98, 97, 37 and 75?%; MBAS by >99, >99, 73 and 96?%; Tot-P by 97, 91, 36 and 78?%; and total nitrogen by 19, 98, 13 and 5?%, respectively. BOD5 and MBAS were efficiently reduced directly from start-up by bark and activated charcoal, while foam needed 30?days to achieve about 50?% reduction in BOD5. Bark was the most efficient filter in reducing thermotolerant faecal coliforms (2.4 log10), while foam achieved the lowest reduction (0.5 log10). Overall, bark and activated charcoal filters appeared to be the most suitable filters for improving greywater quality to reach irrigation quality in terms of organic matter reduction. Performance of these filters under higher and fluctuating loadings and the long-term sustainability of the filter materials need further investigation. 相似文献

Rye was milled into flours having extraction rates between 100 and 65%. The nutritive value of the various fractions was studied by chemical analyses and in balance trials with rats. The concentration of essential nutrients decreased when the extraction rate was lowered. The lysine content (g/16 g N) was 4.23 in whole rye, but only 3.76 in the 65% extraction flour, and a corresponding reduction in biological value was found. A reduction of 50% or more was observed for several minerals with zinc and phosphorus being most affected. The apparent zinc absorption and retention expressed in absolute values, were significantly higher from the flours of high extraction than from the more refined flours, in spite of a much higher phytate content in whole rye and lightly milled flours. It could be concluded that milling of rye into highly refined flours not only preclude considerable amounts of nutrients from human consumption, but the remaining flours have a much poorer nutritive value than flour made from whole rye. 相似文献

Rye products typically induce low insulin responses and appear to facilitate glucose regulation. The objective of this study was to investigate differences in postprandial glucose, insulin, and satiety responses between breads made from five rye varieties. Breads made from whole grain rye (Amilo, Rekrut, Dankowski Zlote, Nikita, and Haute Loire Pop) or a white wheat bread (WWB) were tested in a randomized cross-over design in 14 healthy subjects (50 g available starch). Metabolic responses were also related to the composition of dietary fiber and bioactive compounds in the breads and to the rate of in vitro starch hydrolysis. The Amilo and Rekrut rye breads induced significantly lower insulin indices (II) than WWB. Low early postprandial glucose and insulin responses (tAUC 0-60 min) were related to higher amounts of caffeic, ferulic, sinapic, and vanillic acids in the rye breads, indicating that the phenolic acids in rye may influence glycemic regulation. All rye breads induced significantly higher subjective feelings of fullness compared to WWB. A low II was related to a higher feeling of fullness and a lower desire to eat in the late postprandial phase (180 min). The data indicate that some rye varieties may be more insulin-saving than others, possibly due to differences in dietary fiber, rate of starch hydrolysis, and bioactive components such as phenolic acids. 相似文献

Teosintes are the closest relatives to modern maize, Zea mays L. ssp. mays. They are wild grasses with a native distribution area from Mexico to Nicaragua and represent an important genetic resource.
However, the genetic diversity of Nicaraguan teosinte (Zea nicaraguensis Iltis et Benz) has not yet been determined. This teosinte species has decreased in the last 25 years and now must be regarded
as an endangered species. An analysis of the genetic diversity of Zea nicaraguensis was carried out in a total of 240 individuals from seven populations. Eleven Simple Sequences Repeat (SSR) primer pairs were
used. A total of 42 alleles were found, the range of alleles per locus was 2–5 (mean 3.8) and the numbers of genotypes varied
between primers. The primer Bnlg 1538 showed the highest value, with 45 genotypes through all populations. The genetic diversity observed (Ho) between all
populations varied from 0.51 to 0.63, with an average of 0.563. One of the populations had as many as 40 alleles. Analysis
of molecular variance (AMOVA) revealed that most of the variation was within population, at a significantly high level (P < 0.001). Rare alleles were detected in all populations, but unique alleles were only found in four populations. These results
are highly relevant when developing conservation strategies and show that preserving populations in their natural habits is

Impact of Urban Stormwater on Sediment Quality in an Enclosed Bay of the Lule River, Northern Sweden

Sediment and porewater samples from an enclosed bay receiving stormwater discharge (Skutviken) near the centre of Lule?, northern Sweden were analysed for major and trace elements and 16 polycyclic aromatic hydrocarbons (PAHs). Among the studied metals Cd, Cu, Pb and Zn were enriched at Skutviken. Also, the PAH content was enriched, in particular for phenantrene, anthracene, fluoranthene and pyrene which are regarded as common constituents in stormwater. The use of trace metal ratios provided indications about pollutant sources for the sediment. Cs-137 dating was used to determine historical changes in metal and PAH fixation in the sediment. The bay Skutviken is enclosed through the construction of a road bank since 1962. The enclosure led to reduced water circulation in the bay that promotes the occurrence of anoxic conditions with sulphate reduction within the bay. As a consequence of these conditions, metals are trapped in the sediments as sulphides. This study suggests that enclosed bays with restricted water circulation may be efficient traps for urban pollutants, reducing the present-day input of pollutants to the sea. In areas with postglacial land uplift, where such bays are common, bay sediments are a potential future source of pollutants when uplift results in erosion and oxidation of the sediments. 相似文献
