  1. Systematic conservation planning in freshwater ecosystems faces multiple challenges because of the dynamic nature of rivers and their multiple dimensions of connectivity. In intermittent hydrological systems connectivity is functional when water is available, allowing the exchange of aquatic individuals between isolated freshwater ecosystems. Integrating these isolated systems in their hydrological context is essential when identifying priority areas for conservation, in order to try to minimize the propagation of threats into target water bodies (management units) from the surrounding landscape.
  2. Here, the use of a systematic planning approach is demonstrated to identify a set of priority management units to preserve freshwater biodiversity in an arid system of fragmented water bodies immersed in a landscape subject to a range of impacts.
  3. Twenty-six water-dependent taxa from 59 mountain rock pools (gueltas) of three southern Mauritanian mountains were used as a case study. A conservation planning tool (marxan ) was used to find priority conservation areas to integrate intermittent hydrological systems in their hydrological context, promote connectivity, and minimize the downstream propagation of threats. Three types of connectivity were analysed: (i) no connectivity, (ii) connectivity between gueltas, and (iii) connectivity between gueltas and sub-catchments.
  4. Considering different types of longitudinal connectivity affects the number and spatial allocation of the priority gueltas selected, and the conservation status of the gueltas and their upstream areas. Incorporating connections between gueltas and upstream locations in the modelling resulted in the selection of gueltas in areas with a low human footprint and in the increased connectivity of the solutions.
  5. The results obtained revealed important locations for local biodiversity conservation, and the method presented can be used when assessing the propagation of potential waterborne threats into isolated management units. The framework developed allows connectivity to be addressed in conservation planning. It can be replicated in regions with similar isolated habitats that connect through intermittent hydrological systems and can also be applied to lateral and vertical hydrological connectivity.

  1. Declines in Panulirus echinatus Smith, 1869 populations along their wide distribution in the Atlantic Ocean have spurred efforts to improve their fisheries management and conservation. In this study, the genetic structure of these populations is reported for the first time.
  2. In a survey of 18 species‐specific polymorphic microsatellite markers, 152 individuals were genotyped from five Atlantic oceanic islands, covering most of the insular distribution range of the species. The analyses revealed that P. echinatus is genetically partitioned into two stocks in the Atlantic Ocean. A highly significant genetic structure was observed between north‐east and south‐west Atlantic populations based on fixation index, discriminant analysis of principal components, and structure and barrier analysis.
  3. We suggest that the Equatorial Circulation System represents a biophysical barrier that effectively limits migration among Atlantic subtropical gyre systems, as has been described for other species. Other physical and ecological barriers, such as the Mid‐Atlantic Ridge itself, the distance between the eastern and western sides of the Atlantic (Mid‐Atlantic Barrier) and water mass differences, as well as other biological aspects, may also influence larval dispersal and modulate the insular distribution of this species.
  4. The results show the existence of two distinct genetic stocks of P. echinatus and have implications for fisheries management in the Atlantic Ocean, including their independent management according to their individual status. The Cabo Verde and the Canarian populations (north‐east Atlantic) showed the lowest level of genetic variability in comparison with the south‐western populations. A combination of factors that have occurred or are occurring in the Canary Islands, such as overfishing and volcanic eruptions, is likely to explain the reduced abundance of this lobster species in the area.

In recent years the consumption of tea and herbal infusions has increased. These hot drinks are consumed as daily drinks as well as for medicinal purposes. All tea varieties (white, yellow, green, oolong, black and puerh) originate from the leaves of the tea plant, Camellia sinensis. All extracts made of plant or herbal materials which do not contain Camellia sinensis are referred as herbal infusions or tisanes. During processing and manufacturing fungal contamination of the plant materials is possible, enabling contamination of these products with mycotoxins. In this study a multimycotoxin UPLC-MS/MS method was developed and validated for the analysis of the raw tea and herbal infusion materials as well as for their drinkable products. The samples were analyzed by ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), with a mobile phase consisting of variable mixtures of water and methanol with 0.3% formic acid. The limits of detection for the different mycotoxins varied between 2.1 μg/kg and 121 μg/kg for raw materials and between 0.4 μg/L and 46 μg/L for drinkable products. Afterward 91 different tea and herbal infusion samples were analyzed. Only in one sample, Ceylon melange, 76 μg/kg fumonisin B(1) was detected. No mycotoxins were detected in the drinkable products.  相似文献

Copper biosorption onto chemically modified biomass of marine alga Sargassum filipendula was investigated in a batch reactor and a fixed bed column. Experiments were carried out in the batch reactor to obtain kinetic and equilibrium data and to assess the copper desorption efficiency of different eluent solutions. The pseudo-first-order, pseudo-second-order, and Langmuir kinetic models were used to correlate kinetic data. The experimental data fitted well to the pseudo first order and Langmuir kinetic models. Langmuir and Freundlich models were applied to describe the equilibrium data obtained at a fixed temperature of 30°C and at pH values of 3.0, 4.0, 5.0, and 6.0. The maximum capacities of copper biosorption onto the algal biomass were 1.43, 1.59, 2.40, and 2.36 mequiv./g at pH 3.0, 4.0, 5.0, and 6.0, respectively. The efficiencies of two eluent solutions (calcium chloride and hydrochloric acid) for copper removal from the biomass were evaluated at different concentrations (0.1, 0.2, 0.5, and 1.0 mol/L). The efficiencies of the calcium chloride solutions varied from 1% to 14%, while efficiencies varying from 95% to 99% were obtained when hydrochloric acid solutions were applied. Three adsorption/desorption cycles were carried out in a fixed bed column using 0.1 mol/L hydrochloric acid as eluent solution. The results showed that an increase in the number of cycles led to a reduction in the adsorption capacity of the alga. The desorbed copper fraction presented no significant variation, remaining around 63% in the three adsorption/desorption cycles.  相似文献

The general objective of this study was to evaluate the ordered weighted averaging (OWA) method, integrated to a geographic information systems (GIS), in the definition of priority areas for forest conservation in a Brazilian river basin, aiming at to increase the regional biodiversity. We demonstrated how one could obtain a range of alternatives by applying OWA, including the one obtained by the weighted linear combination method and, also the use of the analytic hierarchy process (AHP) to structure the decision problem and to assign the importance to each criterion. The criteria considered important to this study were: proximity to forest patches; proximity among forest patches with larger core area; proximity to surface water; distance from roads; distance from urban areas; and vulnerability to erosion. OWA requires two sets of criteria weights: the weights of relative criterion importance and the order weights. Thus, Participatory Technique was used to define the criteria set and the criterion importance (based in AHP). In order to obtain the second set of weights we considered the influence of each criterion, as well as the importance of each one, on this decision-making process. The sensitivity analysis indicated coherence among the criterion importance weights, the order weights, and the solution. According to this analysis, only the proximity to surface water criterion is not important to identify priority areas for forest conservation. Finally, we can highlight that the OWA method is flexible, easy to be implemented and, mainly, it facilitates a better understanding of the alternative land-use suitability patterns.  相似文献

OBJECTIVE: To assess the efficacy of prophylactic hoof health examination and trimming during midlactation at reducing the incidence of lameness during late lactation in dairy cows. DESIGN: Randomized field trial. ANIMALS: 333 Holstein cows. PROCEDURES: Cows without apparent lameness were randomly allocated into 1 of 2 groups approximately 204 days after calving. Cows allocated to the treatment group (n = 161) were examined on a tilt table for diagnosis and underwent hoof-trimming procedures, if needed, for treatment of hoof disorders or lesions. Cows in the control group (n = 172) were not examined. Cows were assigned a locomotion score weekly for 28 weeks after allocation to a group. The number of cows classified as lame during late lactation (approx 205 to 400 days after calving) was compared between groups to assess the efficacy of prophylactic examination and trimming. RESULTS: Incidence of lameness during late lactation was 24% in cows in the control group and 18% in cows in the treatment group. CONCLUSIONS AND CLINICAL RELEVANCE: The 25% decrease in number of new cases of lameness in cows undergoing prophylactic hoof health examination and trimming during midlactation may be relevant for the well-being of dairy cows and should not represent a major economic burden to producers.  相似文献

Erythrocytes of bovines and other ruminants have a strikingly anomalous phospholipid composition, with low or absent phosphatidylcholine (PC) together with high sphingomyelin (SM) content. Here, we report the presence in normal bovine serum of high levels of anti-phospholipid antibodies of IgM isotype against, PC and the phosphono analogue of phosphatidylethanolamine, aminoethylphosphonolipid (AEPL), normally produced by rumen ciliates. In contrast, no antibodies were detected against SM or N-acyl-phosphatidylethanolamine (NAPE), the major components of bovine erythrocytes. In addition, we found that exposure of the ciliate Tetrahymena thermophila to bovine serum results in rapid lysis, an effect that was inhibited by adsorption of the serum with SM/AEPL liposomes. Furthermore, incubation with bovine serum had a similar effect on freshly obtained ruminal ciliates, and the lytic activity was eliminated by pre-adsorption of the serum with SM/PE liposomes. The ruminant mode of life with its concomitant ciliate fauna is hereby linked to the peculiar conformation of bovine erythrocyte membranes. We propose that the unique phospholipid composition of bovine erythrocytes appears as an evolutionary adaptation to tolerate the lytic effects of anti-phospholipid antibodies generated against AEPL, a membrane component of the huge mass of ruminal ciliates, necessary commensals of this group of mammals.  相似文献

Soil microbial populations and their functions related to nutrient cycling contribute substantially to the regulation of soil fertility and the sustainability of agroecosystems. A field experiment was performed to assess the medium‐term effect of a mineral fertilizer and two organic fertilization systems with different nitrogen sources on the soil microbial community biomass, structure, and composition (phospholipid fatty acids, pattern, and abundance), microbial activity (basal respiration, dehydrogenase, protease, urease, β‐glucosidase, and total amount of phosphomonoesterase activities), and physical (aggregate stability) and chemical (total organic C, total N, available P and water‐soluble carbohydrates) properties in a vineyard under semiarid Mediterranean conditions after a period of 10 years. The three fertilization systems assayed were as follows: inorganic fertilization, addition of grapevine pruning with sheep manure (OPM), and addition of grapevine pruning with a legume cover crop (OPL). Both treatments, OPM and OPL, produced higher contents of total organic carbon, total N, available P, water‐soluble carbohydrates, and stable aggregates. The organic fertilization systems increased microbial biomass, shifted the structure and composition of the soil microbial community, and stimulated microbial activity, when compared with inorganic fertilization. The abundances of fungi and G+ bacteria were increased by treatments OPM and OPL, without significant differences between them. Organic and inorganic fertilization produced similar grapevine yields. The ability of the organic fertilization systems for promoting the sustainability and soil biological and chemical fertility of an agroecosystem under semiarid conditions was dependent of the organic N source. Banana and plantain are among the most important food crops in developing countries but production is threatened by increasing virulent forms of Fusarium oxysporum f. sp. cubense. Chemical control is not economically effective and,therefore, breeding programs are necessary. Traditional field studies of new genotype resistance to this disease are time-consuming and destructive. Therefore,we developed a rapid and non-destructive procedure to differentiate field-grown banana resistant from susceptible clones. This procedure implicates application of culture filtrates of Fusarium oxysporum f. sp. cubense race 1 onto banana leaves. The relationship between duration of the fungal in vitro incubation, and the fungal culture fresh mass, the culture filtrate absorbency, and the Gross Michel (susceptible cultivar)leaf lesion area (after application of the culture filtrate) were similar and at 24day-incubation the highest values of the recorded indicators were observed. A comparison between Gross Michel and FHIA-01(resistant) was also performed. The most relevant differences between cultivars were observed at 48 hours after application of the culture filtrate, and in the middle-aged leaves. The position of the culture filtrate application in the leaf limb (distal, middle, proximal) was not determinant. A wider comparison among banana cultivars confirmed previous results informed by other researchers using different systems to study this plant-fungus interaction. Such a confirmation validates the effectiveness of the procedure described here to select rapid and non-destructively banana resistance to this disease at field level.
