Heparan and chondroitin sulfate proteoglycans (HSPGs and CSPGs, respectively) regulate numerous cell surface signaling events, with typically opposite effects on cell function. CSPGs inhibit nerve regeneration through receptor protein tyrosine phosphatase sigma (RPTPσ). Here we report that RPTPσ acts bimodally in sensory neuron extension, mediating CSPG inhibition and HSPG growth promotion. Crystallographic analyses of a shared HSPG-CSPG binding site reveal a conformational plasticity that can accommodate diverse glycosaminoglycans with comparable affinities. Heparan sulfate and analogs induced RPTPσ ectodomain oligomerization in solution, which was inhibited by chondroitin sulfate. RPTPσ and HSPGs colocalize in puncta on sensory neurons in culture, whereas CSPGs occupy the extracellular matrix. These results lead to a model where proteoglycans can exert opposing effects on neuronal extension by competing to control the oligomerization of a common receptor.  相似文献

A nanoencapsulation technique was applied to an oxygen-scavenging system, and thermal processing was investigated as an activator to trigger the oxygen-scavenging reaction. α-Tocopherol-loaded polycaprolactone (PCL) nanoparticles (NPs) were prepared using an oil-in-water emulsion solvent evaporation method. The influences of iron(II) chloride, water, and thermal processing on the oxygen-scavenging capability were investigated. NPs without iron(II) chloride, moisture, and thermal processing had no oxygen-scavenging effect. However, the oxygen content (%) in the cup headspace of 20.9% decreased to 20.4% when the oxygen-scavenging system contained NPs, water, and iron(II) chloride. The oxygen content (%) decreased further to 19.5% when water was eliminated from the mixture. In this research, NPs and iron(II) chloride with thermal processing had an oxygen-scavenging capacity of 6.44 cm3 of O2/g and an oxygen-scavenging rate of 0.21 cm3 of O2 g(-1) day(-1). Results indicated that NPs and iron(II) chloride in an oxygen-scavenging system can be used as a heat-activated oxygen scavenger.  相似文献

Dog ownership is considered a risk factor for campylobacteriosis in humans. This study investigated the prevalence and shedding of Campylobacter spp. in kennelled dogs. Faecal samples (n=399) were collected in longitudinal studies from 52 dogs in two kennels. Campylobacter spp. were isolated using charcoal-based selective agars and direct PCR. The prevalence of Campylobacter spp. in dogs in boarding kennels ranged from 46% (95% CI 22, 72) on entry, to 50% (95% CI 30, 70) overall, and in dogs in 'rescue' kennels from 68% (95% CI 49, 84) on entry to 73% (95%, CI 56, 87) overall. C. upsaliensis was isolated from 62% (95% CI 48, 73) of the dogs, whilst C. jejuni was isolated from 15% (95% CI 7, 26) of animals. The majority of infected dogs entered the kennels already carrying Campylobacter spp., and remained infected throughout their stay. However, in some cases, shedding appeared to commence after kennelling. Given that the prevalence of C. upsaliensis and C. jejuni was relatively high in dogs from both boarding and rescue kennels, such animals may pose a zoonotic risk.  相似文献

BACKGROUND: Drip application of insecticides is an effective way to deliver the chemical to the plant that avoids off‐site movement via spray drift and minimizes applicator exposure. The aim of this paper is to present a cascade model for the uptake of pesticide into plants following drip irrigation, its application for a soil‐applied insecticide and a sensitivity analysis of the model parameters. RESULTS: The model predicted the measured increase and decline of residues following two soil applications of an insecticide to peppers, with an absolute error between model and measurement ranging from 0.002 to 0.034 mg kg fw?1. Maximum measured concentrations in pepper fruit were approximately 0.22 mg kg fw?1. Temperature was the most sensitive component for predicting the peak and final concentration in pepper fruit, through its influence on soil and plant degradation rates. CONCLUSION: Repeated simulations of pulse inputs with the cascade model adequately describe soil pesticide applications to an actual cropped system and reasonably mimic it. The model has the potential to be used for the optimization of practical features, such as application rates and waiting times between applications and before harvest, through the integrated accounting of soil, plant and environmental influences. Autofluorescence of salmon and cod muscle was measured and compared with autofluorescence of collagen type I and type V. Similarities between fluorescence of fish muscle and collagen were found in that the same peaks were obtained around 390, 430, and 480 nm. These similarities are supported by principal component analyses. Texture and gaping score were predicted from the fluorescence spectra by partial least-squares regression. However, the predictions did not perform well. Relating fluorescence to the gaping score gave a prediction error of 0.91 and a correlation of 0.43 when measuring gaping on a scale from 0 to 5. There was no relation between texture and fluorescence spectra. Fluorescence of fish muscle could be related to the storage time. However, this relation seemed not to be induced by changes in collagen.  相似文献

We studied the effects of the root endoparasitic nematode Heterodera trifolii on rhizodeposition and the root architecture of white clover (Trifolium repens). Rhizosphere solutions were collected from the root systems of plants growing with and without H. trifolii (200 juveniles per inoculated plant) in sand-based microlysimeters. The organic carbon (C) content of these solutions was analyzed, and they were applied to plant-free soils to investigate microbial responses. Although plant biomass was unaffected by nematodes, the architecture of the root systems was significantly altered, with a decrease in overall root length and an increase in the density of lateral branches from the primary root. The presence of nematodes reduced the concentration of organic compounds in the rhizosphere solutions but only on the final sampling date (75 days). Analysis of microbial signature phospholipid fatty acids revealed no change in the structure of the microbial communities in soils to which rhizosphere solutions were applied. However, these microorganisms did respond with changes in substrate utilization patterns (community-level physiological profiles). Microbes in soils that received rhizosphere solutions from the nematode-infected clover showed lower utilization of most substrates but higher utilization of oligosugars. These responses appear to be related to changes in roots and rhizodeposition associated with nematode infection of clover roots. The results of this study suggest that root herbivory can negatively impact carbon-limited soil microbial communities via changes in root architecture that moderate rhizodeposition.  相似文献

The overall objective of this research was to find a new way to valorize rye bran, by producing a gellifier from the enzymatic solubilization of arabinoxylans (AX). The effects of three pure endo-xylanases from Aspergillus niger (Xyl-1), Talaromyces emersonii (Xyl-2), and Bacillus subtilis (Xyl-3) and of Grindamyl S100 (GS100), a commercial enzyme preparation containing a Xyl-1 type endo-xylanase, were tested on rye bran to study the solubilization of water-unextractable arabinoxylans (WUAX). Eight different extrusion-treated rye brans were also used as substrates to find the best physical treatment to facilitate enzymatic arabinoxylan (AX) solubilization. Arabinoxylans were better solubilized from the bran extruded at high temperature using Xyl-3. This enzyme was then tested in combination with pure (1,4)-beta-d-arabinoxylan arabinofuranohydrolase (AXH) and endo-beta-d-glucanase or ferulic acid esterase (FAE), from A. niger. Only beta-glucanase in combination with Xyl-3 improved the AX extraction, but it did not have a marked effect on the viscosity of the extracts. Xyl-3 was then tested on a high-shear-treated rye bran, and results were compared to those obtained with the high-temperature-extruded rye bran. The high-shear treatment did not improve the bran AX enzymatic solubilization. The combination of FAE with Xyl-1 or Xyl-3 did not improve the AX extraction from untreated and high-shear-treated rye bran. Finally, to study the gelation capacity of the enzymatically solubilized AX, the effect of the hydrogen peroxide/horseradish peroxidase (H(2)O(2)/POD) was tested on the Xyl-3 high-temperature-extruded bran extracts. Solubilized AX did not gel in the presence of the oxidizing system.  相似文献

Streptomycete spores germinated most successfully when placed amongst hyphae of fungi that they could subsequently antagonize. The hyphae of many streptomycetes from Picea litter were able to antagonize fungi from the same habitat, but they were less effective against a group of alien fungi. Streptomycete hyphae were shown to be able to grow at the expense of antagonized fungal hyphae which suggested that they were gaining nutrients from the interaction. The fungal-streptomycete interactions were similar to interfungal hyphal interference reactions. No instances of hyphal penetration were recorded. Many bacteria from the H layer were able to lyse dead Streptomycete hyphae and they were shown to be capable of using both dead and living Streptomycete hyphae as a sole C source. Bacteria from the L and F litter layers were less active in these respects.  相似文献

The influence of the addition of metal chelators on oxidative stability was studied in a milk drink and in a mayonnaise system containing highly polyunsaturated lipids. Milk drinks containing 5% (w/w) of specific structured lipid were supplemented with lactoferrin (6-24 muM) and stored at 2 degrees C for up to 9 weeks. Mayonnaise samples with 16% fish oil and 64% rapeseed oil (w/w) were supplemented with either lactoferrin (8-32 muM), phytic acid (16-124 muM), or EDTA (16-64 muM) and were stored at 20 degrees C for up to 4 weeks. The effect of the metal chelators was evaluated by determination of peroxide values, secondary volatile oxidation products, and sensory analysis. Lactoferrin reduced the oxidation when added in concentrations of 12 muM in the milk drink and 8 muM in the mayonnaise, whereas it was a prooxidant at higher concentrations in both systems. In mayonnaise, EDTA was an effective metal chelator even at 16 muM, whereas phytic acid did not exert a distinct protective effect against oxidation. The differences in the equimolar effects of the metal chelators are proposed to be due to differences in their binding constants to iron and their different stabilities toward heat and low pH.  相似文献

Nitric acid (HNO3) vapor is a significant component of air pollution. Dry deposition of HNO3 is thought to be a major contributor to terrestrial loading of anthropogenically-derived nitrogen (N), but many questions remain regarding the physico-chemical process of deposition and the biological responses to accumulation of dry-deposited HNO3 on surfaces. To examine these processes experimentally, a continuously stirred tank reactor (CSTR) fumigation system has been constructed. This system enables simultaneous fumigation at several concentrations in working volumes 1.3 m dia by 1.3 m ht, allowing for simultaneous fumigation of many experimental units. Evaluation of the system indicates that it is appropriate for long-term exposures of several months duration and capable of mimicking patterns of diurnal atmospheric HNO3 concentrations representative of areas with different levels of pollution.  相似文献
