Background

The process for obtaining monoclonal antibodies against a specific antigen is very laborious, involves sophisticated technologies and it is not available in most research laboratories. Considering that most cytokines remain partially conserved among species during evolution, the search for antibody cross-reactivity is an important strategy for immunological studies in veterinary medicine. In this context, the amino acid sequence from human and canine cytokines have demonstrated 49–96 % homology, suggesting high probability of cross-reactivity amongst monoclonal antibodies. For this, 17 commercially available anti-human monoclonal antibodies [IL-1α, IL-1β, IL-2, IL-4, IL-5, IL-6, IL-8 (#1, #2), IL-10, IL-12, IL-13, IL-17A, IFN-γ (#1, #2), TNF-α (#1, #2) and TGF-β], were evaluated in vitro for intracellular cytokine detection in a stimulated canine blood culture by flow cytometry and confocal microscopy. Lymphocytes from peripheral blood of healthy and two unhealthy dogs were analyzed.

Results

Eleven anti-human mAbs [IL-1α, IL-4, IL-5, IL-6, IL-8 (#1, #2), IL-12, IL-17A, TNF-α (#1, #2) and TGF-β] cross-reacted against canine intracellular cytokines. The specificity of the assays was not affected after Fc-blocking. Three anti-human cytokine mAbs [IL-4, IL-8 (#2) and TGF-β] when evaluated by confocal microscopy also cross-reacted with intracellular canine cytokines. The identification of human mAbs that cross-reacted with canine cytokines may support their use as immunological biomarkers in veterinary medicine studies.

Conclusion

The identification of these 11 anti-human cytokine mAbs that cross-reacted with canine cytokines will be useful immunological biomarkers for pathological conditions by flow cytometry and fluorescence microscopy in dogs.  相似文献

This paper examines return migrants and new migrants to Montana: Who are they? Why do they move? Do return migrants move for different reasons than new migrants? Data from the 1994–1997 Montana Poll, a representative survey of Montana households, are used. A comparison of socio‐economic differences of return and new migrants shows that the two migrant types are very similar in terms of education, income, and age. This stands in contrast to the findings of others who maintain that return migrants are negatively selected with respect to education. Logistic regressions were employed to identify the effect of age and place ties on reasons for moving. Return migrants and new migrants move to Montana for very similar reasons, with family being the most important primary reason for moving. Moving for lifestyle reasons, such as environmental quality and urban amenities, were found to systematically change with age. This could explain why people return to a place they left earlier in life. While other research on return migration compared return migrants and other migrants who left the same place of origin, this paper offers a comparison of return migrants and other migrants who seek out the same destination. Results from the Montana Poll suggest that the same destination attracts return migrants and new migrants with similar socio‐economic characteristics who move there for very similar reasons.  相似文献

Embryogenic calli derived from anther cultures of the two-rowed winter barley cultivar Igri were plated on solid L3 medium containing the proline analogue hydroxyproline (Hyp), 10–20 mmol l–1. Exposure to Hyp caused severe degeneration of most of the calli. Hyp resistant calli, distinguishable by their lighter colour and higher growth rate, and control calli not exposed to Hyp were plated on L3 regeneration medium. From 22,500 anthers exposed to Hyp 46 Hyp resistant regenerates were obtained, which were transferred to soil. After cultivation for 5–10 weeks at normal growth conditions they were cold hardened at 2 C under short day conditions together with control regenerates. Frost tolerance assays with segments of fully grown leaves of unhardened and cold hardened plants revealed that Hyp resistant regenerants were significantly more frost tolerant than the control regenerants. Improved frost tolerance was found also in the progenies R1 to R9, and genotypic segregation in the R1 generation in a 1:2:1 ratio was indicated. Increased proline content was observed in the R2 generation and in subsequent generations and was significantly (P 0.001) correlated with increased frost tolerance in the Hyp lines. Comparative studies of R9 progenies from homozygous R2 plants with the wild type Igri under field conditions in winter at three locations in Europe as well as crossing experiments confirmed the heritable improvement of frost tolerance and winter survival, respectively, in the Hyp lines. The results support the hypothesis that proline accumulation in cold acclimated winter barley plants is causally related to the acquisition of frost tolerance. Moreover, the described biotechnological procedure may be applicable in breeding programs for improved winter hardiness and possibly also for other stress tolerances.  相似文献

The tepary bean (Phaseolus acutifolius) is the object of renewed interest because it possesses some interesting agronomic attributes such as tolerance to drought and salinity. In order to gain a better understanding of the organisation of its genetic diversity, we have examined patterns of diversity for nine polymorphic enzyme systems representing 12 loci, in a sample of 55 wild and 8 cultivated accessions. Several geographic patterns were identified, including an East-West differentiation, (across the Sierra Madre Occidental mountains), a North-South differentiation, and a localized differentiation. Wild teparies displayed a higher number of polymorphic loci, as well as a higher number of alleles per polymorphic locus compared to cultivars, consistent with previous observations indicating a reduction of phaseolin diversity on domestication. No clearcut separation with respect to isozyme differentiation was observed between P. acutifolius var. tenuifolius and var. acutifolius, further questioning the validity of this taxonomic separation based on leaflet shape.  相似文献

A major challenge in sustainable crop management is to ensure adequate P supply for crops, while minimizing losses of P that could negatively impact water quality. The objective of the present study was to investigate the effects of long‐term applications of different levels of mineral fertilizers and farmyard manure on (1) the availability of P, (2) the relationship between soil C, N, and P, and (3) the distribution of inorganic and organic P in size fractions obtained by wet sieving. Soil samples were taken from the top 20 cm of a long‐term (29 y) fertilization trial on a sandy Cambisol near Darmstadt, SW Germany. Plant‐available P, determined with the CAL method, was little affected by fertilization treatment (p < 0.05) and was low to optimal. The concentration of inorganic and organic P extracted with a NaOH‐EDTA solution (PNaOH‐EDTA) averaged about 350 mg (kg dry soil)–1, with 42% being in the organic form (Po). Manure application tended to increase soil C, N, and Po concentrations by 8%, 9%, and 5.6%, respectively. Across all treatments, the C : N : Po ratio was 100 : 9.5 : 2 and was not significantly affected by the fertilization treatments. Aggregate formation was weak due to the low clay and organic‐matter content of the soil, and the fractions > 53 μm consisted predominantly of sand grains. The different fertilization treatments had little effect on the distribution of size fractions and their C, N, and P contents. In the fractions > 53 μm, PNaOH‐EDTA ranged between 200 and 300 mg kg–1, while it reached 1260 mg kg–1 in the fraction < 53 μm. Less than one third of PNaOH‐EDTA was present as Po in the fractions > 53 μm, while Po accounted for 70% of PNaOH‐EDTA in the smallest fraction (< 53 μm). Therefore, 16% and 28% of PNaOH‐EDTA and Po, respectively, were associated with the smallest fraction, even though this fraction accounted for < 5% of the soil mass. Therefore, runoff may cause higher P losses than the soil P content suggests in this sandy soil with a weak aggregate formation. Overall, the results indicate that manure and mineral fertilizer had similar effects on soil P fractions.  相似文献

ABSTRACT: A mysterious disease affecting calves, named bovine neonatal pancytopenia (BNP), emerged in 2007 in several European countries. Epidemiological studies revealed a connection between BNP and vaccination with an inactivated vaccine against bovine virus diarrhea (BVD). Alloantibodies reacting with blood leukocytes of calves were detected in serum and colostrum of dams, which have given birth to calves affected by BNP. To understand the linkage between vaccination and the development of alloantibodies, we determined the antigens reacting with these alloantibodies. Immunoprecipitation of surface proteins from bovine leukocytes and kidney cells using sera from dams with a confirmed case of BNP in their gestation history reacted with two dominant protein species of 44 and 12 kDa. These proteins were not detected by sera from dams, free of BVDV and not vaccinated against BVD, and from sera of animals vaccinated with a different inactivated BVD vaccine. The 44 kDa protein was identified by mass spectrometry analysis as MHC I, the other as β-2-microglobulin. The presence of major histocompatibility complex class I (MHC I) in the vaccine was confirmed by Western blot using a MHC I specific monoclonal antibody. A model of BNP pathogenesis is proposed.  相似文献

Because of few available data on semen parameters in small breed dogs, the study aimed to analyse semen and measure testes of dogs ≤10.0 kg bodyweight. Semen was collected from 41 fertile stud dogs, which were divided based on bodyweight: group 1 ≤ 5.0 kg and group 2 between 5.1 and 10.0 kg. Median values for ejaculate volume (group 1: 1.2 ml; group 2: 2.2 ml), total sperm output (group 1: 110.7 × 106; group 2: 215.1 × 106) and testicular volume (group 1: left testicle 2.8 ml, right testicle 2.7 ml; group 2: left testicle 5.5 ml, right testicle 5.0 ml) were lower in group 1 compared to dogs of group 2 (p = .001; p = .001; both testes: p < .001). There was no difference in sperm concentration (p = .985). Based on these results, introduction of an additional weight group to the commonly used reference values is recommended, since values for ejaculate volume, total sperm output and testicular dimensions for dogs ≤5.0 kg bodyweight differed significantly from values of dogs with a bodyweight from 5.1 to 10.0 kg.  相似文献
