Low impedance at the interface between tissue and conducting electrodes is of utmost importance for the electrical recording or stimulation of heart and brain tissue. A common way to improve the cell-metal interface and thus the signal-to-noise ratio of recordings, as well as the charge transfer for stimulation applications, is to increase the electrochemically active electrode surface area. In this paper, we propose a method to decrease the impedance of microelectrodes by the introduction of carbon nanotubes (CNTs), offering an extremely rough surface. In a multistage process, an array of multiple microelectrodes covered with high quality, tightly bound CNTs was realized. It is shown by impedance spectroscopy and cardiac myocyte recordings that the transducer properties of the carbon nanotube electrodes are superior to conventional gold and titanium nitride electrodes. These findings will be favorable for any kind of implantable heart electrodes and electrophysiology in cardiac myocyte cultures. 相似文献

Cytotoxic Effects of Tropodithietic Acid on Mammalian Clonal Cell Lines of Neuronal and Glial Origin

The marine metabolite tropodithietic acid (TDA), produced by several Roseobacter clade bacteria, is known for its broad antimicrobial activity. TDA is of interest not only as a probiotic in aquaculture, but also because it might be of use as an antibacterial agent in non-marine or non-aquatic environments, and thus the potentially cytotoxic influences on eukaryotic cells need to be evaluated. The present study was undertaken to investigate its effects on cells of the mammalian nervous system, i.e., neuronal N2a cells and OLN-93 cells as model systems for nerve cells and glia. The data show that in both cell lines TDA exerted morphological changes and cytotoxic effects at a concentration of 0.3–0.5 µg/mL (1.4–2.4 µM). Furthermore, TDA caused a breakdown of the mitochondrial membrane potential, the activation of extracellular signal-regulated kinases ERK1/2, and the induction of the small heat shock protein HSP32/HO-1, which is considered as a sensor of oxidative stress. The cytotoxic effects were accompanied by an increase in intracellular Ca2+-levels, the disturbance of the microtubule network, and the reorganization of the microfilament system. Hence, mammalian cells are a sensitive target for the action of TDA and react by the activation of a stress response resulting in cell death. 相似文献

Objective Characterization of a benign disease syndrome involving the eyelids of dogs, describing the signalment, clinical appearance, anatomic location, and pathologic features. Animal studied The records and submitted tissue of 10 dogs with mesenchymal hamartomatous lesions of the periocular connective tissues were retrieved from the Comparative Ocular Pathology Laboratory of Wisconsin (COPLOW) database. Procedure The submitted tissue in each case was stained with hematoxilyn & eosin and Masson’s Trichrome stain and examined histopathologically. Clinical information was collected from the submission requests. The clinical history, treatment, and follow‐up are described in more detail for one of the 10 dogs. Results Seven different breeds, including four Golden Retrievers, were represented. The ages of affected dogs ranged from 6 to 11 years. Eight of 10 lesions were located at the temporal canthus, ranging in diameter from 0.6 to 3 cm. Clinically, the masses were subcutaneous, firm, lobular soft‐tissue growths, which were in some cases adherent to the underlying orbital rim, and in others, freely palpable between the skin and conjunctiva of the eyelid. Histologically, all had distinct margins but were not encapsulated and contained normal appearing collagen‐rich connective tissue with some adipose tissue. Five also contained fully differentiated skeletal muscle tissue arranged in poorly formed aggregates or as individual muscle fibers. Conclusion Mesenchymal hamartoma of the eyelid has not been previously described. The mass has a predisposition to occur at the temporal canthus and should be included in the list of differential diagnoses of benign eyelid masses in dogs. 相似文献

The potential impact of transgenic crops on community ecology will depend on the distribution and establishment of the new transgenic traits, on the sexual transfer of their new genes to the environment (Bartsch &; Pohl-Orf, 1996) and on the potential ecological impact of the transgenic trait. Flowering and pollen dispersal is important for outcrossing of the genetically engineered trait. For a biennial plant, like the cultivars of Beta vulgaris L., overwintering is normally necessary to become generative and to produce pollen and seeds (Abe et al., 1997), which usually does not happen with sugar beet as a field crop harvested in autumn (Longden 1989). The starting point for the project was a transgenic sugar beet, Beta vulgaris L. subsp. vulgaris (Lange et al., 1998), with rhizomania and herbicide ( Basta®, Liberty®) resistance. Cold tolerance is one of the most important factors for survival of sugar beet in Central- and North-Europe. Among other ways, spreading of transgenic traits into weed beet (Boudry et al., 1993) or wild beet can occur if genetically engineered – biennial – plants survive the winter, flower in spring and spread their pollen. Field experiments were performed with transgenic breeding lines and their hybrids, transgenic and non-transgenic hybrids with Swiss chard and three conventional beet cultivars to evaluate winter survival rates at seven different field sites. We could show that survival of sugar beet – transgenic as well as conventional ones – in Germany and at the Dutch border is possible. Survival rates were well correlated with temperature data and were unexpectedly high. Differences between sugar beet hybrids and breeding lines could be detected but not within different breeding lines or hybrids. There were no differences detectable between transgenic and non-transgenic plants. The data are crucial for the risk assessment of the release of transgenic sugar beet and are the basis for further experiments towards outcrossing and establishment. 相似文献

This paper examines return migrants and new migrants to Montana: Who are they? Why do they move? Do return migrants move for different reasons than new migrants? Data from the 1994–1997 Montana Poll, a representative survey of Montana households, are used. A comparison of socio‐economic differences of return and new migrants shows that the two migrant types are very similar in terms of education, income, and age. This stands in contrast to the findings of others who maintain that return migrants are negatively selected with respect to education. Logistic regressions were employed to identify the effect of age and place ties on reasons for moving. Return migrants and new migrants move to Montana for very similar reasons, with family being the most important primary reason for moving. Moving for lifestyle reasons, such as environmental quality and urban amenities, were found to systematically change with age. This could explain why people return to a place they left earlier in life. While other research on return migration compared return migrants and other migrants who left the same place of origin, this paper offers a comparison of return migrants and other migrants who seek out the same destination. Results from the Montana Poll suggest that the same destination attracts return migrants and new migrants with similar socio‐economic characteristics who move there for very similar reasons. 相似文献

Canine leishmaniosis is a common parasitic disease in Central Europe affecting dogs imported or returning from endemic countries around the Mediterranean basin. Through an internet discussion forum owners of dogs with suspected or proven leishmaniosis living in Central Europe (D, A, CH), were questioned about the dog's history. Additionally, serologic examinations of the dogs for anti-Leishmania antibodies (ELISA using antigen of promastigote stages) were offered to the participants. From February to October 2003, 291 dogs imported or returning from Southern Europe (Spain, Italy, Greece, Turkey, France, Malta, Portugal and others) were analysed. Serologically, 111 dogs (38%) were classified positive; 103 being imported and eight travelling dogs. The majority of seropositive dogs originated from Spain (67%). No significant correlation could be established between race, sex and age and the incidence of the disease. The clinical symptoms in the seropositive dogs varied widely and ranged from mild general symptoms to visceral manifestations with chronic renal failure. Skin disorders were found in 78% of the seropositive, symptomatic animals. In the country of origin or after import or return, 174 out of 291 dogs (60%) had been tested for the presence of anti-Leishmania antibodies by different immunofluorescence antibody tests (IFAT). Discrepancies between the ELISA and the various IFATs used were noted in 55 cases (32%), especially in cases of low IFAT titers. Most of the seropositive dogs (80%) had been treated against leishmaniosis. In 91% of these cases, Allopurinol as monotherapy or in combination with Glucantime had been used. An epidemiological study was conducted to determine the impact of maedi-visna (MV) seropositivity on productivity in commercial sheep flocks of the province of Quebec, Canada. A total of 1734 ewes and 220 rams were selected randomly from 29 flocks distributed in the Bas-St-Laurent and Estrie regions. Serostatus was determined with an enzyme-linked immunosorbent assay (ELISA) using recombinant proteins.Flock-specific, animal-level seroprevalence varied from 3 to 70% (median=29%). Seroprevalence increased with age and size of the flock, and was higher in ewes relative to rams (but was not associated with body score). A decrease of 0.94 kg per lamb in weaning weight was seen only for lambs raised by seropositive ewes >/=4 years old, and seropositivity in ewes of any age was associated with an increase in 0-30 days lamb-mortality (OR: 1.65). The impact of MV infection on weaning weight and lamb mortality did not vary between flocks, and seropositivity in ewes was not associated with litter size or lamb's birth weight. 相似文献

The identification of lactic acid bacteria in a complex microbiota using bacteriological culture in combination with phenotypic and genotypic identification techniques is laborious and time-consuming. New molecular methods permit a fast and culture-independent characterisation of such microbiota. Denaturing gradient gel electrophoresis (DGGE) of PCR fragments of the 16S rRNA gene has been proven to be a suitable tool. Here the use of PCR-DGGE with group specific primers is described to investigate the dynamic of sourdough microbiota from addition of the starter until the microbiota remained stable. Species were identified by applying an identification ladder obtained from reference strains or by sequence analysis of the PCR fragments. Furthermore, a method for detection of strains in complex microbiota is described. A strain specific chromosomal DNA fragment of Lactobacillus paracasei LTH 2579 was isolated applying the subtraction hybridisation. Based on the acquired target sequence a specific PCR system was established and combined with a PCR system specific for the species L. paracasei. Use of this detection system permitted to identify and quantitatively detect L. paracasei LTH 2579 in fermented sausages and upon consumption in faecal samples. 相似文献
