In the present work, we studied the acute phase protein response after experimental virus infection in pigs. The animals were experimentally infected with African Swine Fever (ASF) or Aujeszky's disease (AD) viruses. The clinical course of ASF infection correlated with increasingly high levels of pig Major Acute-phase Protein (pig-MAP) (mean value of 6 mg/mL on day 6 post infection (p.i.), from 6 to 9 times higher than day 0) and sharp apolipoprotein A-I (apo A-I) decrease (mean value of 0.5 mg/mL, from 4 to 10 times lower than day 0 on day 4 p.i.). AD-clinical signs appeared at day 3 p.i., both in vaccinated (moderate clinical signs) and non-vaccinated pigs (severe outcome within 48 h p.i.). Pig-MAP and apo A-I profiles also followed clinical signs (changing from 0.70 mg/mL to around 3 mg/mL and from around 3 mg/mL to 0.96 mg/mL, respectively in non-vaccinated animals), with minor changes in concentration in the vaccinated group. Haptoglobin levels significantly increased in ASF and AD infected animals (mean maximum values of 2.77 and 3.96 mg/mL, respectively). Minor differences for the C-Reactive Protein in the case of ASF were observed, whereas its concentration increased more than 7 times in AD-infection. The albumin level was not modified in either case. The correlation of clinical signs to our data suggests the potential use of pig-MAP and apo A-I in monitoring infections in swine.  相似文献

The external microbiome of fish is thought to benefit the host by hindering the invasion of opportunistic pathogens and/or stimulating the immune system. Disruption of those microbial communities could increase susceptibility to diseases. Traditional aquaculture practices include the use of potent surface-acting disinfectants such as potassium permanganate (PP, KMnO4) to treat external infections. This study evaluated the effect of PP on the external microbiome of channel catfish and investigated if dysbiosis leads to an increase in disease susceptibility. Columnaris disease, caused by Flavobacterium columnare, was used as disease model. Four treatments were compared in the study: (I) negative control (not treated with PP nor challenged with F. columnare), (II) treated but not challenged, (III) not treated but challenged, and (IV) treated and challenged. Ribosomal intergenic spacer analysis (RISA) and pyrosequencing were used to analyze changes in the external microbiome during the experiment. Exposure to PP significantly disturbed the external microbiomes and increased catfish mortality following the experimental challenge. Analysis of similarities of RISA profiles showed statistically significant changes in the skin and gill microbiomes based on treatment and sampling time. Characterization of the microbiomes using 16S rRNA gene pyrosequencing confirmed the disruption of the skin microbiome by PP at different phylogenetic levels. Loss of diversity occurred during the study, even in the control group, but was more noticeable in fish subjected to PP than in those challenged with F. columnare. Fish treated with PP and challenged with the pathogen exhibited the least diverse microbiome at the end of the study.  相似文献

A 1.5‐year‐old, 23 kg intact male Dalmatian dog was evaluated for acute respiratory insufficiency without a previous history of trauma or toxic exposition. Imaging revealed pneumomediastinum, pneumothorax, diffuse unstructured interstitial pulmonary pattern, pulmonary interstitial emphysema, and pneumoretroperitoneum. Histopathological evaluation of the lungs revealed perivascular and peribronchial emphysema, mild lymphocytic interstitial pneumonia with atypical proliferation of type II pneumocytes in bronchioles and alveoli. A lung disease resembling fibrosing interstitial pneumonia in man and cats has been previously reported in Dalmatians and should be included as a differential diagnosis for Dalmatians with this combination of clinical and imaging characteristics.  相似文献

Abstract

Since the Deepwater Horizon oil spill in 2010, anecdotal observations of Red Snapper Lutjanus campechanus from the northern Gulf of Mexico exhibiting unusual external lesions have been reported. Two opportunistic bacterial fish pathogens, Vibrio vulnificus and Photobacterium damselae, were recovered from the fish and were deemed responsible for the abnormalities. However, the culturable microbiota of healthy Red Snapper has not yet been characterized. We analyzed the heterotrophic bacteria associated with healthy Red Snapper caught off the Louisiana coast. In total, 179 isolates from 60 fish were recovered from skin and mucus, and 43 isolates were obtained from anterior kidney. All isolates were identified by 16S ribosomal RNA gene sequencing. The Proteobacteria was the predominant phylum in both external and internal samples, followed by the Firmicutes and the Actinobacteria. Within the Proteobacteria, most isolates were members of the genera Vibrio and Photobacterium, and V. natriegens and P. damselae were the predominant species. The results of this study suggest that both Vibrio spp. and Photobacterium spp. are associated with the normal microbiota of healthy Red Snapper. Thus, the opportunistic fish pathogens recovered in previous studies cannot be deemed lesion-forming until Koch's postulates are fulfilled.

The suitability of watermelon cultivars and cucurbit rootstocks as hosts to Meloidogyne incognita and M. javanica was determined in pot and field experiments. Meloidogyne incognita showed higher reproduction than did M. javanica on watermelon and cucurbit rootstocks. The watermelon cultivars did not differ in host status when challenged with these two species and supported lower nematode reproduction than the cucurbit rootstocks. Rootstocks Lagenaria siceraria cv. Pelops and Cucurbita pepo AK15 supported lower reproduction than did the squash hybrid rootstocks (C. maxima × C. moschata). Egg production increased (< 0·05) with a rising initial inoculum level (Pi) in the non‐grafted Sugar Baby but the reproduction factor Rf (eggs per plant/Pi) was similar at two Pi levels. The total egg production in the plants grafted onto squash hybrids RS841 and Titan was greater (< 0·05) at the higher Pi, but the Rf values were lower. The development of field‐grown non‐grafted watermelon plants was significantly stunted in plots where nematodes were detected at planting. However, no differences were observed in plots with grafted plants. In plots with nematodes, non‐grafted and Titan‐grafted plants had similar yields that were higher than that of RS841‐grafted plants. In the commercial plastic houses with grafted watermelon, the average Rf value was 42‐fold, confirming the high susceptibility of squash hybrids as rootstocks for grafted watermelon. The Titan–Sugar Baby combination was tolerant to M. javanica.  相似文献

Probiotic supplementation in fish aquaculture has significantly increased in the last decade due to its beneficial effect on fish performance. Probiotic use at early stages of fish development may contribute to better face metamorphosis and weaning stress. In the present work, we studied the influence of Shewanella putrefaciens Pdp11 supplementation on growth, body composition and gut microbiota in Senegalese sole (Solea senegalensis) during larval and weaning development. S. putrefaciens Pdp11 was incorporated using Artemia as live vector (2.5 × 107 cfu mL?1) and supplied to sole specimens in a co-feeding regime (10–86 DAH) by triplicate. Probiotic addition promoted early metamorphosis and a significantly higher growth in length at 24 DAH larvae. S. putrefaciens Pdp11 also modulated gut microbiota and significantly increased protein content and DHA/EPA ratios in sole fry (90 DAH). This nutritional enhancement is considered especially important after weaning, where significantly higher growth in length and weight was observed in probiotic fish. Moreover, a less heterogeneous fish size in length was detected since metamorphosis till the end of weaning, being of interest for sole aquaculture production. After weaning, fish showed significantly higher growth (length and weight) and less variable lengths in fish when supplemented with probiotics. Both the enhancement of nutritional condition and the decrease in size variability associated with probiotic addition are highly interesting for sole aquaculture production.  相似文献

Recovery of bacteria from infected fish during population sampling can be affected by factors including the type of assay, method of specimen preservation and concentration of bacteria present. Consequently, before use in field sampling, methods should be validated. The three objectives of this study were, first, to determine whether a channel catfish Ictalurus punctatus (Rafinesque) fingerling classified as positive for Gram‐negative Edwardsiella ictaluri infection according to bacterial culture before freezing was also classified as positive after freezing, second, to determine how direct culture from the kidney (DIRECT), culture of homogenate (HOMOG) and standard PCR (PCR) agree with bacterial culture in terms of classifying fish as positive or negative and third, to estimate diagnostic sensitivity (dSe) and diagnostic specificity (dSp) for DIRECT, HOMOG and PCR. In fresh and frozen fish, as bacterial concentration decreased, the ability of each assay to detect positive fish also decreased, especially when there were <104 colony‐forming units per gram (CFU g?1) tissue. HOMOG proved to be the most reliable at correctly classifying catfish, whether they were subclinically or clinically infected. PCR assay was the least reliable. Overall, values for this study population for dSe were 0.66, 0.92 and 0.43, and for dSp were 0.86, 0.91 and 0.95, for DIRECT, HOMOG and PCR respectively.  相似文献

After natural desiccation in the field, anhydrobiotic populations ofPratylenchus thornei Sher & Allen andMerlinius brevidens (Allen) Sidiqqi were treated in three different ways: some were kept dry to maintain nematodes in the anhydrobiotic stage; some were rehydrated and then allowed to dry again to induce a new desiccation cycle; and some were rehydrated and maintained as such, to reactivate and keep nematodes active.P. thornei populations from the dry treatment had a greater survival rate than the two other treatments. Culturing of the nematodes under wheat revealed that the three treatments did not alter the ability ofP. thornei to penetrate the roots, but nematodes given two cycles of desiccation had a greater rate of reproduction than those exposed to one desiccation cycle and kept in dry soil for a longer period (dry treatment). Survival ofM. brevidens rehydrated and retained in the active state was lower than that of nematodes undergoing the two other treatments.  相似文献

The aim of this study was to describe normal ECG patterns and values in four species of conscious raptors (Eurasian kestrel, Griffon vulture, Little owl, and Eurasian Eagle owl). Electrocardiograms were carried out in 75 conscious birds belonging to four species of raptors. Lead II waveforms were analysed to determine amplitudes and durations of waves and intervals. Morphologic patterns of P-QRS-T deflections were analysed in the six limb leads. Rhythm, heart rate, mean electrical axis, presence of Ta wave, ST slurring, and P-on-T phenomenon were also studied. The influence of species, body weight and heart rate in electrocardiographic variables were statistically analysed (P < 0.05). Sinus rhythm was present in all tracings, showing sinus arrhythmia in four cases. Ta wave was present in six tracings and P-on-T phenomenon in four. ST segment could be identified in all tracings, being mainly high above baseline. Significant differences between species were found for all the electrocardiographic parameters. The heart rate and body weight were also found to be a significant influence in most parameters. This study provides electrocardiographic data for four species of raptors that can be used to establish comparisons for clinical purposes.  相似文献

Laboratory‐reared, specific pathogen‐free fingerling channel catfish, Ictalurus punctatus, and adult zebrafish, Danio rerio, were each separately exposed by immersion challenge to the etiological agent of “columnaris disease,”Flavobacterium columnare (Japanese Collection of Microorganisms 21327 strain). At 24‐h post‐immersion, fish exhibiting a “saddleback” lesion were fixed whole in 10% neutral buffered formalin. Skin samples approximately 5 mm2 were excised from both the margin and center of each saddleback lesion as well as from corresponding sites in control, non‐challenged, fish before being prepared routinely for scanning electron microscopy (SEM). Skin samples from control channel catfish and zebrafish had uniform, contiguous epidermal cells with continuous or closely apposed cell margins and well‐defined microridges. Channel catfish skin lesion samples had margins typified by epidermal sloughing and lesion centers that exhibited a multitude of rod‐shaped bacterial cells, approximately 3–10 µm long × 0.3–0.5 µm wide, intermingled with cellular debris across a surface characterized by denuded, strongly ridged, or folded dermal connective tissue. Zebrafish skin lesion samples had a multitude of rod‐shaped bacterial cells and exhibited comparable ultrastructural changes but some lacked scales. These findings are the first published SEM observations of columnaris disease and saddleback lesions in channel catfish and zebrafish and thereby advance our understanding of the ultrastructural characteristics of acute‐stage saddleback lesions and columnaris disease pathogenesis.  相似文献
