Necropsy records for 102 Attwater's prairie chickens (Tympanuchus cupido attwateri) that had been captive-reared and had died at the San Antonio Zoo over a 4-yr period were evaluated to determine the causes of mortality. Gross necropsy, histopathology, and bacterial culture findings are summarized. Mortality was highest in young birds (<6 mo) and embryos. Summary Various genes for disease resistance identified in wild Lactuca are difficult, even impossible to exploit in lettuce breeding, due to sexual incompatibility between L. sativa and wild Lactuca sp. We adapted two cellular biology techniques to overcome these interspecific barriers: in vitro embryo rescue and protoplast fusion. In vitro rescue of immature embryos was used successfully for sexual hybridization between L. sativa and L. virosa. Vigorous hybrid plants were produced between L. sativa and seven accessions of L. virosa. Protoplast fusion permitted the regeneration of somatic hybrids between L. sativa and either L. tatarica or L. perennis. Hybrids between L. sativa and L. tatarica were backcrossed to L. sativa. 相似文献

Summary Rhizomania is a disease of sugar beet caused by the furovirus beet necrotic yellow vein virus (BNYVV). Coat protein mediated resistance has been reported for a number of viral diseases. This approach to virus resistance was therefore attempted for control of rhizomania. Two constructs of the coat protein gene of BNYVV were introduced into sugar beet by Agrobacterium-mediated transformation. The mRNA level was estimated to be 0.01% of the poly A+ RNA. Expression of the coat protein gene was under the detection limit of our western blotting protocol i.e. below 0.01 g/50 g (0.02% of the total soluble protein). One transformation event per construct was tested in a greenhouse assay and in rhizomania infested soil in a field trial. In the greenhouse assay, transgenic plants showed a strong reduction of virus multiplication when compared to non-transgenic plants. This result was confirmed in the field trial, where a significant difference in virus multiplication was shown between plants with and without the coat protein gene. 相似文献

It is increasingly important to be able to monitor and maintain the quality of estuaries for overwintering shorebirds. Bird numbers alone are not sufficient to indicate quality nor, as recent research shows, can it be assumed that site quality is being maintained simply because there is enough food to meet the birds’ physiological demands; i.e., the amount of food available per bird in autumn needs to be greater than the amount eaten. But how much greater? We used a simple individuals-based and behaviour-based model to explore the factors that affect the relationship between overwinter mortality and the amount of food available per bird in autumn (the food:bird ratio). The aims were to explore how the natural history characteristics of a shorebird species affected the shape of this relationship, in order to identify characteristics of the system that should be included in any monitoring programme of site quality. In all cases there was a minimum threshold food:bird ratio above which mortality did not vary and below which mortality increased steadily. The amount of food per bird required at the threshold varied from one to six times the amount actually consumed by each bird during winter, depending on the amount of realism included in the model. The minimum threshold ratio was most strongly influenced by the shape of the relationship between intake rate and prey density - the functional response - and, when interference competition occurred, by the distribution of the birds’ main food supply. Simulations with realistic spatial distributions of prey predicted that the minimum required amount of food per bird would be between two and five times the amount actually consumed. The threshold approach could provide a simple method of monitoring estuary quality based on food supply and bird numbers. It also provides a potentially clearly-defined and unambiguous measure of the carrying capacity of a site. If applied to a sufficient number of real estuaries, general guidelines for maintaining estuary quality based on estuary characteristics, food supply and bird species might be established. 相似文献

The origin of dimethyl sulfide (DMS) produced during wine aging was examined through different assays. The production of DMS during the model aging of a wine and the concomitant decrease of residual potential DMS (PDMS), as DMS released by heat-alkaline treatment in 0.5 M sodium hydroxide at 100 degrees C for 1 h, were demonstrated. Then, dimethyl sulfoxide (DMSO), methionine sulfoxide (MSO), S-methylmethionine (SMM), and dimethylsulfonium propanoic acid (DMSPA), reported previously as possible DMS precursors, were investigated for their ability to be DMS precursors in wine in the conditions of this model aging and of the heat-alkaline treatment. The results showed that DMSO, MSO, and DMSPA could hardly be DMS precursors in the conditions used, whereas SMM appeared to be a good candidate. Finally, the use of [(2)H(6)]-DMSPA as an internal standard for PDMS determination was proposed, because it provided better reproducibility than [(2)H(6)]-DMS used as an external standard. 相似文献

Constructed ponds are an important consideration in the conservation of wetland biota in agricultural landscapes. Twenty-two natural ponds and 22 adjacent constructed ponds (farm dams) were surveyed on the Southern Tablelands of New South Wales to compare patterns of use by frogs and develop frog conservation recommendations. Farm dams supported similar numbers of frog species to natural ponds, although differences in frog assemblage were observed between the pond types. Limnodynastes tasmaniensis and Uperolia laevigata were significantly more likely to occur at farm dams while L. peronii was more likely to occur at natural ponds. Results suggest waterbodies with high levels of emergent vegetation cover that lack fish are likely to support a high number of frog species, regardless of origin (i.e. natural or constructed). However, it is important for landholders to conserve natural waterbodies as these environments appear likely to support frog species that do not use farm dams. 相似文献

Bioavailability of dietary folate might be impaired by the polyglutamate chain to which approximately 70% of dietary folates are bound. This chain must be removed enzymatically in the intestine before folate is absorbed as a monoglutamate. To increase formation of monoglutamate folate in vegetables, the vegetables were subjected to various processing treatments. Treatments included freezing (-18 degrees C, 16 h) and thawing (4 degrees C, 24 h) and hydrostatic high-pressure treatment (200 MPa, 5 min). Both freezing/thawing and high-pressure treatment increased the proportion of folate in the monoglutamate form in leeks, cauliflower, and green beans 2-3-fold. However, loss of total folate after these treatments was >55%. It is concluded that conversion of folate polyglutamate to the monoglutamate form in vegetables is possible by certain processing treatments. Potentially this could lead to vegetables with higher folate bioavailability. However, to prevent folate loss into processing water, processing in a closed system should be applied. 相似文献

Antioxidant activity, total phenolic content, anthocyanin content, and six other fruit characters including titratable acid concentration, soluble solids, firmness, and percentage of bruised berries were determined for nine blueberry (Vaccinium L. sp.) cultivars at harvest and at various postharvest intervals after storage at 5 degrees C. Berries from MSU-58, Brigitta, and Legacy stored successfully for 7 weeks, Bluegold stored for 3-5 weeks, Bluecrop, Elliott, and Nelson stored for 3 weeks, and Jersey and Little Giant stored for fewer than 3 weeks. During the time they retained marketable quality, one cultivar (MSU-58) demonstrated a 29% increase in antioxidant activity. None of the cultivars showed a significant decrease from the harvest antioxidant activity value during storage. Antioxidant activity, total phenolic content, and anthocyanin content were strongly correlated with each other (r = 0.87-0.99, P < 0.01). All three parameters were moderately correlated with soluble solids (r = 0.47, P < or =0.05; r = 0.44, P < or = 0.05; and r = 0.64, P < or = 0.01, respectively), and antioxidant activity and total phenolic content were both moderately correlated with pH (r = 0.53 and 0.49, respectively; P < or = 0.05). However, antioxidant activity, total phenolic content, and anthocyanin content showed no correlation with firmness, percent severely bruised berries, or weight loss. Antioxidant activity and total phenolic content at harvest both correlated with titratable acidity at harvest (r = 0.68, P < or = 0.05 and r = 0.70, P < or = 0.05, respectively) on a cultivar mean basis. Berries from Elliott were also harvested from plants at two levels of bush ripeness (30-50% and 60-80% ripe berries on plants) and separated into three fruit maturity classes on the basis of percent blue color. The level of bush ripeness had no significant effect on antioxidant activity, total phenolic content, or anthocyanin content; however, fruit maturity had a significant effect on antioxidant activity, total phenolic content, and anthocyanin content, and bush ripeness x fruit maturity interactions were significant for these three traits. Berries with 50-75% blue coloration harvested from bushes with 60-80% mature fruit showed a significant increase in antioxidant activity, total phenolic content, and anthocyanin content during the first 3 weeks in storage. Our results demonstrate that increases in antioxidant activity, total phenolic content, and anthocyanin content may occur in the blueberry during cold storage and are cultivar-dependent. The increases that occur in immature fruit, such as in Elliott, may be advantageous for producers who wish to delay marketing of the fruit. 相似文献
