The Sfh protein is encoded by self-transmissible plasmids involved in human typhoid and is closely related to the global regulator H-NS. We have found that Sfh provides a stealth function that allows the plasmids to be transmitted to new bacterial hosts with minimal effects on their fitness. Introducing the plasmid without the sfh gene imposes a mild H-NS(-) phenotype and a severe loss of fitness due to titration of the cellular pool of H-NS by the A+T-rich plasmid. This stealth strategy seems to be used widely to aid horizontal DNA transmission and has important implications for bacterial evolution.  相似文献

Transforming growth factor-β (TGFβ) signaling drives aneurysm progression in multiple disorders, including Marfan syndrome (MFS), and therapies that inhibit this signaling cascade are in clinical trials. TGFβ can stimulate multiple intracellular signaling pathways, but it is unclear which of these pathways drives aortic disease and, when inhibited, which result in disease amelioration. Here we show that extracellular signal-regulated kinase (ERK) 1 and 2 and Smad2 are activated in a mouse model of MFS, and both are inhibited by therapies directed against TGFβ. Whereas selective inhibition of ERK1/2 activation ameliorated aortic growth, Smad4 deficiency exacerbated aortic disease and caused premature death in MFS mice. Smad4-deficient MFS mice uniquely showed activation of Jun N-terminal kinase-1 (JNK1), and a JNK antagonist ameliorated aortic growth in MFS mice that lacked or retained full Smad4 expression. Thus, noncanonical (Smad-independent) TGFβ signaling is a prominent driver of aortic disease in MFS mice, and inhibition of the ERK1/2 or JNK1 pathways is a potential therapeutic strategy for the disease.  相似文献

A neutered female cat presented with a 9-day history of hyporexia and depression. The referring veterinarian had identified moderate non-regenerative anaemia, haematuria and suspected unilateral obstructive ureterolithiasis. Subsequent ultrasonography revealed moderate distension of the left renal pelvis with echogenic material, ureteral distension and ureterolithiasis. A partial ureteral obstruction was suspected. After 4 days of medical management, there was further distension of the renal pelvis with well-delineated echogenic material and an accumulation of perinephric fluid. A left nephroureterectomy was performed. Renal pelvic rupture with intrapelvic haematoma and retroperitoneal haemorrhage was confirmed by histopathology. Eighteen months following surgery, the cat remained clinically well with normal renal values.  相似文献

Modeling how crop plants evolve under domestication requires estimating among-plant variation in important parameters of the reproductive system, including fecundity – the number of propagules produced – and propagule quality. Measuring these traits poses particular problems in vegetatively propagated crop plants. Unlike seeds, vegetative propagules are not intrinsic biological entities but are prepared by farmers. Propagule number and quality are thus determined by the interaction between plant traits and how farmers prepare propagules. We conducted observations, interviews and experiments to study this interaction in cassava grown by Makushi Amerindians, examining how both sources of variation, in plant traits and in farmers’ practices, combine to determine the number and quality of propagules produced. Increased stake mass, determined mostly by stem diameter, leads to increased yield and also to increased asexual ‘fecundity’ of the resulting plant. Farmers’ practices reflect knowledge of this relationship. Diameter is the key criterion in the selection of stems for stakes. Larger diameters are preferred; when thinner stems are used, stakes are cut longer, partially compensating for reduced mass. These results suggest that conscious and unconscious selection to increase ‘fecundity’ and propagule quality in cassava would act to favor plants with thicker stems. Mean stem diameter is greater, and variation in stem diameter is lower, in little-branched plants. Selection for increased asexual ‘fecundity’ can thus have led to reduction in the degree of branching, one of the most striking differences between domesticated cassava and its wild ancestors. Measuring variation in asexual fecundity is a key step in analyzing evolution of the mixed clonal/sexual reproductive systems that characterize many vegetatively propagated crop plants.  相似文献

Pre‐Columbian agricultural raised fields are widespread in Neotropical wetlands. Many seasonally flooded coastal savannahs of French Guiana are dotted with thousands of small, round mounds, which current research indicates are vestiges of raised fields. It is surprising that these elevated structures persist under the erosive force of heavy tropical rainfall. One hypothesis for this stability is that “engineer” organisms such as plants, earthworms, termites and ants colonize abandoned raised fields and maintain the raised structures against erosion. This “erosion/deposition balance” hypothesis assumes that the landscape is subject to non‐negligible rates of erosion, but that actions of engineer organisms reduce the erodibility of mounds, compensate for erosion by transporting materials to mounds, or both. The hypothesis also predicts greater bioturbation on mounds than in other parts of the landscape. To test these assumptions and predictions, we estimated soil erosion and bioturbation rates in abandoned raised fields in a seasonally flooded savannah in French Guiana by using two radionuclides (137Cs and unsupported 210Pb) as tracers for soil redistribution. Analysis of the vertical and horizontal distribution of radionuclides at the study site showed that soil erosion in the mounds either occurs at a rather low rate or that organisms counteract erosion, or both. We also showed that mounds were heavily influenced by bioturbation. These results validate the fundamental assumptions of the erosion/deposition balance hypothesis.  相似文献
