Summary Three mollisols, typical of the Palouse winter wheat region of eastern Washington and northern Idaho, were analyzed for microbial biomass, total C and total N after 10 years of combined tillage and rotation treatments. Treatments included till, no-till and three different cereal-legume rotations. All crop phases in each rotation were sampled in the same year. Microbial biomass was monitored from April to October, using a respiratory-response method. Microbial biomass, total C and total N were highest under no-till surface soils (0–5 cm), with minimal differences for tillage or depth below 5 cm. Microbial biomass differences among rotations were not large, owing to the relative homogeneity of the treatments. A rotation with two legume crops had the highest total C and N. Microbial biomass was significantly higher in no-till surface soils where the current crop had been preceded by a high-residue crop. The opposite was true for the tilled plots. There was little change in microbial biomass over the seasons until October, when fresh crop residues and rains had a strong stimulatory effect. The seasonal pattern of biomass in no-till surface soils reflected the dry summer/winter rainfall climate of the region. The results of this study show that numerous factors affect soil microbial biomass and that cropping history and seasonal changes must be taken into account when microbial biomass data are compared.Scientific paper no. 7634  相似文献

Batch and upflow column leaching experiments were used to evaluate the nature and extent of Cu and Zn solubilization from contaminated soil by nitrilotriacetic acid (NTA) in 0.025 M NaClO4. In batch soil suspensions, NTA levels of 10?5 to 10?3 M substantially promoted Cu and Zn release from the metal-enriched soil. The ability of NTA to enhance Cu and Zn solubility decreased with increasing solution acidity probably due to competitive binding of NTA by protons and Fe released by hydrous oxide dissolution. However, in the pH range typically encountered in northeastern U.S. soils, soluble metal levels were nearly constant for a given NTA concentration. Leaching soil columns with NTA solutions enhanced Cu release more than Zn, as the enrichment ratio (cumulative metal leached by NTA compared to the 0.025 M NaClO4 control leachate) after 85 pore volumes displacements was 23.6 and 4.3 for Cu and Zn, respectively. While Cu release by 0.01 M CaCl2 differed little from the control, 0.01 M CaCl2 was substantially more effective than 10?5 M NTA in displacing bound Zn. When the use of tylosin as a feed additive was forbidden by Council Regulation 2821/98, the necessity of a chemical confirmation method for the monitoring of the ban was created. Recently a method was developed for the detection of tylosin in animal feed by means of LC-MS/MS. During the validation high deviating values for the decision limit, detection capability, and repeatability for tylosin in cattle feed were observed, and the presence of urea and the formation of a tylosin urea adduct (TUA) were suggested as possible explanations. In this study two hydrolysis approaches for the TUA adduct were compared, namely, a chemical hydrolysis and an enzymatic hydrolysis with urease. The latter yielded a more complete hydrolysis of urea and was used for further validation. The recovery increased by approximately 15-25% depending on the amount of urea present in the feed (0.5-2%). The decision limit and detection capability were hardly influenced by the enzymatic hydrolysis.  相似文献

Work in swine and poultry units is associated with exposure to significant levels of organic dust and endotoxins with the highest concentrations found in poultry houses, whereas values found in dairy and in cattle farming are much lower. Corresponding to this is an excess of work-related respiratory symptoms in swine farmers. A dose-response relationship exists between symptoms and number of working hours. Longitudinal studies have demonstrated an accelerated decline of lung function in swine farmers large enough to cause clinically significant disease in some farmers. Because of the large number of people needed in swine farming and the long working hours, swine farming has emerged as the major respiratory problem in farming. Experimental studies indicate that exposure has to be lowered substantially to avoid acute effects and longitudinal studies demonstrate that loss of lung function occurs in non-smoking swine farmers without respiratory symptoms and that accelerated decline in lung function occurs below endotoxin concentrations in dust (100 ng/m3) proposed as a safe threshold.  相似文献

Because the economics of soil extraction processes depend on conservation and reuse of costly chelating agents, the ability of various electrolytes to modify EDTA extraction of Pb from a grossly-contaminated soil (PbT=21%) was investigated using batch equilibration experiments. In the absence of added electrolyte, a single 5-hr. extraction with 0.04 M EDTA (corresponding to 1∶1 PbT to EDTA ratio) released 65% of PbT over the pH 5 to 9 range. Under these conditions, Na+-, Li+-, and NH4ClO4 salts at 0.5 M increased Pb desorption to nearly 80%, probably from exchange displacement of soilbound Pb2+ and increased solubility of Pb-containing phases at higher ionic strength. Because Cl? and ClO4 ? salts were equally effective, chlorocomplex formation was insignificant. Under slightly acidic conditions, Ca(ClO4)2 and Mg(ClO4)2 at 0.167 M caused roughly the same elevation in Pb recovery as 0.5M of the monovalent electrolytes. However, with progressively higher pH, Ca, and to a lesser extent Mg, suppressed Pb solubilization by competitive chelation of EDTA. Pb recovery by EDTA soil washing could be enhanced by addition of Ca salts at pH 4 to 6. Subsequent pH elevation in the presence of Ca would promote decomposition of Pb-EDTA complexes and separation of Pb as a hydroxide precipitate.  相似文献

The development of an assay for the detection of streptomycin residues in pasteurized whole milk using an optical biosensor (Biacore) is reported. Streptomycin-adipic hydrazide coupled to bovine thyroglobulin was used to produce a sheep polyclonal antibody. The antibody displayed excellent cross-reactivity with dihydrostreptomycin (106%). There was no significant cross-reaction with other aminoglycosides or common antibiotics. Streptomycin was also immobilized onto a CM5 sensor chip to provide a stable, reusable surface. The developed assay permitted the direct analysis of whole milk samples ( approximately 3.5% fat) without prior centrifugation and defatting. Results were available in 5 min. The limit of detection of the assay was determined as 4.1 ng/mL, well below the European maximum residue limit (MRL) of 200 ng/mL. Repeatability (or coefficient of variation) between runs was determined as 3.5% (100 ng/mL; 0.5 x MRL), 5.7% (200 ng/mL; MRL), and 7.6% (400 ng/mL; 2 x MRL).  相似文献

Alterations in thyroid indices in critically ill dogs were studied retrospectively to determine the incidence of the euthyroid sick syndrome and to assess its prognostic relevance to survival. Sixty-seven dogs were classified as euthyroid bases on a TSH stimulation test. Forty-one of 67 (61%) of the euthyroid dogs had low baseline serum T4 concentrations (<1.5 ug/dl) and 38 of 67 (56%) of the euthyroid dogs had low baseline serum T3 concentrations (<75 ng/dl). There was no significant difference between either the baseline or the post-stimulation serum T4 concentrations when values of dogs that survived were compared with those that died. There was, however, a significant difference (P<0.05) between both the baseline and the post-stimulation values for T3 when survivors (100.3± 81 ng/dl, 143± 66 ng/dl) and non-survivors (64.2 ± 17.1 ng/dl, 96.6 ± 38.3 ng/dl) were compared. The euthyroid sick syndrome occurs commonly in critically ill dogs. Further, the extent of depression of serum concentrations of T3 may be correlated to mortality.  相似文献
