Previous studies have indicated a relationship between genome size and cold tolerance in plants. Many species adapted to growth in cool environments have large genome sizes. These studies are based on interspecific DNA content variation. In this study, the nuclear DNA content of eight maize populations was determined. These populations were obtained from the University of Nebraska and represent populations selected for cold tolerance and their respective unselected original populations. Intraspecific DNA content variation was observed between the selected and unselected populations. Upon assessing the data based solely on cold tolerance, no clear relationship between genome size and cold tolerance was apparent. When both freeze tolerance and cold tolerance were considered, populations which were cold tolerant and exhibited a certain degree of freeze tolerance were observed to have significantly larger genomes relative to the unselected populations. Thus, it appears that the relationship between intraspecific genome size variation and cold tolerance is similar to the relationship between interspecific genome size variation and growth at cooler temperatures in plants. 相似文献

An in vitro growth system was used to determine the virulence of two samples of Striga gesnerioides from Zakpota in southern Benin. Cowpea variety B301, previously considered resistant to all races of S. gesnerioides, was susceptible to both samples of the parasite. Two other cowpea varieties, 58–57 and IT81D-994, were totally resistant. Resistance in 58–57 was associated with a hypersensitive necrosis of infected roots, whilst IT81D-994 supported production of small S. gesnerioides tubercles with stems which failed to develop. Striga gesnerioides from southern Benin is the fourth race of the parasite to be identified, and the first with virulence on variety B301. The implications for breeding cowpeas with resistance to S. gesnerioides are discussed.Abbreviations IITA

Summary Cultivar specific DNA profiles in rye were revealed by polymerase chain reaction (PCR) using randomly amplified polymorphic DNA (RAPD) sequences. Ten base primers were used for the amplification of genomic DNA of rye cultivars by PCR. RAPD analysis was found to be reproducible among samples between PCR runs. When amplification profiles of different rye cultivars were compared using various primers, the overall profiles were cultivar specific. However, not all primers revealed polymorphisms. These primers appear to amplify conserved sequences in all rye cultivars. Intracultivar studies were conducted on two of the cultivars. In the cultivar Imperial, no polymorphisms were observed among ten plants analyzed with five primers. In the cultivar Balboa, polymorphisms were observed among fifty plants with four of the ten primers analyzed. Despite the small amount of intracultivar variability, RAPD analysis has the potential to be a rapid and reliable method of cultivar identification in this outcrossing species. 相似文献

Literature searches and recent surveys of the bat fauna of Singapore indicate that of the 24 species of Microchiroptera and six species of Megachiroptera documented for this small equatorial island just 15 and 5, respectively, are still present. These recorded declines in chiropteran species richness almost certainly understate the true losses as extensive land transformation/habitat loss (>95%) and biota loss occurred early in Singapore’s colonial history before comprehensive surveys of bats were made. In an effort to reconstruct the pre-settlement bat fauna, we inferred an upper bound of pre-settlement species richness using a checklist from a well-known bat assemblage in neighbouring Peninsular Malaysia, and a lower bound based on species common to Peninsular Malaysia, Borneo and Sumatra. The Singapore records were compared with these two species list predictions. Based on this analysis, we infer that between 60 and 72 species would have inhabited Singapore before 1819. We also estimate that between 33% (based on the confirmed inventory) and 72% of the species (based on the upper-bound estimate of species richness) are now locally extinct. For Microchiroptera the data suggest that the documented local extinction rate of 38% may project to between 69% and 75%. Forest-dependant bats are particularly affected and comprise a much lower proportion of the bat fauna than is seen in intact forest in Peninsular Malaysia. All hipposiderids and 40% of the documented rhinolophid taxa have been lost and almost half (6) of the surviving microchiropteran species are locally endangered. Projected local extinction rates for Megachiroptera raise the documented value (17%) to about 60%, with most of the survivors being widespread species known to forage in cultivated or secondary forest habitats or to commute long distances between fragmented resources. The dramatic decline in Singapore bat species richness and a concomitant change in chiropteran guild and trophic structure (Microchiroptera vs. Megachiroptera) reflect patterns of diversity change seen elsewhere in the region in response to loss of forest habitat. In Singapore, the decline in diversity (species richness and abundances) for both mega- and microbats may also relate to urbanisation and deliberate or accidental destruction of bats and their roost sites in a land that has one of the highest human population densities on the planet. Although these losses (actual and inferred) represent a microcosm of mainly local extinctions, a wider geographical extrapolation over the 21st century indicates that heavy deforestation in progress in Southeast Asia might be expected to lead to extinction of many bat taxa, with upper-bound estimates of regional species losses exceeding 40% and global extirpation anticipated for at least 23% of Southeast Asia’s bat fauna by 2100. 相似文献

The essential Cdc13 protein in the yeast Saccharomyces cerevisiae is a single-stranded telomeric DNA binding protein required for chromosome end protection and telomere replication. Here we report the solution structure of the Cdc13 DNA binding domain in complex with telomeric DNA. The structure reveals the use of a single OB (oligonucleotide/oligosaccharide binding) fold augmented by an unusually large loop for DNA recognition. This OB fold is structurally similar to OB folds found in the ciliated protozoan telomere end-binding protein, although no sequence similarity is apparent between them. The common usage of an OB fold for telomeric DNA interaction demonstrates conservation of end-protection mechanisms among eukaryotes. 相似文献

The Rho guanosine triphosphatases (GTPases) Rac1 and Rac2 are critical signaling regulators in mammalian cells. The deletion of both Rac1 and Rac2 murine alleles leads to a massive egress of hematopoietic stem/progenitor cells (HSC/Ps) into the blood from the marrow, whereas Rac1-/- but not Rac2-/- HSC/Ps fail to engraft in the bone marrow of irradiated recipient mice. In contrast, Rac2, but not Rac1, regulates superoxide production and directed migration in neutrophils, and in each cell type, the two GTPases play distinct roles in actin organization, cell survival, and proliferation. A reflectance spectrum of Phobos (from 200 to 1100 nanometers) has been compiled from the Mariner 9 ultraviolet spectrometer, Viking lander imaging, and ground-based photometric data. The reflectance of the martian satellite is approximately constant at 5 percent from 1100 to 400 nanometers but drops sharply below 400 nanometers, reaching a value of 1 percent at 200 nanometers. The spectral albedo of Phobos bears a striking resemblance to that of asteroids (1) Ceres and (2) Pallas. Comparison of the reflectance spectra of asteroids with those of meteorites has shown that the spectral signature of Ceres is indicative of a carbonaceous chondritic composition. A physical explanation of how the compositional information is imposed on the reflectance spectrum is given. On the basis of a good match between the reflectance spectra of Phobos and Ceres and the extensive research that has been done to infer the composition of Ceres, it seems reasonable to believe that the surface composition of Phobos is similar to that of carbonaceous chondrites. This suggestion is consistent with the recently determined low density of Mars's inner satellite. Our result and recent Viking noble gas measurements suggest different modes of origin for Mars and Phobos. 相似文献

Models describing the evolution of the partial pressure of atmospheric oxygen over Phanerozoic time are constrained by the mass balances required between the inputs and outputs of carbon and sulfur to the oceans. This constraint has limited the applicability of proposed negative feedback mechanisms for maintaining levels of atmospheric O(2) at biologically permissable levels. Here we describe a modeling approach that incorporates O(2)-dependent carbon and sulfur isotope fractionation using data obtained from laboratory experiments on carbon-13 discrimination by vascular land plants and marine plankton. The model allows us to calculate a Phanerozoic O(2) history that agrees with independent models and with biological and physical constraints and supports the hypothesis of a high atmospheric O(2) content during the Carboniferous (300 million years ago), a time when insect gigantism was widespread. 相似文献
