The effects of porcine sarcoplasmic proteins (SP) on the physicochemical properties of meat emulsion gel were examined. Meat emulsion was prepared from water‐washed pork meat (WWM), corn oil and SP. Whole SP (W‐SP) enhanced the breaking stress of the WWM emulsion gel as well as other animal proteins in the presence of 0.2 mol/L NaCl. The breaking stress of the emulsion with W‐SP increased with an increase in corn oil content. Furthermore, this tendency was more noticeable at a lower NaCl concentration (0.15 mol/L) rather than at 0.45 mol/L NaCl. Ammonium sulfate (AS) treatment fractionated W‐SP into three portions (SP‐f1, SP‐f2 and SP‐f3), which were the precipitates at 0–50% and 50–75% AS saturation and the supernatant at 75% AS saturation, respectively. The fractions SP‐f2 and SP‐f3 increased the gel strength more than W‐SP. In particular, the fraction SP‐f3 increased the gel strength approximately 10‐fold compared to the control. Sodium dodecyl sulfate‐polyacrylamide gel electrophoresis analysis showed that SP‐f3 had several kinds of proteins and a main protein with a molecular mass of 35 kDa, which corresponded to glyceraldehyde 3‐phosphate dehydrogenase. These results indicate that the influence of SP should not be ignored when processing of low‐salt meat products and the fraction SP‐f3 has a gel‐enhancing factor for myofibrillar proteins.  相似文献

Fifty canine lymphomas were classified cytomorphologically using the updated Kiel classification scheme. Aspirates of lymph nodes from dogs with lymphoma were stained using 5 canine-specific antibodies and 3 human-specific antibodies that cross-react with canine lymphocytes. The antibody-stained aspirates were analyzed by flow cytometry. A total of 32 (64%) of the 50 lymphomas were characterized as B-cell origin and 18 (36%) were of T-cell origin. B-cell lymphomas were identified in 12 females and 20 males with a mean age of 8.35 years. T-cell lymphomas were identified in 8 females and 10 males with a mean age of 7.9 years. A minority of the lymphomas were low-grade B-cell and T-cell lymphomas (6/50, 12% and 4/50, 8%, respectively). The most common morphologic types were high-grade centroblastic and unclassifiable plasmacytoid for B- and T-cell lymphomas (18/50, 36% and 7/50, 14%, respectively).  相似文献

Vitrification of in vitro matured oocytes of Mangalica and Large White pigs   总被引:1,自引:0,他引:1  
The breeding of Mangalica, a native pig breed in Hungary, had been started in 1833, but this pig breed almost became extinct in Hungary in the past decades. In 1991, the number of sows was only 200. Although in these days the existing Mangalica population consists of more than 6000 animals representing different colour variations, the preservation of this traditional pig breed is still very important. Vitrification is a potential tool for the preservation of gametes and embryos of these animals. The aim of this study was to investigate the effects of vitrification on the developmental competence of Mangalica (M) and Large White (LW) oocytes following fertilisation. The oocytes were vitrified by the Open Pulled Straw (OPS) method using different concentrations of ethylene glycol and dimethyl sulphoxide as cryoprotectants. After rehydration the oocytes underwent in vitro fertilisation; the resultant zygotes were then cultured in vitro for four days to assess embryonic development. In the first experiment, in vitro maturation of M and LW oocytes was compared. No significant difference was observed in the nuclear maturation rate of LW and M oocytes. In the second experiment, the sensitivity of oocytes to vitrification was examined by evaluating oocyte morphology after thawing. A higher percentage of LW oocytes showed normal morphology compared to M oocytes, indicating that Mangalica oocytes are more sensitive to cryoprotectants than Large White oocytes. After warming and in vitro fertilisation, more than 50% of the oocytes started embryonic development and by the end of the incubation period morula stage embryos had developed in both groups. The results show that the OPS vitrification technique is well suited to preserve Mangalica oocytes and from these oocytes morula embryos can be produced.  相似文献

Chondracanthus chamissoi is an important source of carrageenan in Chile. Presently, all the production is harvested from wild populations. This study reports the first attempt to cultivate C. chamissoi. Experiments were conducted with female gametophytic and sporophytic thalli by inserting them among braids of a 7 mm polypropylene rope. Inoculated ropes were placed at 1, 3 and 5 m depths in two sheltered bays in northern Chile. The fronds adapted well to cultivation and grew at all the depths tested, although the greatest increase in biomass was observed at 1 m depth. On a yearly cycle, higher biomass increases were observed in autumn and winter months compared with the spring and summer. We believe that by adapting the cultivation methodology, this species could be cultivated year round.  相似文献

Chlamydomonas reinhardtii is a unicellular green alga whose lineage diverged from land plants over 1 billion years ago. It is a model system for studying chloroplast-based photosynthesis, as well as the structure, assembly, and function of eukaryotic flagella (cilia), which were inherited from the common ancestor of plants and animals, but lost in land plants. We sequenced the approximately 120-megabase nuclear genome of Chlamydomonas and performed comparative phylogenomic analyses, identifying genes encoding uncharacterized proteins that are likely associated with the function and biogenesis of chloroplasts or eukaryotic flagella. Analyses of the Chlamydomonas genome advance our understanding of the ancestral eukaryotic cell, reveal previously unknown genes associated with photosynthetic and flagellar functions, and establish links between ciliopathy and the composition and function of flagella.  相似文献

The objective of this review is to make current knowledge on the nitrogen (N) load throughout the atmosphere (airborne N load) available to readers, with special emphasis on Japanese and Chinese agroecosystems. Key species of airborne N are ammonia, nitrogen dioxide, nitrogen oxide, nitric acid, nitrous acid and particulate ammonium and nitrate. Organic N also exists in the atmosphere. The main processes in terms of the airborne N load involve emission, atmospheric transportation and transformation, atmospheric deposition and environmental impacts. Agricultural activities are the largest emitters of ammonia through emissions mainly from livestock waste and field-applied N fertilizers. The ammonia emissions in China in 1995 from chemical fertilizers and organic fertilizers, such as animal excreta, were estimated to be 3.56 and 2.04 Tg N year−1, respectively, and the emissions in Japan were 0.059 and 0.069 Tg N year−1, respectively. The most fundamental causes of the airborne N load in relation to Japanese and Chinese agriculture were intensive livestock farming in Japan and over use of N fertilizers in China. However, agroecosystems are also a sink for airborne N. Atmospheric N deposition was up to 20 and 60 kg N ha−1 year−1 in Japan and China, respectively. The unrelenting load of airborne N continues in Japan and China. The development of a simple, but accurate method to determine the dry deposition flux that is applicable to simultaneous and multipoint observations would be valuable. The establishment of cross relationships among in situ observations, remote sensing and numerical modeling is also needed to cope with the issue by assessing the actual status, predicting the future status and working out effective measures.  相似文献

The present study aimed to elucidate ammonia (NH3) volatilization loss following surface incorporation (0–15 cm mixing depth) of nitrogen (N) fertilizer in an upland field of light-colored Andosol in central Japan. A dynamic chamber technique was used to measure the NH3 effluxes. Poultry manure, pelleted poultry manure, cattle manure, pelleted cattle manure and ammonium sulfate were used as N fertilizers for basal fertilization to a bare soil with surface incorporation. All three experiments in summer and autumn 2007 and in summer 2008 showed negligible NH3 volatilization losses following the application of all N fertilizers with the same application rate of 120 kg N ha−1 as total N; these negligible losses were primarily ascribed to chemical properties of the soil, that is, its high cation exchange capacity (283 mmolc kg−1 dry soil) and relatively low pH(H2O) (5.9). In addition, the surface incorporation, the very small ratio of ammoniacal N to total N for the manure, and the decrease in soil pH to ≤5.5 following applications of ammonium sulfate were also advantageous to the inhibition of NH3 volatilization loss from the field-applied N fertilizers.  相似文献

Abstract

The humus composition was analyzed and the humic acid characterized by UV and visible absorption spectroscopy in order to investigate the rotting and maturing process of city refuse compost according to the method of Kumada et al. During the composting process, the following findings were obtained: (1) the HT value was almost constant, but the HE/HT ratio varied somewhat, (2) HA increased with decrease in FA, and the PQ value so increased clearly, (3) the shoulder-like absorption at a wavelength near 270 nm weakened, and (4) the RF value of humic acid increased, whereas the Δ log K value seldom varied.

The IR spectrum of humic acid gradually changed as follows: (1) the absorption band in the 1700-1600 cm-1 region and in the 1550-1500 cm-1 region increased slightly, (2) the band in the 1100-1000 cm-1 region decreased, and (3) the bands at 835 and 710 cm-1 com pletely disappeared. On the whole, the shape of the IR spectrum of the city refuse compost became featureless. These changes were probably due to the oxidation which occurred in the composting process.  相似文献
