Initiation and maintenance of mitosis require the activation of protein kinase cyclin B-Cdc2 and the inhibition of protein phosphatase 2A (PP2A), which, respectively, phosphorylate and dephosphorylate mitotic substrates. The protein kinase Greatwall (Gwl) is required to maintain mitosis through PP2A inhibition. We describe how Gwl activation results in PP2A inhibition. We identified cyclic adenosine monophosphate-regulated phosphoprotein 19 (Arpp19) and α-Endosulfine as two substrates of Gwl that, when phosphorylated by this kinase, associate with and inhibit PP2A, thus promoting mitotic entry. Conversely, in the absence of Gwl activity, Arpp19 and α-Endosulfine are dephosphorylated and lose their capacity to bind and inhibit PP2A. Although both proteins can inhibit PP2A, endogenous Arpp19, but not α-Endosulfine, is responsible for PP2A inhibition at mitotic entry in Xenopus egg extracts.  相似文献

The aim of this study was to determine a temporary strategy for increasing the legume population in an established pasture of brachiaria grass (Brachiaria brizantha) and forage peanut (Arachis pintoi). The treatments comprised four previous long‐term canopy heights (10, 20, 30 and 40 cm) from the establishment (the first 32 months after treatment initiation). From September 2014, all of the experimental units were kept at 10 cm height. The pastures were evaluated for their forage harvest, forage mass, botanical composition and stolon and tiller density. The tiller density and brachiaria grass mass were maintained in all pastures since the first season of evaluation (p > 0.10). The stolon density and legume mass were initially lesser at 30 and 40 cm than those at 10 and 20 cm, early in the experiment, but progressively increased over time. After 1 year under 10 cm height, the population and mass of legume were similar in all treatments. Therefore, reducing the canopy height to 10 cm at the beginning of the rainy season is an efficient method to increase the botanical composition of forage peanut in mixed pastures with brachiaria grass.  相似文献

In strawberry, Fragaria × ananassa Duch., fruit, two different endo-β-1,4-glucanase genes, FaEG1 and FaEG3, also named Cel1 and Cel2, are expressed during the softening process that occurs during fruit ripening. It has also been suggested that FaEG3, which contains a putative cellulose-binding domain, could play a key role in fruit development, since previous attempts to down-regulate this gene through transgenesis have been unsuccessful. In this investigation, we obtained transgenic strawberry plants containing an antisense sequence of the FaEG3 gene under the control of the 35S promoter. Ripened fruit from transgenic lines (Acel lines) showed large variation in FaEG3 silencing, but fruit firmness was similar to control fruit in all the lines. Two Acel lines showing almost 95% reductions in FaEG3 mRNA levels were selected for further study. In these lines, FaEG3 down-regulation was high, from 78 to 95%, at all fruit developmental stages, whereas FaEG1 was only slightly suppressed. In spite of the high FaEG3 silencing achieved, EGase activity was not modified in ripe fruit. At the cell wall level, walls from transgenic ripe fruit contained a significantly higher amount of the 4 M KOH fraction, which is enriched in hemicellulosic polymers. The analysis of this fraction by size exclusion chromatography showed that transgenic cell walls contained a smaller amount of higher molecular mass polymers than controls. Altogether, these results indicate that FaEG3 does not play a key role either in fruit development or fruit softening. However, its silencing affects the amount and, in a minor way, the size of hemicellulosic polymers.  相似文献

Warm-season grasses are economically important for cattle production in tropical regions, and tools to aid in management and research of these forages would be highly beneficial. Crop simulation models synthesize numerous physiological processes and are important research tools for evaluating production of warm-season grasses. This research was conducted to adapt the perennial CROPGRO Forage model to simulate growth of the tropical species palisadegrass [Brachiaria brizantha (A. Rich.) Stapf. cv. Xaraes] and to describe model adaptation for this species. In order to develop the CROPGRO parameters for this species, we began with values and relationships reported in the literature. Some parameters and relationships were calibrated by comparison with observed growth, development, dry matter accumulation and partitioning during a 2-year experiment with Xaraes palisadegrass in Piracicaba, SP, Brazil. Starting with parameters for the bahiagrass (Paspalum notatum Flugge) perennial forage model, dormancy effects had to be minimized, and partitioning to storage tissue/root decreased, and partitioning to leaf and stem increased to provide for more leaf and stem growth and less root. Parameters affecting specific leaf area (SLA) and senescence of plant tissues were improved. After these changes were made to the model, biomass accumulation was better simulated, mean predicted herbage yield per cycle was 3573 kg ha−1, with a RMSE of 538 kg DM ha−1 (D-Stat = 0.838, simulated/observed ratio = 1.028). The results of the adaptation suggest that the CROPGRO model is an efficient tool to integrate physiological aspects of palisadegrass and can be used to simulate growth.  相似文献

This paper concerns a disease affecting a group of African grey parrots, which involves intranuclear inclusion bodies composed of filamentous material. The disease was characterized by either sudden death or death within 2-3 days from onset of non-specific symptoms. At necropsy, gross lesions included enlarged liver, mild hepatic congestion and focal necrosis. Samples from five birds were fixed in 10% formol and routinely processed for light and electron microscopy. In four birds, numerous hepatocytes displayed an enlarged nucleus, with peripheral margination of chromatin; the nucleus was partially or wholly filled by a basophilic inclusion body. In the remaining bird, inclusion bodies were acidophilic and completely filled the nucleus; nuclear enlargement was less evident than in the other birds. At ultrastructural examination, and in both types of IIB, nuclei contained looped filaments but no evidence of viral structures. However, virion-like structures were observed in the cytoplasm of some hepatocytes.  相似文献

Standard quality parameters, consumer acceptability, emission of volatile compounds and ethylene production of ‘Mondial Gala®’ apples (Malus × domestica Borkh.) were determined in relation to storage atmosphere, storage period and shelf-life period. Fruit were harvested at the commercial date and stored in AIR (21 kPa O2:0.03 kPa CO2) or under three different controlled atmospheres (CAs): LO (2 kPa O2:2 kPa CO2), ULO1 (1 kPa O2:1 kPa CO2), or ULO2 (1 kPa O2:2 kPa CO2). Fruit samples were analysed after 12 and 26 weeks of storage plus 1 or 7 d at 20 °C.Apples stored in CA maintained better standard quality parameters than AIR-stored fruit. The volatile compounds that contributed most to the characteristic aroma of ‘Mondial Gala®’ apples after storage were butyl, hexyl and 2-methylbutyl acetate, hexyl propanoate, ethyl butanoate, ethyl hexanoate, ethyl, butyl and hexyl 2-methylbutanoate. Data obtained from fruit analysis were subjected to principal component analysis (PCA). The apples most accepted by consumers showed the highest emission of ethyl 2-methylbutanoate, ethyl hexanoate, tert-butyl propanoate and ethyl acetate, in addition to the highest titratable acidity and firmness values.  相似文献

Thrips tabaci (Thysanoptera: Thripidae) is a major pest of onion worldwide. In 2011, research was conducted in a commercial onion field in northwestern Italy to: (i) assess the presence of autochthonous onion thrips predators on the crop; and (ii) evaluate the impact of the commonly used insecticides and alternative pest management strategies on onion thrips and its autochthonous predators. Toxicity of the active ingredients on local populations of onion thrips and its predatory thrips was also evaluated in laboratory bioassays. During field surveys, the highest and lowest thrips infestations were observed in plots treated with lambda-cyhalothrin and spinosad, respectively. The effectiveness of spinosad on T. tabaci was also confirmed in laboratory bioassays. The dominant zoophagous species Aeolothrips intermedius (Thysanoptera: Aeolothripidae) was more adversely affected by treatment with lambda-cyhalothrin, confirmed by a decrease in predator/prey ratios. The use of spinosad and acibenzolar-S-methyl is suggested as an alternative to conventional insecticides for the preservation of A. intermedius, which proved to be a potential biological control agent of T. tabaci.  相似文献

The aim of this study was to evaluate the addition of artificial substrates in biofloc technology systems that are used for the intensive production of Litopenaeus vannamei. The experiment lasted 35 days. Tanks each with a useful volume of 800 L were filled with water containing bioflocs (25 % of the total volume) and filtered seawater. Three treatments with three replicates each were compared: (1) Ctrl (control), i.e., without the addition of artificial substrate, (2) 200 and (3) 400, with added artificial substrate equivalent to 200 and 400 % of the lateral area of the tanks, respectively. The shrimp with initial weight of 0.40 ± 0.15 g were stocked at a density of 300 shrimp/m2. Feed (Guabi/38 Active, Brazil) was supplied two times per day. The water quality parameters were suitable for the production of L. vannamei. The settleable solids (SS) were higher in the Ctrl treatment (82.1 ± 19 mL L?1) and differed statistically from the 200 and 400 treatments that presented average SS values below 10 mL L?1. The reuse of water with bioflocs accelerated nitrification in the tanks, and no significant differences were shown between tanks. The presence of biofilm did not interfere with ammonium or nitrite levels, but it did serve as a food source that optimized shrimp performance, as shown by higher final weights. More studies are needed to evaluate the circulation and mixing intensity effects of the substrates on water throughout the production cycle.  相似文献

Corn gluten meal (CGM) has a high protein content and absence of antinutrients. However, it has high levels of carotenoids that can cause a yellowing of fish fillets and impair further commercialisation. Graded levels of CGM were incorporated in pacu diets to replace soybean meal (SM) protein. The experimental diets were formulated to be isonitrogenous (220 g kg?1 digestible protein) and isoenergetic (13.4 MJ kg?1 digestible energy) with 0, 25, 50, 75, and 100 % replacement of SM protein by CGM protein over a 60-day period. One hundred fish (32.79 ± 3.43 g) were randomly distributed in 20 experimental cages (70 L) placed within five (1000 L) aquaria with continuous water renewal. The experimental design was completely randomised with five treatments and four replicates. A quadratic effect (P < 0.05) was observed for fillet and carcass yields, feed conversion ratio, weight gain, and specific growth rate, with the optimum values for replacement of SM protein by CGM protein estimated as 21.95, 29.13, 30.94, 37.71, and 38.75 %, respectively. There were no differences (P > 0.05) for blood parameters, proximal composition, water-holding capacity, pH, lipid oxidation, and a* value of fillets. The L* and b* values of fillets showed differences (P < 0.05). Replacement between 21.95 and 38.75 % of SM protein by CGM protein (5.37 and 9.48 % of inclusion in the diet) improves the growth performance and body yield of pacu juveniles without altering blood parameters or affecting the fillet quality.  相似文献

The phytoplasmas of groups 16SrI (‘Candidatus Phytoplasma asteris’) and 16SrVII (‘Ca. Phytoplasma fraxini’) have been associated with phytoplasma diseases in several urban tree species in Bogotá, Colombia and surrounding areas. The insect vectors responsible for this phytoplasma transmission are unknown. The objectives of this study were to test for the presence of phytoplasmas in leafhopper species (Cicadellidae) collected in areas with diseased trees and to determine the phytoplasma transmission ability of two of these species. Leafhoppers of nine species were collected at two sampling sites and tested by nested or double nested PCR using primers for the 16S rRNA gene. The amplicons were subjected to RFLP and/or sequencing analysis. Phytoplasmas of group 16SrI were detected in morphospecies MF05 (Haldorus sp.), group 16SrVII in MF07 (Xestocephalus desertorum), MF08 (Empoasca sp.) and MF09 (Typhlocybinae), and both groups 16SrI and 16SrVII in MF01 (Empoasca sp.), MF02 (Typhlocybinae), MF03 (Scaphytopius sp.), MF04 (Amplicephalus funzaensis) and MF06 (Exitianus atratus). Transmission tests to uninfected bean plants (Phaseolus vulgaris) were performed using field collected A. funzaensis and E. atratus individuals in separate assays. After 5 weeks, the test plants exposed to individuals of both species of leafhoppers showed symptoms, suggesting phytoplasma infection. Phytoplasma groups 16SrI and 16SrVII were detected in the two groups of exposed plants, indicating that A. funzaensis and E. atratus were able to transmit both groups of phytoplasmas. This is the first report of insect vectors for phytoplasmas of group 16SrVII in the world and of 16SrI in South America.  相似文献
