The objectives of the present study were to investigate the relationship between the morphological status of cumulus cells surrounding canine oocytes after maturation culture and the meiotic stage of the oocytes. In addition, the effect of the removal of cumulus cells from canine cumulus-oocyte complexes (COCs) during maturation culture on their meiotic competence was examined. Canine COCs were collected from bitches at the anoestrous and dioestrous stages and only COCs with >110 microm in vitelline diameter were cultured in medium 199 with 10% canine serum for 72 h. In the first experiment, the relation between the morphological status of cumulus cells surrounding oocytes cultured for 72 h and their meiotic stages was examined. At the end of maturation culture, the proportions of intact, partially nude and completely nude oocytes were 65.2%, 22.9% and 11.9%, respectively. The proportion of maturation to metaphase II of completely nude oocytes was highest among the oocytes with different morphological status of cumulus cells. In the second experiment, the cumulus cells were partially or completely removed from COCs at 48 h after the start of maturation culture and the oocytes were cultured for a further 24 h. The proportion of oocytes reaching metaphase II in the completely denuded oocytes was significantly higher than that in the control oocytes without the removal treatment of cumulus cells. The results indicate that morphological status of cumulus cells surrounding oocytes may be related to the nuclear maturation of canine oocytes, and the removal of cumulus cells from COCs during maturation culture can promote the completion of oocyte meiotic maturation. 相似文献

Mammalian sperm undergo a series of biochemical transformations in the female reproductive tract that are collectively known as capacitation. Cyclodextrins added to the sperm culture medium have been described to induce in vitro sperm capacitation, enabling its use in protein‐free media. However, the additive capacitating effect of methyl‐β‐cyclodextrin (MβCD) in the medium containing bovine serum albumin (BSA) is unknown in the bovine species. In this study, we evaluated the effects of incubating frozen–thawed bovine spermatozoa in a BSA‐containing medium supplemented with MβCD on different sperm quality and functional parameters. Sperm viability decreased with the addition of MβCD in a dose‐dependent manner (p < 0.05), and DNA damage could be observed but only with the highest concentration of MβCD. However, pre‐incubation of spermatozoa in MβCD‐supplemented medium improved the capacitation status as assessed by the increase in plasma membrane fluidity, intracellular calcium concentration, induced acrosome reactivity and zona pellucida (ZP)‐binding ability (p < 0.05). Thus, we conclude that MβCD supplementation is able to enhance the capacitation status of frozen–thawed bovine spermatozoa cultured in capacitation medium containing BSA and could result in a valid strategy for its application on artificial reproductive technologies such as in vitro fertilization or intracytoplasmic sperm injection. 相似文献

The recombination dynamics of a transition metal redox system monitored by femtosecond pump-probe spectroscopy are shown to be sensitive to high magnetic fields at times shorter than 10 picoseconds. The effect, based on coherent population beats of different spin states, is quantitatively accounted for and allows direct access to spin relaxation rates far beyond the time resolution of the fastest electron paramagnetic resonance technique. The presence of this ultrafast magnetic field effect helps in understanding complex reaction schemes in transition metal chemistry, which occur in a wide range of fields, such as bioinorganic chemistry and catalysis. 相似文献

ABSTRACT Eighty-eight isolates of Fusarium oxysporum f. sp. niveum, collected from wilted watermelon plants and infested soil in Maryland and Dela-ware, were characterized by cross pathogenicity to muskmelon, race, and vegetative compatibility. Four isolates (4.5%) were moderately pathogenic to >/=2 of 18 muskmelon cultivars in a greenhouse test, and one representative isolate also was slightly pathogenic in field microplots. The four isolates all were designated as race 2, and were in vegetative compatibility group (VCG) 0082. Of the 74 isolates to which a VCG could be assigned, 41 were in VCG 0080, the VCG distributed most widely; 27 were in VCG 0082, and were distributed in half of the 20 watermelon fields surveyed; and 6 were in the newly described VCG 0083, and were restricted to three fields. Among the isolates in VCG 0080, 8 were designated as race 0, 21 as race 1, and 12 as race 2. Of the isolates in VCG 0082, 6 were designated as race 0, 11 as race 1, and 10 as race 2. All isolates in VCG 0083 were designated as race 2. Isolates from more than one race within the same VCG or isolates from more than one VCG were recovered from single plants and fields. No differences in aggressiveness on differential watermelon cultivars were observed among isolates from different VCGs of the same race. A diverse association between virulence and VCG throughout the Mid-Atlantic region suggests that the pathotypes of F. oxysporum f. sp. niveum may be of local origin or at least long existent in the region. 相似文献

An experiment was conducted to test the hypothesis that the supplementation of crude protein (CP) results in rumen acidosis and increased histamine concentrations in dairy cows. Six ruminally fistulated, non-pregnant dry cows were fed three experimental rations in a replicated 3 × 3 Latin square design. The CP contents in the low-CP, the high rumen undegradable protein (high-RUP) and the high rumen degradable protein (high-RDP) rations were 112, 259 and 266 g/kg dry matter (DM) respectively. The cows were fed 7.7 kg DM of the concentrates and 2.7 kg DM of rice straw. High levels of RDP in the ration significantly increased the ammonia, total volatile fatty acid (VFA) and histamine concentrations in the rumen fluid. However, supplemental CP, whether degradable or undegradable, did not significantly affect the pH of rumen fluid. Plasma urea nitrogen concentration was higher in both high-RDP and high-RUP in relation to cows fed the low-CP ration. The rise in ruminal histamine concentrations was physiologically non-relevant, most likely because rumen pH was not affected by supplemental CP at the installed level of DM intake. Therefore, it can be concluded that the issue of supplemental CP, rumen pH and ruminal histamine concentrations has not yet been settled. South American camelids show high embryo loss rate, during the first 60 days of pregnancy. One of the factors which may be related to this situation is that over 98% of the embryos implant in the left uterine horn (LUH) even though both ovaries contribute similarly to ovulation. There is scarce information about the uterine environment of female camelids at any physiological state that could explain the capability of the LUH to attract the embryo and maintain pregnancy. We describe, for the first time, the biochemical and protein profile of uterine fluid (UF), addressing the right and LUH environment in non‐pregnant and pregnant alpacas. Different substrates, electrolytes and metabolites were assayed in both uterine horn fluids. Small changes were observed in glucose and total protein levels, which were more noticeable during pregnancy. In addition, 10 specific proteins were found in the left horn fluid in 5‐week‐pregnant alpacas, and two protein bands were identified in non‐pregnant alpaca right horn fluid. These results would provide basic information for identification of possible markers for pregnancy diagnosis, reproductive diseases and hormone‐treated animals evaluation and hence contributing to improve the pregnancy rate. 相似文献
