Measurement of urinary metanephrines in spot samples is used for the diagnosis of canine pheochromocytoma (PC). We describe a simple analytical method based on liquid chromatography tandem mass spectrometry (LC-MS/MS) for measuring free metanephrine (MN) and normetanephrine (NMN) in spot urine samples. Using the developed method, we evaluated the stability of urinary free-MN and free-NMN at various storing conditions. In addition, we assessed the feasibility of urinary free-MN and -NMN measurement for diagnosing PC. Urine samples were mixed with stable isotope internal standards and thereafter purified by ultrafiltration. The purified samples were analyzed by LC-MS/MS in the multiple reaction monitoring mode after separation on a multimode octa decyl silyl column. The coefficient of variation of free-MN and -NMN measurement was 7.6% and 5.5%, respectively. The linearity range was 0.5–10 µg/l for both analytes. Degradation was less than 10% for both analytes under any of the storage conditions. The median free-NMN ratio to creatinine of 9 PC dogs (595, range 144–47,961) was significantly higher (P<0.05) than that of 13 dogs with hypercortisolism (125, range 52–224) or 15 healthy dogs (85, range 50–117). The developed method is simple and may not require acidification of spot urine. The results of this preliminary retrospective study suggest that the measurement of urinary free metanephrines is a promising tool for diagnosing canine PC.  相似文献

This study was conducted to evaluate theeffect of plant proteinbased diets on gonadaldevelopment and plasma 17 -estradiol (E2) levelin female Nile tilapia Oreochromis niloticus.Fish with a mean body weight of 6.7 (0.1) g were fedfour different diets with the same digestible protein(DP) and digestible energy (DE) containing gradedlevels of a mixture of plant ingredients as partial ortotal replacement of fish meal protein for 20 weeks.The control diet (D0) was based on fish meal, twodiets containing 33% (D33) and 66% (D66) of plantprotein, and one diet containing only plant protein(D100). Fish were sampled at 12 and 20 weeks. Nosignificant differences were found in different stagesof oocyte development and plasma E2 levels betweentilapia fed diets D0 and D100 at 12 weeks. Eight weekslater tilapia fed diet D0 showed a higher (P < 0.05)level of E2 than the D100 group. This difference andthe reduced proportion of vitellogenic and matureoocytes demonstrated that diets containing only plantprotein are less efficient in terms of tilapia growthand consequently ovarian development.  相似文献

Throughout the Amazon of Brazil, manioc (Manihot esculenta) is a staple crop produced through slash-and-burn agriculture. Nutrient losses during slash-and-burn can be large and nutrient demand by food crops so great that fields are often abandoned after two years. In recent decades, farmers have reduced the fallow phase from 20 to ~5 years, limiting plant nutrient accumulation to sustain crop yields. Improved fallows through simultaneous planting of trees with food crops may accelerate nutrient re-accumulation. In addition, slash-and-mulch technology may prevent loss of nutrients due to burning and mulch decomposition may serve as a slow-release source of nutrients. This study in Pará, Brazil, in a 7-year-old secondary forest following slashing and mulching of the vegetation, involved two main plot treatments (with and without P and K fertilizers) and two sub-plot treatments (with or without a N2-fixer Inga edulis). A mixed-culture of trees and manioc was planted in all plots. P and K fertilizer increased tree mortality due to weed competition but growth of surviving trees in four of the five tree species tested also increased as did biomass production of manioc. In the N2-fixer treatment trends of greater growth and survival of four of five tree species and manioc biomass were also observed. Fertilization increased the biomass of competing vegetation, but there was a fertilizer by N2-fixer interaction as I. edulis caused a reduction in competing biomass in the fertilized treatment. After one year, fertilization increased decomposition of the mulch such that Ca, Mg, and N contents within the mulch all decreased. In contrast, P and K contents of mulch increased in all treatments. No influence of the N2-fixer on 0–10 cm soil N contents was observed. Two years after establishment, this agroforestry system succeeded in growing a manioc crop and leaving a well-maintained tree fallow after the crop harvest.  相似文献

The inefficiency of conservation efforts based exclusively on natural habitat patches has called the attention of some conservationists to the matrix. Described as the major component of a landscape, the matrix is often agricultural, particularly in the tropics. In this context, agroforestry practices have been recognized for their ability to support a rich fauna and flora. Besides the extensive literature concerning bird communities in agroforestry systems, very few studies analyze how different species respond to the management of such practices. Our study describes the diet and habitat use frequency of five frugivorous bird species in agroforest home gardens, secondary forests, and pastures in the region of Pontal do Paranapanema, Brazil. The focal species were Ramphastos toco (Toco Toucan), Pteroglossus castanotis (Chestnut-eared Aracari), Amazona aestiva (Blue-fronted Amazon), Ara chloroptera (Green-winged Macaw), and Cyanocorax chrysops (Plush-crested Jay). We gathered both habitat use frequency and diet using the “feeding-bout” method. Overall frequency was higher in the secondary forest when compared to pasture and home gardens for all bird species except A. aestiva. The number of feeding bouts was higher in home gardens than in forests for all species with the exception of C. chrysops. Differences in monthly median feeding activity were only statistically significant for C. chrysops and for A. aestiva. The latter was the only species observed feeding in pasture habitats. The total number of food taxa was larger in home gardens than in the forest. Our results reinforce the importance of agroforestry systems as a resource-rich habitat for frugivorous birds.  相似文献

Relationships between the chemical composition of the 9th- to 11th-rib section and the chemical composition of the carcass and empty body were evaluated for Bos indicus (108 Nellore and 36 Guzerah; GuS) and tropically adapted Bos taurus (56 Caracu; CaS) bulls, averaging 20 to 24 mo of age at slaughter. Nellore cattle were represented by 56 animals from the selected herd (NeS) and 52 animals from the control herd (NeC). The CaS and GuS bulls were from selected herds. Selected herds were based on 20 yr of selection for postweaning BW. Carcass composition was obtained after grinding, homogenizing, sampling, and analyzing soft tissue and bones. Similarly, empty body composition was obtained after grinding, homogenizing, sampling, analyzing, and combining blood, hide, head + feet, viscera, and carcass. Bulls were separated into 2 groups. Group 1 was composed of 36 NeS, 36 NeC, 36 CaS, and 36 GuS bulls and had water, ether extract (EE), protein, and ash chemically determined in the 9th- to 11th-rib section and in the carcass. Group 2 was composed of 20 NeS, 16 NeC, and 20 CaS bulls and water, EE, protein, and ash were determined in the 9th- to 11th-rib section, carcass, and empty body. Linear regressions were developed between the carcass and the 9th- to 11th-rib section compositions for group 1 and between carcass and empty body compositions for group 2. The 9th- to 11th-rib section percentages of water (RWt) and EE (RF) predicted the percentages of carcass water (CWt) and carcass fat (CF) with high precision: CWt, % = 29.0806 + 0.4873 × RWt, % (r(2) = 0.813, SE = 1.06) and CF, % = 10.4037 + 0.5179 × RF, % (r(2) = 0.863, SE = 1.26), respectively. Linear regressions between percentage of CWt and CF and empty body water (EBWt) and empty body fat (EBF) were also predicted with high precision: EBWt, % = -9.6821 + 1.1626 × CWt, % (r(2) = 0.878, SE = 1.43) and EBF, % = 0.3739 + 1.0386 × CF, % (r(2) = 0.982, SE = 0.65), respectively. Chemical composition of the 9th- to 11th-rib section precisely estimated carcass percentages of water and EE. These regressions can accurately predict carcass and empty body compositions for Nellore, Guzerah, and Caracu breeds.  相似文献

Bovine leukemia virus (BLV) is among the most widespread livestock pathogens in many countries. Despite advances in understanding the pathogenesis of this disease, little is known about the involvement of oxidative stress. Therefore, this study examined the antioxidant status and the markers of oxidative stress in BLV-infected dairy cows. BLV infection was associated with an increase in triacylglycerol levels, a decrease in glutathione peroxidase (GSH-Px) activity and a tendency toward lower superoxide dismutase activity in the infected animals. No significant difference was observed in other markers of oxidative stress (i.e., conjugated dienes, hydroperoxides and malondialdehyde) in the infected animals compared to controls. A novel method for the analysis of oxidative stress, Z-scan based on the measurement of the mean-value of θ in low density lipoprotein indicated that the infected animals had low-density lipoprotein particles that were slightly less modified than those from the healthy group. Thus, we conclude that BLV infection is associated with a selective decrease in GSH-Px activity without any alteration in the common plasma markers of oxidative stress.  相似文献

In 2008, in the west zone of Rio de Janeiro municipality-Brazil, the leishmaniasis control program identified 155 dogs with titers ≥ 40 by Indirect ImmunoFluorescence (IIF) on blood collected onto filter paper. The objective of this study was to describe the laboratory test findings performed in dogs euthanized by the leishmaniasis program control of Rio de Janeiro municipality. Dogs were examined, subjected to euthanasia and collection of clinical specimens. Parasite isolation was obtained in 29 animals: Leishmania chagasi was isolated in 14 dogs; Leishmania braziliensis was isolated in five dogs; Trypanosoma caninum was obtained in seven animals and one dog had mixed infection (L. braziliensis and L. chagasi). By Polymerase Chain Reaction, seventeen animals were positive in intact skin fragments. In the serological reassessment of serum samples, 28% and 22% were positive for IIF and enzyme immunoassay, respectively. Ninety-one (59%) dogs were negative for all tests performed in this study. The findings indicate that the visceral leishmaniasis control program needs to be adjusted in order to avoid non-infected dogs from being removed or permit that dogs infected with L. chagasi to remain undetected in endemic areas.  相似文献

The aim of this study was to test the functional status of uterine oxytocin receptors in cows in vivo around parturition. The animals received consecutive, intra‐arterial injections of 800, 1600 and 3200 mU of oxytocin at three different stages: during late gestation (days 260–274), at 12 h and at 24 h after intramuscular injection of a prostaglandin F analogue at day 275 to induce parturition. Cows (n=6) had been provided with myometrial electrodes and a catheter had been installed in the aorta and in a branch of the uterine vein (UV). Regular blood samples were obtained from the UV from 5 min before until 45 min after each oxytocin injection to measure plasma levels of prostaglandin F (PGF) and oxytocin. Uterine electromyographic (EMG) activity was registered continuously during each experiment. The increase of oxytocin levels in UV plasma after intra‐arterial injections was dose dependent (p < 0.02). Pre‐ and post treatment oxytocin levels at 24 h after induction of parturition were significantly increased (p=0.0313). Both during late pregnancy and at 12 h after induction of parturition, oxytocin caused a significant increase in EMG activity (p=0.022). After the 3200 mU dose the increase was significantly higher than with the other 2 doses (p=0.004). After each dose, EMG activity returned to baseline levels within some 15 min. At 24 h after induction of parturition, the pre‐treatment level of EMG activity had increased. Doses of 800 mU and 1600 mU of oxytocin produced a significant (p=0.022) increment of EMG activity, which was of the same magnitude as during the preceding stages; after 3200 mU of oxytocin the response was significantly higher than before (p=0.008). No significant increases of PGF levels in UV plasma could be measured after oxytocin injections at any of the three stages. It is concluded that the myometrium of the pregnant cow responds in vivo to physiological doses of oxytocin. At 24 h after induction of parturition, when luteolysis has occurred and a parturient pattern of parturient myometrial activity has already started to develop, the response is enhanced. Physiological doses of oxytocin did not evoke a spurt release of PGF in uterine venous blood during the peripartal period.  相似文献
