Vascular endothelial growth factor (VEGF) is a major regulator of angiogenesis and a potential autocrine growth factor for neoplastic cells in various malignancies. In the present study, we have investigated expression of VEGF and VEGF receptors in canine mastocytomas and the canine mastocytoma cell line C2. As assessed by immunostaining of tissue sections and cytospin slides, primary neoplastic mast cells (MC) and C2 cells were found to express the VEGF protein. In Northern blot and RT-PCR experiments, C2 cells expressed VEGF mRNA in a constitutive manner. VEGF mRNA expression in C2 cells was counteracted by LY294002 and rapamycin, suggesting involvement of the PI3-kinase/mTOR pathway. Moreover, C2 cells were found to express VEGF receptor-1 (Flt-1) and VEGF receptor-2 (KDR). However, recombinant VEGF failed to promote (3)H-thymidine uptake in C2 cells, and a neutralizing anti-VEGF antibody (bevacizumab) failed to downregulate spontaneous proliferation in these cells. In addition, rapamycin decreased the expression of VEGF in C2 cells at the mRNA and protein level without suppressing their proliferation. Together, canine mastocytoma cells express VEGF as well as VEGF receptors. However, despite co-expression of VEGF and its receptors, VEGF is not utilized as an autocrine growth regulator by canine mastocytoma cells.  相似文献

Spatio-temporal heterogeneity in physico-chemical conditions associated with the annual expansion/contraction cycle in a complex glacial flood plain of the Swiss Alps was investigated employing a landscape approach. The diverse and dynamic aquatic habitats of the flood plain were visualized as an aquatic mosaic or riverscape. Based on samples collected at ca. monthly intervals for 1.5 yr along 17 floodplain transects, the 3 components of riverscape heterogeneity, extent, composition, and configuration, were quantified using categorical maps and indices of landscape patterns for turbidity and specific conductance. Changes in the spatial heterogeneity of 13 other physico-chemical parameters were further analyzed by means of a within-dates principal component analysis. Riverscape heterogeneity (RH), quantified by applying several indices of landscape pattern to turbidity and specific conductance data, was minimum during groundwater-dominated base flow in winter. Despite an increase in surface connectivity in the channel network with rising discharge, RH rose in spring and summer as additional chemically-distinct water sources (i.e., snowmelt runoff and glacial ablation) contributed to surface flow within the flood plain. Most other physico-chemical variables measured during this study exhibited the same spatio-temporal heterogeneity as turbidity and specific conductance. Overall, the glacial flood plain shifted from a monotonous physico-chemical riverscape in winter to a complex mosaic in summer, this seasonal pattern being clearly driven by hydrological factors operating at the catchment scale rather than by autogenic processes within individual water bodies. Although RH exhibited a predictable annual pattern in response to the seasonal flow regime, we expect the channel network to undergo future modifications from stochastic factors associated with flood events and long-term changes reflecting movements of the glaciers.  相似文献

Phosphorus availability in terrestrial ecosystems is strongly dependent on soil P speciation. Here we present information on the P speciation of 10 forest soils in Germany developed from different parent materials as assessed by combined wet‐chemical P fractionation and synchrotron‐based X‐ray absorption near‐edge structure (XANES) spectroscopy. Soil P speciation showed clear differences among different parent materials and changed systematically with soil depth. In soils formed from silicate bedrock or loess, Fe‐bound P species (FePO4, organic and inorganic phosphate adsorbed to Fe oxyhydroxides) and Al‐bound P species (AlPO4, organic and inorganic phosphate adsorbed to Al oxyhydroxides, Al‐saturated clay minerals and Al‐saturated soil organic matter) were most dominant. In contrast, the P speciation of soils formed from calcareous bedrock was dominated (40–70% of total P) by Ca‐bound organic P, which most likely primarily is inositol hexakisphosphate (IHP) precipitated as Ca3‐IHP. The second largest portion of total P in all calcareous soils was organic P not bound to Ca, Al, or Fe. The relevance of this P form decreased with soil depth. Additionally, apatite (relevance increasing with depth) and Al‐bound P were present. The most relevant soil properties governing the P speciation of the investigated soils were soil stocks of Fe oxyhydroxides, organic matter, and carbonate. Different types of P speciation in soils on silicate and calcareous parent material suggest different ecosystem P nutrition strategies and biogeochemical P cycling patterns in the respective ecosystems. Our study demonstrates that combined wet‐chemical soil P fractionation and synchrotron‐based XANES spectroscopy provides substantial novel information on the P speciation of forest soils.  相似文献

Management-oriented models of cattle habitat use often treat grazing pressure as a single variable summarizing all cattle activities. This paper addresses the following questions: How does the spatial pattern of cattle effects vary between cattle activities in a highly heterogeneous landscape? Do these patterns change over the grazing season as forage availability decreases? What are the respective roles of natural and management-introduced structures? We estimated the intensity of herbage removal, dung deposition and trampling after each of three grazing periods on a grid of 25 m ×25 m cells covering an entire paddock in the Swiss Jura Mountains. We found no significant positive correlations between cattle effects. Spatial patterns weakened through the season for grazing and trampling, whereas dunging patterns changed little between grazing periods. Redundancy analysis showed that different cattle effects were correlated with different environmental variables and that the importance of management-introduced variables was highest for herbage removal. Autocorrelograms and partial redundancy analyses using principal coordinates of neighbour matrices suggested that dunging patterns were more coarse-grained than the others. Systematic differences in the spatial and seasonal patterns of cattle effects may result in complex interactions with vegetation involving feedback effects through nutrient shift, with strong implications for ecosystem management. In heterogeneous environments, such as pasture-woodland landscapes, spatially explicit models of vegetation dynamics need to model cattle effects separately.  相似文献

Landraces are thought to be well adapted to the environmental conditions of the region where they have evolved and to be highly tolerant to biotic and abiotic stress. There is indeed experimental evidence of that for several cereal species, but to our knowledge only scarce data is available on rye and none on rye landraces from South Tyrol in particular. For this region, assumptions rely mainly on historic documents or reports from the owners of the landraces. In a 3-year field experiment we described 13 landraces from Val Venosta/Vinschgau (South Tyrol, Italy), along with six modern population cultivars (CUL), at a mountain location. Depending on the available passport data, the landraces were assigned to two variety types: well documented landrace status (DLS) and uncertain landrace status (ULS). 14 traits concerning phenology, agronomy, indirect milling and baking quality as well as seed colour were investigated. Most of the traits representing the usual target of modern breeding were found to be dependent on the variety type. The results reflect well the expected differences between CUL and DLS, while ULS had an intermediate character. Winter hardiness and ripening time of both DLS and ULS, however, were found to be affected by the altitude of the site at which the landraces were collected, suggesting a certain adaptation of the landraces to the climatic conditions at the sites of origin.  相似文献

APC-germline mutation creates predisposition for intestinal tumorigenesis. APCMin/+ mice, developing tumors preferentially in the small intestine and only minimally in the colon, were fed pectin-enriched diets (10% galacturonan; degree of methoxylation=37.0 and 70.4%) or standard diet. Pectins used in the present study do not inhibit intestinal tumorigenesis and rather accelerate it in APCMin/+ mice. Both pectins exhibited prebiotic effects associated with high fermentative formation of acetate but producing low butyrate. The differences of the short-chain fatty acid concentrations between cecum and colon and those between colon and feces were larger than expected and increased with cancer progression, indicating an inhibition of butyrate absorption. Pectins transported more bile acids toward the colon than the standard diet and caused a higher generation of secondary bile acids despite lower pH values. Overexpression of COX-2 resulted in lower antioxidative capacity, thus promoting cancer. Apoptosis increased in hyperplasia but decreased in late adenomas. When biological modular design principles are taken into consideration, it can be expected that pectin also reinforces colorectal tumorigenesis of patients suffering from APC gene defects.  相似文献

The effects of germination time and temperature on the quality of rye malt and worts derived thereof were investigated using Response Surface Methodology. Amylolytic and proteolytic enzyme activities were increased by long germination periods, while β-glucanase activity was not influenced. Total and Soluble Nitrogen content were also not significantly affected by the variations in germination conditions. Free Amino Nitrogen (FAN) was found in higher amounts in worts prepared from rye malts with long germination times. Extract contents were higher in rye malt than in the control barley malt and could be increased by a favourable germination regime, while no such impact on wort fermentability was found. High wort viscosities could be significantly reduced by a long germination period at low temperatures, but were still unacceptably high. The same conditions favoured the development of endoxylanase activity. Arabinoxylan (AX) accumulated during the germination process and their extractability increased. The results suggest that longer germination periods resulted in an increased number of AX molecules with lower molecular mass. Optimal rye malt qualities within the limits of this study were found for a germination time of 144 h at 10 °C, which resulted in an acceptable FAN content and the lowest measured viscosity.  相似文献

Since 2000, the Chinese government has implemented emergency water diversion measures to restore the damaged riparian forest ecosystem with dominant tree species Euphrat poplar(Populus euphratica Oliv.)at the lower reaches of the Tarim River. In the present study, comparative analysis of variations in the vitality of P. euphratica trees were made using 2005 and 2010 data to illustrate the revitalization process of riparian forest. Poplar trees within 300 m of the riverbed were positively revitalized, while the vitality of trees farther than 300 m from the river decreased. Population structure was studied to demonstrate the development of poplar community. In the first belt, the class structure for the diameter at breast height(DBH) of P. euphratica fit a logistic model, and the 2nd, 3rd and 4th belt curve fittings were close to a Gaussian model; in other plots they were bimodal. Cluster analysis of the composition of the DBH class of poplar trees demonstrated that those within 16–36 cm DBH were the most abundant(58.49% of total) in study area, under 16 cm of DBH were second(31.36%), and trees 40 cm DBH were the least abundant(10.15%). More than 80% of the trees were young and medium-sized, which means that the poplar forest community in the vicinity of the lower Tarim River is at a stable developmental stage. The abundance of juvenile trees of P. euphratica in the first and second measuring belts was 12.13% in 2005 and increased to 25.52% in 2010, which means that the emergency water transfer had a positive impact on the generation of young P. euphratica trees in the vicinity of the river.  相似文献

Barley yellow dwarf (BYD) is one of the main viral diseases of small-grain cereals. This disease, reported on numerous plant species of the Poaceae family, is caused by a complex of eight viral species including the species Barley Yellow Dwarf Virus-PAV (BYDV-PAV), frequently found in western Europe. Resistance sources against BYDV-PAV are scarce and only identified in perennial Triticineae. Some BYDV-resistant wheat lines have been obtained by introgressing these resistances into bread wheat germplasms. Genetic and biological characterization of the resulting lines has been undertaken. However, little information on the resistant behaviour of these lines during the early stages of the infection process is available. To evaluate the resistance of two genetically distinct resistant lines (Zhong ZH and TC14), 1740 young plantlets, belonging to susceptible reference hosts (barley cv. Express and wheat cv. Sunstar), Zhong ZH or TC14 wheat lines, were inoculated in controlled conditions with French BYDV-PAV isolates. The infection process was monitored during the first 21 days after inoculation (DAI) using a semi-quantitative ELISA. A standardized protocol including five successive samplings of leaves from all inoculated plants and the collection of plant roots at the end of the monitored period was carried out. This protocol enabled an assessment of the infection percentage and the evolution of the viral load in plants from the 7th DAI to the 21st DAI. Statistical analyses of the BYDV infection kinetics using raw ELISA data, a model of the time-dependent variation of the percentage of infected plants and the area under concentration progress curves (AUCPC) demonstrated that Zhong ZH and TC14 lines (1) reduce the development rate of the BYD disease during the first days of infection, (2) decrease the infection efficiency of BYDV-PAV isolates, in the leaves, from 98.7% for susceptible plant genotypes to 81.9% and 71.7% for Zhong ZH and TC14, respectively, (3) reduce the virus load in the leaves of infected plants and (4) are not spared from BYDV infection, as 95.1% of Zhong ZH and 90.2% of TC14 inoculated plants accumulated viral particles in roots and/or in leaves at 21 DAI. These results confirm the BYDV-partial resistant behaviour of both Zhong ZH and TC14 lines. The development rate of the disease was the single parameter that allowed the distinction between the two resistant sources present in the tested lines.  相似文献
