AIM: To discover specific neural stem cell (NSC) proliferation-promoting factor, which will contribute to study on development of nervous system and treatment of nervous system diseases. METHODS: The extracts of forebrain, midbrain,afterbrain and cerebellum of neonatal mice were prepared, and the NSCs of newborn mice were cultured in vitro. Neurospheres were observed, immunocytochemical staining of characteristic protein, nestin, and MTT assay were performed to identify NSCs and their proliferative properties. RESULTS: A great deal of neurospheres were formed in the presence of the extracts of afterbrain and cerebellum, which were positive for characteristic protein (Nestin) of NSC showed by immunocytochemical staining. CONCLUSION: The afterbrain and cerebellum extracts can increase the total number of NSCs isolated from newborn mice in vitro in a dose-dependent manner.  相似文献

AIM: To investigate the characterization of absorption of hematoporphyrin monomerthyl ether (HMME), a domestic new generation photosensitizer product, by activated T cells from human peripheral blood. METHODS: Evaluation was performed by flow cytometry on the effects of incubating concentration and time of HMME on absorption by activated T cells. Lymphocytes were separated from human peripheral blood by density gradient centrifugation with Ficoll and T cells were activated with polyclonal stimulators PHA and PDB+Ion. To analyze the effects of HMME incubating doses on the absorption of activated T cells, the cultural lymphocytes were incubated with a serial doses of HMME for 1 h and HMME absorption were measured by FACS after immuno-staining with anti-CD3 antibody. To test the impact of HMME incubating time on the absorption of activated T cells, the cultural lymphocytes were incubated with HMME for various times and HMME absorption were measured by FACS after immuno-staining with anti-CD3 antibody. RESULTS: The HMME absorption-dose curve and absorption-time curve were shifted to right and up in the activated T cells as compared to resting T cells. HMME absorptions of activated T cells were statistic significantly larger than that of resting T cells in the doses between 5 mg/L to 20 mg/L. HMME absorptions of either activated T cells or resting T cells underwent a gradual increase with the incubation-time in HMME at concentration of 10 mg/L. HMME absorptions of activated T cells were statistic significantly larger than that of resting T cells in the incubation-time between 15 to 60 min. CONCLUSION: The differences of HMME absorption between activated T cells and resting T cells depend on the incubation times and doses of HMME. HMME absorption of activated T cells are significantly larger than that of resting T cells in certain incubation-times and doses. These results suggest that incubation time and dose associated with HMME-PDT therapeutic windows will be created for selective deletion of activated T cells.  相似文献

AIM: To evaluate the immunogenicity of a novel orthopedics materials (graded zirconia-hydroxyapatite composite) in vitro by using peripheral blood mononuclear cells (PBMCs) from healthy young people, and simple zirconia-hydroxyapatite composited material was used as control materials. METHODS: Proliferation of PBMCs cultured in different liquid after 5 days was measured by MTT methods. ELISA was used to detect TNF-α and IL-6 concentration in the supernatant of PBMCs cultured in the extracts after 24 hours. Flow cytometery was used to measure CD69 and CD25 in activated PBMCs cultured in the extracts of the two kinds of materials after 24 hours. RESULTS: The proliferation rate of the simple composite group was significantly lower than that in negative group (P<0.05), but there was no difference between the graded composited group and negative group. After 24 hours culture with LPS, the concentrations of TNF-α and IL-6 in the simple composited group were significantly higher than that in graded composited group, respectively (P<0.05). Cultured with PHA for 24 hours, the ratio of CD69 and CD25 positive PBMCs in the simple composited group was all significantly higher than that in graded composited material group (P<0.01). CONCLUSION: The number of PBMCs activated by the graded composited material is less than the simple composited material and the immunogenicity of the graded composited material is lower than the simple composited material.  相似文献

AIM: To study the level of serum cystatin C (Cys C) and its prognostic value in acute renal failure (ARF). METHODS: 215 critically ill patients were studied prospectively, and each blood sample was determined daily. Serum creatinine (Scr) was measured by enzymic method. Cys C was detected by particle-enhanced turbidimetric immunoassay (PETIA) and glomerular filtration rate (GFR) was estimated by Cockroft-Gault equation. ARF was diagnosed according to ADQI (RIFLE) criteria. RESULTS: 41 patients developed ARF to some degree, among which 7, 13 and 21 patients fulfilled R-, I-, and FLE-criteria, respectively. 21 were death and 20 survival in total 41 ARF patients. 174 patients without ARF served as controls. In patients with ARF, Cys C was significantly increased than that in patients who did not develop ARF (P<0.01). In different RIFLE criteria of ARF, serum Cys C was linearly correlated with serum creatinine (P<0.01), also [Cys C]-1 with estimated GFR (P<0.01). When ARF was diagnosed according to serum creatinine increased ≥150%, serum Cys C was demonstrated a highly diagnostic value to detect ARF as indicated by area under the curve (AUC) of the ROC curve of 0.983 (95% confidence interval, 0.960-1.006) (P<0.01). Logistic regression showed serum Cys C and Scr were not independently risk factors for the mortality of ARF (P>0.05). Also ROC analysis demonstrated serum Cys C and Scr had no predicting value of the fatality and survival in ARF (P>0.05). CONCLUSION: Serum Cys C is significantly increased in ARF and correlated well with the severity of ARF. Serum Cys C can be one of the detectable markers of ARF, but it is independent of the mortality and does not predict the prognosis of these patients.  相似文献

 采用抗原直接包被和双抗体夹心酶联免疫吸附测定法(ELISA)对采自云南、福建、湖南烟区烟草花叶病样品进行了病毒种类检测,利用三抗体夹心ELISA对黄瓜花叶病毒(Cucumber mosaic virus,CMV)的亚组类型进行了鉴定。在云南采集的520个花叶病样品中,烟草花叶病毒(Tobacco mosaic virus,TMV)、CMV和马铃薯Y病毒(Potato virus Y,PVY)总检出率分别为71.74%、55.01%和6.35%;在福建采集的150个花叶病样品中,TMV、CMV和PVY的总检出率分别为94%、24.66%和8.00%;在湖南采集的74个花叶病样品中,TMV、CMV和PVY的总检出率分别为58.11%、51.35%和2.70%。部分样品为2种以上病毒复合侵染。云南、福建和湖南采集的64个CMV阳性样品中,属亚组Ⅰ的样品为57个,占89.1%;属亚组Ⅱ的样品为10个,占15.6%;其中3个样品为亚组Ⅰ和亚组Ⅱ的复合侵染。  相似文献

本文对青岛市美国白蛾Hyphandria cunez种群的几个重要生物学特性进行了调查,获得了如下有意义的结果。(1)在生态光周期范围内,该虫显示了一个典型的长日照型昆虫,临界日长为14h 30min。然而在全光条件下,有41.2%的个体滞育;在全暗条件下则有83.7%的个体发育。(2)第1代蛹在自然条件下羽化的结果表明,部分个体的蛹期显著延长,呈现了夏季滞育。(3)滞育蛹置于25℃,LD16:8下滞育能够解除,但死亡率极高。(4)雄虫前翅黑斑的出现是由光周期控制的,只有在短日照下诱导的滞育蛹,滞育解除后羽化出来的雄虫才会出现黑斑。  相似文献

根据已报道的烟草线条病毒外壳蛋白基因序列,设计出一对特异性引物,提取大豆病叶的总RNA,反转录成cDNA,PCR扩增出约400bp大小的产物,产物克隆到pGEM-T Easy载体上,转化大肠杆菌DH 5α,抽提的质粒用Not Ⅰ酶切,出现430bp大小的目标插入片段。经序列测定确认了插入片段的大小为404bp。  相似文献

杭州地区有蝶类113种,隶属于12科76属。其中古北东洋共有种48种,东洋种42种,古北种17种,广布种5种。区系分析结果表明,杭州地区蝴类以古北东洋共有种和东洋种为主要成份,属东洋区系范畴,并具有由东洋区向古北区过渡的特征。与江、浙其他地区的蝶类资源进行相似度分析,杭州地区与宁波地区的蝶类资源相似度最高。  相似文献

毒死蜱微囊悬浮剂的制备及微囊化条件的优化   总被引:3,自引:4,他引:3  
以脲醛树脂为壁材,采用原位聚合法制备了20%毒死蜱微囊悬浮剂,探讨了影响微囊粒径大小及分布的因素,优化了微囊化条件,并就其与毒死蜱乳油对韭菜迟眼蕈蚊Bradysia odoriphaga幼虫的毒力进行了对比。结果表明,选用苯乙烯-马来酸酐共聚乳液(SMA)为分散剂(质量分数为总体系的2.5%),乳化条件为1 500 r/min搅拌30 min,60 min内调节体系pH值至2.5,反应后期升温至45℃,在此条件下制备的毒死蜱微胶囊外形较好,平均粒径为8.9 μm,分散系数为0.182 6,包封率高达99.54%。毒力测定结果表明,乳油和微胶囊对韭菜迟眼蕈蚊幼虫的初始LC50值分别为2.05和 3.02 μg/mL;而在自然环境中放置32 d后,乳油对该虫的毒力下降较快,LC50值为100.23 μg/mL,而微囊悬浮剂毒力仍保持较高水平,LC50值为15.63 μg/mL。表明毒死蜱微囊悬浮剂兼具速效性高和持效期长的优点。  相似文献
