The mutant prevention concentration (MPC) is a new concept meant to face the increased prevalence of antibiotic resistance by using antibiotic concentrations able to prevent the selection of single-step resistant mutants. In the present study, the MPCs of ciprofloxacin and enrofloxacin were evaluated against fully susceptible strains of Escherichia coli, Salmonella Typhimurium and Pseudomonas aeruginosa. Additionally, representative single-step mutants arising after exposure to sub-MPC antibiotic concentrations were investigated for molecular basis of their fluoroquinolone resistance phenotypes. MPC value was recorded when more than 1010 CFU/mL were spread on Muëller Hinton Agar supplemented with different antibiotic concentrations (from 1X to 16X MIC value). MICs of original strains as well as single-step mutants were determined in presence or absence of the Efflux Pump Inhibitor Phe-Arg-β-naphthylamide (PAβN). Moreover point mutations in the QRDR of the gyrA and parC genes were investigated by sequencing. The enrofloxacin MPC values were 4–16-fold higher than ciprofloxacin values. E. coli and S. Typhimurium representative single-step mutants showed reduced susceptibilities associated with point mutations in the QRDR of the gyrA gene or efflux pump system. P. aeruginosa mutants showed resistance phenotypes associated predominantly with efflux pump system activity. According to in vitro MPC data, ciprofloxacin showed a better efficacy than enrofloxacin, in preventing the selection of E. coli, S. Typhimurium and P. aeruginosa single-step mutants. However, in relation to AUC/MPC ratio, the MPC concept can be applied in vivo to ciprofloxacin and enrofloxacin for E. coli and S. Typhimurium but not for P. aeruginosa.  相似文献

Tree thinning reduces tree-to-tree competition and likely contributes to the improvement of tree water status and productivity in water-limited systems. In this study, we examined the importance of competition for water among Quercus ilex trees in open woodlands by comparing the water consumption and physiological status of trees located along stand density gradients which ranged from 10% (low density; LD) to 100% (high density; HD) of canopy cover. The study was carried out at two sites which differed in mean annual rainfall (506 and 816 L m−2; Dsite and Wsite, respectively). Predawn and midday leaf water potential (ψd and ψm, respectively) and CO2 assimilation rate (A) were measured every two weeks from mid May to mid September, in eight trees located along a stand density gradient at each site. Sap flow and soil moisture were measured only at Dsite. Sap flow was continuously recorded by sap flowmeters (constant heating method) installed in 12 trees along two stand density gradients. Soil moisture (?) was measured every 20 cm for the first meter and then every 50 cm up to 250 cm. Measurements were conducted in 18 soil profiles, 6 located in HD and 12 in LD (six beneath and six out the canopy). At Wsite, differences among stand densities for ψ and A were very small and emerged only at the end of the dry season. At Dsite, ψ (both predawn and midday), A, ?, and sap flow density were significantly higher in LD trees than in HD ones. At Dsite, some water remained unused in the soil at the end of the dry season beyond the canopy in the LD areas, and trees did not experienced such an acute water deficit (ψd > −1 MPa) as the HD trees did (ψd < −3 MPa). Summer tree transpiration at the stand level (Estand) tended to saturate with the increase of canopy cover. Estand increases by 32% when canopy cover goes from 50% to 100%. Results confirmed that the increase of tree-to-tree competition with stand density was much more significant at dry sites. In these sites, tree thinning is recommended as a way to maintain tree functioning.  相似文献

Phakopsora pachyrhizi, the cause of soybean rust, is an economically important pathogen of soybean in South America. Understanding the pathogenicity of indigenous fungal populations is useful for identifying resistant plant genotypes and targeting effective cultivars against certain populations. Fifty-nine rust populations from Argentina, Brazil, and Paraguay were evaluated for pathogenicity in three cropping seasons, 2007/2008–2009/2010, using 16 soybean differentials. Only two pairs of P. pachyrhizi populations displayed identical pathogenicity profiles, indicating substantial pathogenic variation in the rust populations. Comparative analysis of 59 South American and five Japanese samples revealed that pathogenic differences were not only detected within South America but also distinct between the P. pachyrhizi populations from South America and Japan. In addition, seasonal changes in rust pathogenicity were detected during the sampling period. The differentials containing resistance genes (Rpp: resistance to P. p achyrhizi) Rpp1, Rpp2, Rpp3, and Rpp4, except for Plant Introduction (PI) 587880A, displayed a resistant reaction to only 1.8–14, 24–28, 22, and 36 % of South American P. pachyrhizi populations, respectively. In contrast, PI 587880A (Rpp1), Shiranui (Rpp5), and 3 Rpp-unknown differentials (PI 587855, PI 587905, and PI 594767A) showed a resistant reaction to 78–96 % of all populations. This study demonstrated that P. pachyrhizi populations from South America vary geographically and temporally in pathogenicity and that the known Rpp genes other than Rpp1 in PI 587880A and Rpp5 have been less effective against recent pathogen populations in the countries studied.  相似文献

Abstract

A perennial forage legume species tolerant to water stress would be useful to improve pasture and animal productivity in the central zone of Chile. The growth, dry-matter (DM) production, and drought tolerance of twelve accessions of Lotus tenuis Waldst & Kit, Syn. L. glaber, naturalized in Chile were evaluated with the objective to select contrasting genotypes, tolerant and sensitive to drought, for future breeding programmes. The accessions were sown in pots of 1.3 L containing a volcanic soil (Andisol). A completely randomized design with all combinations of Lotus accessions and four water treatments [100, 70, 40, and 10% of soil water availability (SWA)] was used. The relative rate of stem elongation (RRSE), the shoots and roots DM production, the relative water content (RWC), and the specific leaf area (SLA) were measured. A drought-sensitivity index (DSI) and the interaction genotype×environment were calculated. The RRSE, the DM production, RWC, and SLA all showed a significant reduction (P<0.05) in the treatment with the higher water restriction (10% SWA). There were significant differences (P<0.05) within RRSE and DM production genotype means, but the RWC and SLA did not differ among populations. The DSI varied broadly among genotypes, from 0.49 to 1.34, and was correlated negatively with DM production under water stress (10% SWA). It was concluded that the Lotus tenuis populations showed water-stress tolerance genetic variability, with accessions Lt14 and Lt4 the most contrasting. These findings will permit us to identify chromosomal regions associated with drought-tolerant genotypes which will allow us to accelerate the development of cultivars adapted to water-restricted environments.  相似文献

The area occupied by colonies of the black-tailed prairie dog (Cynomys ludovicianus; BTPD henceforth) in northwestern Mexico was dramatically reduced between 1988 and 2005. We conducted a quantitative assessment of the distributional changes of these BTPD colonies during the periods 1988–2000 and 2000–2005 focusing on the potential roles of plant biomass, landscape configuration, habitat loss, and habitat suitability. We used remote sensing and GIS tools to characterize habitat and landscape conditions at areas of extinction and persistence at the beginning of each period. Based on this information, we contrasted areas of extinction and persistence within single BTPD colonies, and used logistic regression to model extinction of entire colonies. The greatest levels of reduction, fragmentation and extinction of colonies occurred between 1988 and 2000. The trend of reduction continued between 2000 and 2005 because the largest colony became smaller and fragmented, driving the area down, however, the rest of the towns showed a marginal increase. During the first period, extinction of entire colonies was higher in smaller colonies occupying low-biomass areas. Both extinction models and raw data indicate that colony area lost to agriculture and urbanization was relatively low for both periods. Because the period exhibiting the highest colony area loss coincided with a severe period of drought (1994–1995), we hypothesized that an unusually high food shortage could be the driving force behind this generalized reduction in colonies. Our results suggest that BTPD populations are sensitive to size and isolation effects mainly in the context of severe drought. Because changes in precipitation have profound impacts on plant productivity and composition in arid ecosystems, BTPD populations at the southernmost edge of their geographic range are especially vulnerable to drought and desertification processes, and therefore to climate disruption.  相似文献

We evaluated the ability of Brassica napus L. (oilseed rape), Helianthus annus L. (sunflower), and Glycine max L. (soybean) plants grown inoculated with or without bacteria to utilize organic P sources. Plants were supplied with inorganic (dibasic sodium phosphate) and organic P sources (phytate and glucose phosphate) at three concentrations and grown for 40 d under sterile conditions. Three inoculation treatments were compared: control (non‐inoculated plants), inoculation with Bacillus amyloliquefaciens BNM340, and inoculation with Pseudomonas fluorescens BNM296 (two bacteria with proven phytase activity). Oilseed rape, sunflower and soybean could utilize organic P sources. For example, when phytate (0.5 mM) P was used as the external P source, the increase factors over the no‐P treatments were 4.5, 1.4, and 1.4 for oilseed rape, sunflower, and soybean P uptake, respectively. When glucose 1‐phosphate disodium salt (G1P, 0.5 mM) was the P source, the increase factors were 8.8, 1.7, and 1.9 respectively. Positive responses to the organic P sources were found for the biomass accumulation of oilseed rape and soybean but not for sunflower. The inoculation with bacteria did not exert a promoting effect on P uptake. We demonstrate that the three species can effectively use organic P sources. The existence of crop plants that are more efficient in the utilization of different soil P sources would be particularly beneficial to improve P recycling and use of P fertilizers in agriculture.  相似文献

Eleven soft tissue- and five osteosarcoma canine patients were subjected to: (i) periodic subcutaneous injection of irradiated xenogeneic cells secreting hGM-CSF and hIL-2 mixed with allogeneic or autologous tumor homogenates; and (ii) injections of cIFN-β and HSVtk-carrying lipoplexes and ganciclovir, marginal (after surgery) and/or intratumoral (in the case of partial tumor resection, local relapse or small surface tumors). This treatment alone (4 patients) or as surgery adjuvant (12 patients), was safe and well tolerated. In those patients presenting local disease (6/11), the suicide gene plus cIFN-β treatment induced local antitumor activity evidenced by the objective responses (3 complete, 2 partial) and stable diseases (2). In addition, the treatment prevented or delayed local relapse, regional metastases (lymph nodes developed only in 3/16) and distant metastases (0/16), suggesting a strong systemic antitumor immunity. The most encouraging result was the long survival times of 10 patients (>1 year, with good quality of life).  相似文献

This study was conducted to determine whether flushing or the stimulus of estrogenized goats is necessary to achieve a high reproductive response in anestrous goats on rangeland. Does were feed-supplemented on rangeland (flushed, n = 20). Other group was exposed to estrogenized does during the mating period (stimulated, n = 20). A third group was both supplemented and stimulated (stimulated–flushed, n = 20) and other group grazed on rangeland only (control, n = 18). More goats in the stimulated and stimulated–flushed groups showed estrus during the first 5 days of joining (45% and 60%, respectively) than the control and flushed groups (11% and 5%, respectively; P < 0.05). Flushing did not improve prolificacy and reduced (P < 0.05) kidding rates (40% and 35% for the flushed and stimulated–flushed groups, respectively) compared to 67% and 55% for the control and stimulated groups, respectively. This experiment demonstrates potential shortcomings of use of flushing in range goats with inadequate nutrition during gestation.  相似文献
