Mammalian X-chromosome inactivation involves a coordinate shutting down of physically linked genes. Several proposed models require the presence of specific sequences near genes to permit the spread of inactivation into these regions. If such models are correct, one might predict that heterologous genes transferred onto the X chromosome might lack the appropriate signal sequences and therefore escape inactivation. To determine whether a foreign gene inserted into the X chromosome is subject to inactivation, transgenic mice harboring 11 copies of the complete, 17-kilobase chicken transferrin gene on the X chromosome were used. Male mice hemizygous for this insert were bred with females bearing Searle's translocation, an X-chromosome rearrangement that is always active in heterozygous females (the unrearranged X chromosome is inactive). Female offspring bearing the Searle's translocation and the chicken transferrin gene had the same amount of chicken transferrin messenger RNA in liver as did transgenic male mice or transgenic female mice lacking the Searle's chromosome. This result shows that the inserted gene is not subject to X-chromosome inactivation and suggests that the inactivation process cannot spread over 187 kilobases of DNA in the absence of specific signal sequences required for inactivation. 相似文献

Histoplasma capsulatum is an effective intracellular parasite of macrophages and causes the most prevalent fungal respiratory disease in the United States. A "dimorphic" fungus, H. capsulatum exists as a saprophytic mold in soil and converts to the parasitic yeast form after inhalation. Only the yeasts secrete a calcium-binding protein (CBP) and can grow in calcium-limiting conditions. To probe the relation between calcium limitation and intracellular parasitism, we designed a strategy to disrupt CBP1 in H. capsulatum using a telomeric linear plasmid and a two-step genetic selection. The resultingcbp1 yeasts no longer grew when deprived of calcium, and they were also unable to destroy macrophages in vitro or proliferate in a mouse model of pulmonary infection. 相似文献

The mechanism by which Helicobacter species are transmitted remains unclear. To examine the possible role of environmental transmission in marine mammals, we sought the presence of Helicobacter spp. and non-Helicobacter bacteria within the order Campylobacterales in water from the aquatic environment of marine mammals, and in fish otoliths regurgitated by dolphins. Water was collected from six pools, two inhabited by dolphins and four inhabited by seals. Regurgitated otoliths were collected from the bottom of dolphins' pools. Samples were evaluated by culture, PCR and DNA sequence analysis. Sequences from dolphins' water and from regurgitated otoliths clustered with 99.8-100% homology with sequences from gastric fluids, dental plaque and saliva from dolphins living in those pools, and with 99.5% homology with H. cetorum. Sequences from seals' water clustered with 99.5% homology with a sequence amplified from a Northern sea lion (AY203900). Control PCR on source water for the pools and from otoliths dissected from feeder fish were negative. The findings of Helicobacter spp. DNA in the aquatic environment suggests that contaminated water from regurgitated fish otoliths and perhaps other tissues may play a role in Helicobacter transmission among marine mammals. 相似文献

Awareness is growing of health‐promoting functional foods and the use of various plants as nutraceuticals. Due to a suite of organosulfur compounds, onion, and other vegetable Alliums possess a unique antiplatelet effect that may promote cardiovascular health because aggregating platelets can lead to heart attack and stroke. Investigation of the consequences of S nutrition in onion has primarily focused on levels of S ranging from deficiency to just above sufficiency; however, little work has been conducted to examine the effects of supraoptimal S nutrition on organosulfur‐based traits. Four onion selections were grown in hydroponic solutions with 2, 7, and 12 mM SO42—‐S in greenhouse experiments during 1995—1996 and 1996—1997. Onion plants were separated into root, leaf, and bulb portions and analyzed for mineral content. Onion bulb extracts were assayed for in vitro human antiplatelet activity. Bulb and leaf S were not affected by increasing solution culture S, but root S was increased by 98 % when solution culture S was increased from 2 mM to 12 mM. Similar increases in root Ca and Al were detected, suggesting CaSO4 and AlSO4+ were accumulated in and on the root. No directional change in antiplatelet activity was detected as S was increased from 2 to 12 mM. This lack of significant change in antiplatelet activity with increasing S levels suggests that modification of this trait by simply increasing S levels may not be feasible in a solution culture environment. 相似文献
