PrP genetics in ruminant transmissible spongiform encephalopathies   总被引:2,自引:0,他引:2  
Scrapie, bovine spongiform encephalopathy (BSE), and chronic wasting disease (CWD) are prion diseases in ruminants with considerable impact on animal health and welfare. They can also pose a risk to human health and control is therefore an important issue. Prion protein (PrP) genetics may be used to control and eventually eradicate animal prion diseases. The PrP gene in sheep and other representatives of the order Artiodactyles has many polymorphisms of which several are crucial determinants of susceptibility to prion diseases, also known as transmissible spongiform encephalopathies (TSE). This review will present the current understanding of PrP genetics in ruminants highlighting similarity and difference between the species in the context of TSE.  相似文献

A method based on a gel permeation chromatography (GPC) extraction procedure combined with an additional cleanup by solid-phase extraction (SPE) on silica gel and liquid chromatography-mass spectrometry (LC-MS) detection has been validated for the analysis of seven glycidyl esters (GEs) including glycidyl laurate, myristate, palmitate, stearate, oleate, linoleate, and linolenate in various edible oils. This method was conjointly developed and validated by two different laboratories, using two different detection systems, a LC time of flight MS (LC-ToF-MS) and a LC triple-quadrupole MS (LC-MS/MS). The extraction procedure allowed targeting low contamination levels due to a highly efficient matrix removal from the 400 mg oil sample loaded on the GPC column and is suitable for routine analysis as 24 samples can be extracted in an automated and reproducible way every 12 h. GPC extraction combined with SPE cleanup and LC-MS/MS detection leads to a limit of quantification in oil samples between 50 and 100 μg/kg depending on the type of glycidyl ester. Recoveries ranged from 68 to 111% (average = 93%). Quantification was performed by automated standard addition on extracts to compensate matrix effects artifacts. To control recoveries of each sample four isotopically labeled GEs ((13)C(3) or (13)C(4)) were included in the method.  相似文献

ABSTRACT: The application of genetic breeding programmes to eradicate transmissible spongiform encephalopathies in goats is an important aim for reasons of animal welfare as well as human food safety and food security. Based on the positive impact of Prnp genetics on sheep scrapie in Europe in the past decade, we have established caprine Prnp gene variation in more than 1100 goats from the United Kingdom and studied the association of Prnp alleles with disease phenotypes in 150 scrapie-positive goats. This investigation confirms the association of the Met142 encoding Prnp allele with increased resistance to preclinical and clinical scrapie. It reveals a novel association of the Ser127 encoding allele with a reduced probability to develop clinical signs of scrapie in goats that are already positive for the accumulation of disease-specific prion protein in brain or periphery. A United Kingdom survey of Prnp genotypes in eight common breeds revealed eleven alleles in over thirty genotypes. The Met142 encoding allele had a high overall mean allele frequency of 22.6%, whereas the Ser127 encoding allele frequency was considerably lower with 6.4%. In contrast, a well known resistance associated allele encoding Lys222 was found to be rare (0.9%) in this survey. The analysis of Prnp genotypes in Mexican Criollas goats revealed nine alleles, including a novel Phe to Leu substitution in codon 201, confirming that high genetic variability of Prnp can be found in scrapie-free populations. Our study implies that it should be feasible to lower scrapie prevalence in goat herds in the United Kingdom by genetic selection.  相似文献

Next-generation sequencing technologies have enabled the rapid generation of high-resolution genetic maps to enable the identification of genomic regions associated with traits of interest. In certain cases these regions of interest need to be narrowed down via fine mapping to increase resolution to the level of the individual gene. In those cases, there is a choice of genotyping options, whereby cost, time, and information content need to be considered. We developed detailed, customizable models to compare the cost of genotyping by sequencing and allele-specific PCR for fine-mapping genomic regions of interest. The models were validated experimentally with data from a fine-mapping experiment designed to identify candidate genes within a 3.2-Mb disease resistance locus in the cereal crop sorghum (Sorghum bicolor (L.). Moench), which has a sequenced genome. The size of the mapping population, genetic diversity of the parental lines, choice of reagents, and labor cost were shown to influence the overall cost of the two methods, and, consequently, which method would be most cost-effective for a given experiment. The model can be easily customized to reflect experiments with different species, genetic populations, experimental and personnel costs to determine the most cost-effective procedure for a new positional cloning project.  相似文献

Rotational grazing management approaches are regarded as strategies for sustaining rangeland productivity and continue to be applied across many parts of the world. In Africa, livestock farmers implementing rotational grazing often switch from traditional loosely bunched herding (LBH), in which animals within a herd are allowed to spread out naturally when foraging, to tightly bunched herding (TBH) with limited herd spread to increase animal impact on the range. However, there is little scientific information on the actual direct (short-term) effects of this altered herding strategy on livestock productivity. We investigated the direct effects of TBH versus LBH on foraging behavior, nutrition, and performance (weight gain) of cattle in a semiarid savanna rangeland in central Kenya. We conducted the study across two habitat types: a heterogeneous red soil habitat and a relatively homogeneous black cotton soil habitat. Across both habitats, cattle traveled 9–15% less, foraged 10–29% more efficiently, and put on 14–39% more weight when managed with TBH as compared with LBH. These changes occurred despite the fact that stock densities were double to several times higher under TBH, and cattle under this herding regime foraged less selectively, consuming preferred plants less (especially in the black cotton soil habitat) and consuming diets with lower crude protein content (in the red soil habitat). Financial projection showed that the benefit of increased cattle performance under TBH could sufficiently outweigh increased cost of additional labor required to implement this herding strategy. These findings suggest that TBH, as practiced here, can be implemented without livestock production or financial losses. Further, the research demonstrated reduced grazing selectivity under TBH indicates that this herding strategy could potentially be used to reduce grazing pressure on preferred forage plants and maintain herbaceous species diversity without sacrificing cattle performance.  相似文献

There have been increasing efforts to understand the dynamics of organic carbon (OC) associated with measurable fractions of bulk soil. We compared the decomposition of native OC (native C) with that of an added substrate (glucose) on physically separated fractions of a diverse suite of soils. Five soil orders were selected from four contrasting climate zones (Mollisol from temperate, Ultisol and Oxisol from tropics, Andisol from sub-arctic, and Gelisol from arctic region). Soils from the A horizon were fractionated into particulate OC (POC) and mineral-associated OC (MOC) by a size-based method. Fractions were incubated at 20 °C and 50 % water-holding capacity in the dark after the addition of unlabeled d-glucose (0.4 mg C g?1 fraction) and U–14C glucose (296 Bq g?1 fraction). Respiration of glucose 14C indicated 64 to 84 % of added glucose 14C which was respired from POC and 62 to 70 % from MOC within 150 days of incubation, with more than half of the cumulative respiration occurring within 4 days. Native C respiration varied widely across fractions: 12 to 46 % of native C was respired from POC and 3 to 10 % was respired from MOC fractions. This suggested that native C was more stabilized on the MOC than on the POC, but respiration from the added glucose was generally similar for MOC and POC fractions. Our study suggests a fundamental difference between the behavior of freshly added C and native C from MOC and POC fractions of soils.  相似文献

The genetics of the prion protein gene (PRNP) play a crucial role in determining the relative susceptibility to transmissible spongiform encephalopathies (TSEs) in several mammalian species. To determine the PRNP gene variability in European red deer (Cervus elaphus), roe deer (Capreolus capreolus) and chamois (Rupicapra rupicapra), the PRNP open reading frame from 715 samples was analysed to reveal a total of ten single nucleotide polymorphisms (SNPs). In red deer, SNPs were found in codons 15, 21, 59, 78, 79, 98, 136, 168 and 226. These polymorphisms give rise to 12 haplotypes, and one of which is identical to the PRNP of American wapiti (Rocky Mountain elk, Cervus elaphus nelsoni). One silent mutation at codon 119 was detected in chamois and no SNPs were found in roe deer. This analysis confirmed that European wild ruminants have a PRNP genetic background that is compatible with TSE susceptibility, including chronic wasting disease.  相似文献
