Relative to the atmosphere, much of the aerobic ocean is supersaturated with methane; however, the source of this important greenhouse gas remains enigmatic. Catabolism of methylphosphonic acid by phosphorus-starved marine microbes, with concomitant release of methane, has been suggested to explain this phenomenon, yet methylphosphonate is not a known natural product, nor has it been detected in natural systems. Further, its synthesis from known natural products would require unknown biochemistry. Here we show that the marine archaeon Nitrosopumilus maritimus encodes a pathway for methylphosphonate biosynthesis and that it produces cell-associated methylphosphonate esters. The abundance of a key gene in this pathway in metagenomic data sets suggests that methylphosphonate biosynthesis is relatively common in marine microbes, providing a plausible explanation for the methane paradox.  相似文献

It has long been established that the sheep Prnp genotype influences the susceptibility to scrapie, and some studies suggest that it can also determine several aspects of the disease phenotype. Other studies, however, indicate that the source of infection may also play a role in such phenotype. To address this question an experiment was set up in which either of two different natural scrapie sources, AAS from AA136 Suffolk and VVC from VV136 Cheviot sheep, were inoculated into AA136, VA136 and VV136 sheep recipients (n = 52). The immunohistochemical (IHC) profile of disease-associated PrP (PrPd) accumulation in the brain of recipient sheep was highly consistent upon codon 136 homologous and semi-homologous transmission, but could be either similar to or different from those of the inoculum donors. In contrast, the IHC profiles were highly variable upon heterologous transmission (VVC to AA136 and AAS to VV136). Furthermore, sheep of the same Prnp genotype could exhibit different survival times and PrPd profiles depending on the source of infection, and a correlation was observed between IHC and Western blot profiles. It was found that additional polymorphisms at codons 112 or 141 of AA136 recipients resulted in a delayed appearance of clinical disease or even in protection from infection. The results of this study strongly suggest that the scrapie phenotype in sheep results from a complex interaction between source, donor and recipient factors, and that the Prnp genotype of the recipient sheep does not explain the variability observed upon codon 136 heterologous transmissions, arguing for other genetic factors to be involved.  相似文献

Understanding long-term changes in forest ecosystem carbon stocks under forest management practices such as timber harvesting is important for assessing the contribution of forests to the global carbon cycle. Harvesting effects are complicated by the amount, type, and condition of residue left on-site, the decomposition rate of this residue, the incorporation of residue into soil organic matter and the rate of new detritus input to the forest floor from regrowing vegetation. In an attempt to address these complexities, the forest succession model LINKAGES was used to assess the production of aboveground biomass, detritus, and soil carbon stocks in native Eucalyptus forests as influenced by five harvest management practices in New South Wales, Australia. The original decomposition sub-routines of LINKAGES were modified by adding components of the Rothamsted (RothC) soil organic matter turnover model. Simulation results using the new model were compared to data from long-term forest inventory plots. Good agreement was observed between simulated and measured above-ground biomass, but mixed results were obtained for basal area. Harvesting operations examined included removing trees for quota sawlogs (QSL, DBH >80 cm), integrated sawlogs (ISL, DBH >20 cm) and whole-tree harvesting in integrated sawlogs (WTH). We also examined the impact of different cutting cycles (20, 50 or 80 years) and intensities (removing 20, 50 or 80 m3). Generally medium and high intensities of shorter cutting cycles in sawlog harvesting systems produced considerably higher soil carbon values compared to no harvesting. On average, soil carbon was 2–9% lower in whole-tree harvest simulations whereas in sawlog harvest simulations soil carbon was 5–17% higher than in no harvesting.  相似文献

After detection of a high prevalence of scrapie in a large dairy goat herd, 72 infected animals were examined by immunohistochemistry with prion protein (PrP) antibody Bar224 to study the pathogenesis of the infection. Tissues examined included the brain and thoracic spinal cord (TSC), a wide selection of lymphoreticular system (LRS) tissues, the distal ileum and its enteric nervous system (ENS), and other organs, including the mammary gland. The whole open reading frame of the PRNP gene was sequenced and antibodies to caprine arthritis-encephalitis virus (CAEV) infection were determined. Unexpectedly, accumulation of disease-associated PrP (PrPd) in the brain was more frequent in methionine carriers at codon 142 (24/32, 75.0%) than amongst isoleucine homozygotes (14/40, 35.0%). The latter, however, showed significantly greater amounts of brain PrPd than the former (average scores of 9.3 and 3.0, respectively). A significant proportion of the 38 goats that were positive in brain were negative in the ENS (44.7%) or in the TSC (39.5%). These results, together with the early and consistent involvement of the circumventricular organs and the hypothalamus, point towards a significant contribution of the haematogenous route in the process of neuroinvasion. Chronic enteritis was observed in 98 of the 200 goats examined, with no association with either scrapie infection or presence of PrPd in the gut. Lymphoproliferative interstitial mastitis was observed in 13/31 CAEV-positive and scrapie-infected goats; PrPd in the mammary gland was detected in five of those 13 goats, suggesting a possible contribution of CAEV infection in scrapie transmission via milk.  相似文献

Atypical/Nor98 scrapie cases in sheep were diagnosed for the first time in Norway in 1998. They are now identified in small ruminants in most European countries and represent an increasingly large proportion of the scrapie cases diagnosed in Europe. Atypical/Nor98 scrapie isolates have shown to be experimentally transmissible into transgenic mice and sheep but the properties of the TSE agent involved, like its biological and biochemical features, are so clearly distinct from the agent involved in classical scrapie that they have provided a challenging diagnostic for many years. No strain diversity has yet been identified among the atypical/Nor98 scrapie sample cases. The genetic predisposition of the sheep affected by atypical/Nor98 scrapie is almost inverted compared to classical scrapie, and the exact origin of this sporadic TSE strain is still speculative, but a spontaneous, non-contagious origin, like sporadic Creutzfeldt-Jakob disease in humans, can not be excluded. Further transmission and epidemiological studies are needed to better address this hypothesis.  相似文献

On rangelands worldwide, cattle interact with many forms of biodiversity, most obviously with vegetation and other large herbivores. Since 1995, we have been manipulating the presence of cattle, mesoherbivores, and megaherbivores (elephants and giraffes) in a series of eighteen 4-ha (10-acre) plots at the Kenya Long-term Exclosure Experiment. We recently (2013) crossed these treatments with small-scale controlled burns. These replicated experimental treatments simulate different land management practices. We seek to disentangle the complex relationships between livestock and biodiversity in a biome where worldwide, uneasy coexistence is the norm. Here, we synthesize more than 20 yr of data to address three central questions about the potentially unique role of cattle in savanna ecology: 1) To what extent do cattle and wild herbivores compete with or facilitate each other? 2) Are the effects of cattle on vegetation similar to those of wildlife, or do cattle have unique effects? 3) What effects do cattle and commercial cattle management have on other savanna organisms? We found that 1) Cattle compete at least as strongly with browsers as grazers, and wildlife compete with cattle, although these negative effects are mitigated by cryptic herbivores (rodents), rainfall, fire, and elephants. 2) Cattle effects on herbaceous vegetation (composition, productivity) are similar to those of the rich mixture of ungulates they replace, differing mainly due to the greater densities of cattle. In contrast, cattle, wild mesoherbivores, and megaherbivores have strongly guild-specific effects on woody vegetation. 3) Both cattle and wild ungulates regulate cascades to other consumers, notably termites, rodents, and disease vectors (ticks and fleas) and pathogens. Overall, cattle management, at moderate stocking densities, can be compatible with the maintenance of considerable native biodiversity, although reducing livestock to these densities in African rangelands is a major challenge.  相似文献
