The detection and mapping of genetic markers linked to quantitative trait loci (QTL) can be utilized to enhance genetic improvement of livestock populations. With the completion of the bovine genome sequence assembly, single nucleotide polymorphisms (SNP) assays spanning the whole bovine genome and research work on large scale identification, validation and analysis of genotypic variation in cattle has become possible. The objective of the present study was to perform a whole genome scan to identify and map QTL affecting milk production traits and somatic cell scores using linkage disequilibrium (LD) regression and 1536 SNP markers. Three and 18 SNP were found to be associated with only milk yield (MY) at a genome and chromosome wise significance (p < 0.05) level respectively. Among the 21 significant SNP, 16 were in a region reported to have QTL for MY in other dairy cattle populations and while the rest five were new QTL finding. Four SNP out of 21 are significant for the milk production traits (MY, fat yield, protein yield (PY), and milk contents) in the present study. Six and nine SNP were associated with PY at a genome and chromosome wise significant (p < 0.05) level respectively. Three and 17 SNP were found to be associated with FY at a genome and chromosome wise significant (p < 0.05) level. Five and seven SNP were mapped with somatic cell score at a genome and chromosome wise significant (p < 0.05) level respectively. The results of this study have revealed QTL for MY, PY, protein percentage, FY, fat percentage, somatic cell score and persistency of milk in the Canadian dairy cattle population. The chromosome regions identified in this study should be further investigated to potentially identify the causative mutations underlying the QTL.  相似文献

Background: Laser lithotripsy has been used as an alternative to surgical removal of uroliths.
Objective: To describe the procedure and efficacy of laser lithotripsy for removal of lower urinary tract uroliths in horses. Additionally, the ultrastructure and the differences in mineralogy and microstructure from 1 successful and 1 unsuccessful laser lithotripsy case are described.
Animals: Six client-owned horses with 7 episodes of naturally occurring urocystoliths, urethroliths, or both.
Methods: Retrospective study of all horses treated between 2006 and 2008 by laser lithotripsy. All horses were sedated followed by laser lithotripsy. Quantitative urolith analysis was performed in all cases. Ultrastructure and microstructure analyses were performed on uroliths from 2 horses.
Results: Procedural success was achieved in 5 of 7 laser lithotripsy procedures. No complications occurred as a result of laser lithotripsy. One horse developed uroabdomen likely as a result of manual lithotrite disruption of the bladder after failure of laser lithotripsy. There were differences in microstructure between 1 urolith that was successfully fragmented by laser lithotripsy and 1 urolith that was resistant to laser fragmentation.
Conclusions and Clinical Importance: Laser lithotripsy is an effective procedure for removal of some urocystoliths, urethroliths, or both in horses.  相似文献

The primary impacts of urban development on biodiversity are loss and fragmentation of habitat, and changes in the structure, composition and function of remnant native ecosystems. Forest dependent species, including arboreal mammals, are particularly sensitive to these changes due to their highly specific habitat requirements and inability or reluctance to cross the urban matrix. We addressed this problem using a case study of the squirrel glider (Petaurus norfolcensis) in fragmented urban landscapes of southeast Queensland, Australia. We applied a mixed effect modeling approach clustered by patch to quantify the importance of site-level habitat factors relative to edge contrast and habitat patch size on squirrel glider abundance. Analysis of variance was used to test for differences in glider age and sex in interior habitats compared to road and residential edges. We found that edge contrast had a strong negative effect on the abundance of squirrel gliders but is conditional on the presence of a low numbers of unreliable flowering overstorey trees and the availability of critical resources such as nest hollows at a site. We also found that older male gliders prefer forest fragment interiors over road and residential edge habitats. We conclude that although interior habitats are ideal to maintain large stable populations, the conservation value of low contrast edges containing key site-level resources should not be underestimated for arboreal mammals such as the squirrel glider.  相似文献

Exotic predators can have detrimental impacts on indigenous fauna. Lethal predator control is commonly used to reduce predator impacts, but is not always feasible, effective or ethical. A promising non-lethal alternative is refuge supplementation for prey. We conducted a Before-After Control-Impact (BACI) experiment over 3 years to determine the relative effects of predator removal (by exclosure fencing) and artificial refuge supplementation on survival of McCann’s skink (Oligosoma maccanni) in duneland on Kaitorete Spit (South Island, New Zealand). Skink populations on 0.0625 ha-grids were randomly assigned to four treatment groups, each replicated four times: (a) predator exclosure only; (b) artificial retreats only; (c) exclosure + artificial retreats, and (d) control (no exclosure or artificial retreats), and monitored annually by pitfall trapping. Capture-recapture analysis was used to estimate the difference in annual survival probability between pre- and post-treatment periods. On average, survival increased only at grids that received the exclosure-only treatment (effect size of 0.03 (0.017-0.043; unconditional 95% CI)). Reduction in predator abundance (by lethal predator control or predator exclusion), but not artificial refuge supplementation, is predicted to benefit McCann’s skink. Our findings add to other studies highlighting the detrimental impacts of exotic predators on indigenous prey and calls for improved means of reducing predator impacts.  相似文献

This study provides new and practical information to implement the use of photoperiodic manipulation for the control of the sexual maturation of Canadian 6+ Atlantic wolffish (Anarhichas lupus) and 3+ first-time spawners spotted wolffish (Anarhichas minor) from Canadian and Norwegian populations. Wolffish reproductive cycle (gametogenesis and oocyte maturation and spermiation) was monitored for 23 consecutive months. Control groups were held under a 12-month simulated natural photoperiod and treatment groups under an 8-month compressed photoperiod. Plasma sex steroid concentrations (estradiol-17β and 11-ketotestosterone), oocyte diameter growth, and milt production were assessed monthly. For all groups under study, fish subjected to the compressed photoperiod spawned 2–6 months earlier than the controls. Complete out-of-shift cycle was not achieved based on the completion of two reproduction cycles, and this is briefly discussed. Photoperiod treatment induced temporal shifts in sex steroid profiles, which are the likely mediators of altered timing of ovulation/final maturation. Photoperiod has a strong influence on the timing of wolffish maturation and could be used as an efficient and inexpensive tool to secure wolffish reproduction operations (year-round supply of egg and milt and/or timing with optimal temperature regimes).  相似文献

Computational models are useful to estimate agricultural greenhouse gas emissions at regional scales. However, empirically based parameter values are required for the models to accurately represent carbon (C) and nitrogen (N) mineralization rates of different organic amendments in more and less humid regions or during wet and dry periods of the growing season. A controlled environment study was conducted to assess the rates of C and N mineralization in differently processed sewage sludge (biosolids) in wet and dry soil. Parameter values were estimated for use in modelling the degradation of three types of biosolids. A loam soil with either 49% water-filled pore space (WFPS) or 29% WFPS was amended with mesophilic anaerobically digested (digested), alkaline-stabilized, or composted biosolids. Headspace samples were collected and analysed for carbon dioxide (CO2) and nitrous oxide (N2O), and soil samples for nitrate ( NO 3 ) and ammonium ( NH 4 + ). Four different first-order models were fitted to the cumulative CO2–C and N2O–N data (R2 > 0.98), and soil NO 3 (R2 > 0.65) and NH 4 + (R2 > 0.93) concentrations. CO2–C data indicated that C mineralization was higher in soil with 49% WFPS than in soils with 29% WFPS. Seventy-nine percent of the C compounds in digested biosolids degraded in soil with 49% WFPS, compared with 52% for alkaline-stabilized biosolids and 8% for composted biosolids. The fitted coefficient values were similar for all of the four first-order models used in this study and provide useful information for parameterizing more sophisticated mechanistic models of the degradation of biosolids in soil.  相似文献

A new method for measuring dough densities is presented, based on weighing small dough samples in air and immersed in xylene. The method can be used to evaluate the air content of low‐density doughs and to follow the changing density of a proofing dough sample. The method is applied to evaluate the effect of flour strength and surfactant addition on dough aeration and subsequent proofing. Doughs were mixed in a high‐speed mixer from two flours, a strong breadmaking flour and a weak flour. Surfactants sodium stearoyl lactylate (SSL) and diacetyl tartrate esters of monoglyceride (DATEM) were added at three levels, and the air content, proofing dynamics, and baked loaf quality were evaluated. The air content of dough was proportional to headspace pressure in the mixer, while the strong flour occluded less air than the weak flour. Surfactants greatly improved the volume of baked loaves but appeared to have no significant effect on air incorporation during mixing. The addition of surfactants appeared to increase the rate of growth of the dough piece during proofing, possibly due to increased bubble breakup during mixing or to increased rates of mass transfer of CO2 into bubbles during proofing.  相似文献
