The fluorescent-antibody technique was employed for the detection of bluetongue virus in bovine foetal kidney cell cultures inoculated with tissues and blood from experimentally infected animals.

In the first series, a total number of 79 inoculated suckling-mouse brains were examined, 36 as frozen sections alone and 43 as impression slides in conjunction with tissue culture inoculation of the same material. With the combined tissue culture immunofluorescent methods, 36 suspicious were detected by impression smears and 37 positives by the tissue culture out of 43 brains examined. Twenty-two were suspicious out of the 36 examined as frozen sections.

Results obtained with the second series, using sheep tissues, showed that the combined tissue culture-fluorescent antibody method was satisfactory for demonstrating the virus in blood of infected animals 1 to 9 days postinfection, and in some organs after death. No false positive reactions were obtained.

The prefemoral lymph nodes of two calves and a sheep infected with a stock of Trypanosoma congolense transmitted by Glossina morsitans were examined histologically for the presence of trypanosomes. Ten days after infection trypanosomes were found in the subcapsular sinuses of the nodes of a calf and the sheep but parasites were absent from the blood at this time. Trypanosomes were also detected in the prefemeral lymph node of the other calf on examination 30 days after infection, when parasites were also present in the blood. These observations provide further evidence that extravascular foci of trypanosomes develop in infections with T congolense and indicate that it should not be regarded as a strict plasma parasite.  相似文献

Animals' sera collected on Easter Island from December 1964 to February 1965 were tested by appropriate methods for the presence of antibodies to various infections. These included, ornithosis, Q-fever, brucellosis, Johne's disease, leptospirosis, toxoplasmosis and vesicular stomatitis viruses. It appeared that the cattle and sheep were exposed to the ornithosis group of agents. The sheep were also exposed to toxoplasmosis. The low-grade reactions observed on the cattle sera with the leptospira and brucella antigens were not sufficient to indicate past infection. All sera tested with Q-fever and Johne's disease antigens gave negative reactions. The results suggested that neither strain of vesicular stomatitis virus had yet been introduced into this restricted animal population.  相似文献

The amylase and haemolytic tests recommended for the diagnosis of hog cholera were used on normal swine, hog cholera-infected swine, hog cholera-immune swine and swine with some other virus diseases.
