AIM: To characterise and explain the increase in density evident by computerised tomography (CT) and radiography in companion studies as a response to training, in bone in the palmar and dorsal regions of the condyles of the third metacarpal bone (Mc3) of 2-year-old Thoroughbred horses.

METHODS: Compositional back-scattered electron (BSE) imaging in scanning electron microscopy (SEM) and confocal scanning laser microscopy (CSLM) were conducted on polymethyl methacrylate (PMMA)-embedded mediolateral slices of the right distal Mc3 from seven 2-year-old Thoroughbred horses trained on a racetrack and seven untrained horses kept at pasture. One left Mc3 from each group was studied in transverse section planes. This study focussed on regions of Mc3 found to differ in density between the trained and untrained horses in companion studies using CT and radiography.

RESULTS: The increase of bone density in the condyles of Mc3 in trained horses compared with untrained horses occurred, without prior osteoclastic resorption, via the deposition of new bone on pre-existing internal surfaces. Within prior marrow spaces of cancellous bone, there was also rapid formation of immature strands and fronds of bone which were more cellular and mineralised, and more lamellar bone tissue was deposited on these new scaffolding elements in the trained horses. Both resulted in increased bone volume fraction (BVF). The microscopic mineralisation density of the bulk of the new tissue was lower than in pre-existing bone, and CT and radiography underestimated the increase in BVF. The new tissue was thus probably less stiff at the microscopic scale than pre-existing bone, though its addition would stiffen the global structure.

CONCLUSIONS: In Mc3 of all the trained horses, there were obvious differences in microscopic structure compared with those from the untrained horses. Moderate, industry-standard levels of exercise used to prepare young horses for racing induced the formation of new bone in non-bone spaces in bone tissue, such that the bone organ should better withstand later increased levels of exercise.  相似文献

AIMS: This is the first in a series of papers reporting studies in 2-year-old Thoroughbred racehorses that aimed to determine the response of musculoskeletal tissues to early training on grass and sand racetracks. In this paper, the experimental set-up of the whole study is described, and nutritional, workload, and clinical, radiographic and pathological outcomes are reported, including semi-quantitative assessment of macroscopic changes in articular cartilage.

METHODS: The study group comprised 14 two-year-old Thoroughbred fillies reared entirely at pasture. Of these, seven were selected by a licensed racehorse trainer to undergo a 4-week period of initial training in which they were taught to accept saddle and rider, followed by a 13-week period of flatrace training at a racetrack (Weeks 1–13); the other seven fillies were confined to large grass enclosures and were not trained. Nutrient, including macro- and trace-element intakes were estimated. Distances cantered or galloped and average velocities were quantified for the trained horses. All horses were observed daily, weighed approximately weekly, and underwent a clinical lameness examination at Weeks 5, 9 and 13. Distal forelimbs were radiographed prior to Week 1, during Weeks 7–8, and again at the end of the study, when macroscopic changes in articular cartilage of the proximal surface of the proximal phalanx were also scored after staining with India ink.

RESULTS: Dietary intakes met or exceeded recommended requirements for all nutrients except sodium, which was low in the trained horses. Bodyweight increased throughout the study in the untrained horses, and increased until Week 7 and then decreased slightly in the trained horses. Mean velocity data were used to define three stages of the training programme: Stage 1 comprised canter in Weeks 1–4; Stage 2 comprised canter in Weeks 5–8; and Stage 3 comprised canter in Weeks 9–13 and galloping twice weekly. Four of seven horses completed training. These covered a mean distance of 179.2 km at mean velocities (excluding gallops) of 7.63 m/sec (SD 0.58), 8.99 m/sec (SD 0.56), and 8.43 m/sec (SD 0.74) for Stages 1–3, respectively, and galloped 4.45 km at 14.4 (SD 0.1) m/sec. The three horses that did not complete training became lame in Weeks 9, 10 and 11, and covered 147.9 km at velocities of 7.38 m/sec (SD 0.44), 8.88 m/sec (SD 0.33) and 8.43 m/sec (SD 0.59) and galloped 2.1 km. Overall, slight or intermittent lameness in trained horses was noted on 76/655 (12%) of horse observation days. Swelling was evident on 284/655 (41%) of horse observation days in the metacarpophalangeal (MCPJ) and metatarsophalangeal (MTPJ) joints (92%), palmar metacarpal tendon region (7%) or carpus (1%). Swelling of the MCPJ or MTPJ was not associated with obvious lameness. Radiographic changes were minor and no gross lesions in bone or tendon tissue were evident except for one case of dorsal metacarpal disease. Post mortem, the cartilage of some MCPJ and MTPJ had obvious wear lines and high lesion scores, which were not consistently related to clinical evidence of pain, lameness or joint swelling. Mean lesion scores were not significantly different between the MCPJ and MTPJ, or between trained and untrained horses.

CONCLUSIONS: Workload can be readily quantified in racehorses under semi-commercial training conditions. Obvious lesions in cartilage of the MCPJ or MTPJ were present in some trained and some untrained horses and not consistently associated with clinical evidence of lameness, joint swelling or change in other connective tissues.

CLINICAL RELEVANCE: Workload data in racing horses are likely to be highly relevant for studying the pathogenesis of changes in bone, tendon and cartilage during training, for training management and for risk analysis in racehorse populations. Although obvious cartilage lesions produced little clinical effect, such lesions have previously been shown to be progressive and to prejudice athletic capability. Detection of such occult lesions in young horses will require more sophisticated detection methods.  相似文献

OBJECTIVE: To determine associations between age, sex, breed, and month and year of admission and the diagnosis of lead toxicosis in cattle. DESIGN: Retrospective case-control study. Sample Population-Records of all cattle evaluated at North American veterinary teaching hospitals during the years 1963 to 2002, which were available through the Veterinary Medical Database. PROCEDURES: Logistic regression was used to evaluate the associations between postulated risk factors and the occurrence of lead toxicosis in cattle and predict the occurrence of the diagnosis of lead toxicosis in cattle. RESULTS: 413 cases of lead intoxication and 202,363 control cattle were identified and met the inclusion criteria. Cattle < 4 years of age were at increased risk for the diagnosis of lead intoxication relative to cattle > or = 4 years of age. Cattle > or = 2 months and < 6 months of age had the greatest risk for lead intoxication (odds ratio, 12.3). Angus cattle were at greater risk for toxicosis (odds ratio, 1.95), compared with other breeds. The risk of lead toxicosis was greater before 1985 (odds ratio, 1.94) than the risk thereafter. The risk of lead toxicosis diagnosis was greatest in the months of May, June, July, and August. CONCLUSIONS AND CLINICAL RELEVANCE: Lead toxicosis in cattle was associated with age < 4 years and the Angus breed. A seasonal pattern existed with peak occurrence in the late spring and summer. The occurrence of lead toxicosis has declined over time.  相似文献

OBJECTIVE: To determine if once-daily colonic irrigation results in fecal continence for a 24-hour period in dogs with colostomies and if colonic volume increased in response to the irrigation. STUDY DESIGN: A prospective controlled experimental study. ANIMALS: Four intact male and one intact female mixed breed dogs. MATERIALS: All dogs received left end-on paralumbar colostomies. Four dogs received once-daily colonic irrigation for 8 weeks, whereas the control dog did not. Daily fecal weights were recorded for the length of the study in all dogs. Barium enema studies and volumetric studies were used to determine colonic volumes. RESULTS: Daily fecal weights were significantly decreased in treatment dogs compared with the control dog. Colonic volume increased in irrigated dogs in response to daily irrigation over the 8 week period of the study. CONCLUSIONS: Colonic irrigation resulted in significantly decreased fecal production over a 24-hour period. Therefore management of dogs with colostomies would be more practical and cost effective. It did not result in complete fecal continence in this study. Further clinical studies are indicated to determine if longer periods of irrigation would result in complete continence.  相似文献

A comparative study concerning the physicochemical, monomeric composition and biological characters among different fucoidan fractions is presented. Common purification techniques for fucoidan usually involve many steps. During these steps, the important structural features might be affected and consequently alter its biological activities. Three purified fractions were derived from Fucus vesiculosus water extract which, afterwards, were purified by a recently-developed dye affinity chromatography protocol. This protocol is based on dye-sulfated polysaccharide interactions. The first two fractions were obtained from crude precipitated fucoidan at different pH values of the adsorption phase: pH 1 and 6. This procedure resulted in fucoidan_1 and 6 fractions. The other, third, fraction: fucoidan_M, however, was obtained from a buffered crude extract at pH 1, eliminating the ethanol precipitation step. All of the three fractions were then further evaluated. Results revealed that fucoidan_M showed the highest sulfur content (S%), 12.11%, with the lowest average molecular weight, 48 kDa. Fucose, galactose, and uronic acid/glucose dimers were detected in all fractions, although, xylose was only detected in fucoidan_1 and 6. In a concentration of 10 µg·mL−1, Fucoidan_6 showed the highest heparin-like anticoagulant activity and could prolong the APTT and TT significantly to 66.03 ± 2.93 and 75.36 ± 1.37 s, respectively. In addition, fucoidan_M demonstrated the highest potency against HSV-1 with an IC50 of 2.41 µg·mL−1. The technique proved to be a candidate for fucoidan purifaction from its crude extract removing the precipitation step from common purification protocols and produced different fucoidan qualities resulted from the different incubation conditions with the immobilized thiazine toluidine blue O dye.  相似文献

Botrytis calthae is a necrotrophic plant pathogen, closely related to the ubiquitous broad host range fungus Botrytis cinerea, but highly host specific. Botrytis isolates from lesions of Caltha palustris grown at different locations were classified with genetic markers as either B. calthae or Botrytis pseudocinerea, or less frequently as B. cinerea. A PCR‐based identification of B. calthae was developed. Seven haplotypes of B. calthae could be distinguished. Compared to B. cinerea, mycelium growth of B. calthae was similar, but conidiation less abundant, and sclerotia formation was only partially repressed by light. Conidia of B. calthae germinated more slowly, and showed a highly acidic optimum (pH 2·5) compared to B. cinerea conidia (pH 5·3). All B. calthae isolates were sensitive to common anti‐Botrytis fungicides, but showed partial resistance to the succinate dehydrogenase inhibitors boscalid, fluopyram and carboxin. Infection experiments revealed a weak capability of B. calthae to induce necrotic lesions on plants that are hosts for B. cinerea. On C. palustris leaves, B. calthae induced similar lesions to B. cinerea. These data provide a basis for comparative molecular investigation of the physiology and host specificity of B. calthae and closely related Botrytis species.  相似文献

Soybean is the most important oilseed and protein crop worldwide, but in Europe, the acreage is comparably low. Thus, Europe strongly depends on imports of soy products and consequently recent efforts aim at expanding the acreage of soybean, particularly in Central Europe. The aim of this study was, therefore, to assist these breeding efforts by characterizing Central European soybean germplasm, employing a genotyping‐by‐sequencing approach yielding 7741 genomewide distributed markers. Our analysis on genetic diversity and population structure revealed that the Central European lines are most closely related to Swiss and Canadian lines and somewhat more distant from the investigated Chinese and US lines. In addition, we analysed patterns of allelic diversity and the extent of linkage disequilibrium. Collectively, our results can assist breeding of Central European soybean and suggest that further progress can be made by crosses among adapted material but a long‐term success also requires introgression of alleles from non‐European germplasm to further broaden the genetic diversity and incorporate novel traits.  相似文献

Winter triticale, an abundant biomass source for producing biogas in Central Europe, has been bred until now for increased grain yield but not for early biomass yield. Therefore, our objectives were to assess the feasibility of breeding triticale for simultaneous improvement of early biomass and grain yield as well as to predict the early biomass yield using an index of traits measured at maturity stage. A representative sample of 100 triticale lines was evaluated in Experiment I at four locations in Germany. Traits were measured at two growth stages (early dough and full maturity). Genetic variances were significantly different from zero for all traits. The correlated response for early biomass yield estimated from grain yield (0.53) was high, which points to the efficiency of indirect selection. A multiple regression model was developed to predict early biomass yield. The reliability of the model was confirmed in an independent Experiment II with 49 lines. The calibration model explained 55% of total variation for early biomass yield. This clearly underlines that at least during earlier stages of selection, field testing can be carried out for grain yield and related traits to select potential genotypes with high biomass yield based on the developed regression model.  相似文献

Various approaches have been developed to achieve sustainability in forestry, under changing social needs and, consequently, changing definitions of sustainability. This has led to the confusing situation in which various groups have different understandings of the meaning of ‘sustainability’. Likewise, the concepts utilized to achieve sustainability, often with a poorly defined objective, are sometimes not clear and/or inconsistent as a systematic overview regarding definitions and concepts is lacking. Based on a literature review, this paper discusses related terms such as sustainability, sustained yield management, sustainable forestry, sustainable forest management and sustainable development: their history, concepts and relationships, from a European perspective. Finally, flexibility is proposed as a solution to overcome the identified shortcomings at all scales, while focussing on the enterprise level. The origin of the sustainability concept in forestry was first driven by forest experts, while participatory elements have been considered since the more recent idea of sustainable development. Since then, much effort has been made to achieve intragenerational fairness by creating an improved participatory process. Concurrently, the original idea of sustainable forestry as long-term and future-oriented management, considering future generations’ needs fell behind. An increasing standing timber volume in Europe and the discussion on climate change brought new interest in how to cope with risks in the context of pervasive future uncertainties within the scope of promoting sustainable development. Although the consideration of risk has been concentrated on as a topic in forest science in recent years, studies have mainly focused on the enhancement of forest resistance against disturbances. However, precaution and risk avoidance alone are probably insufficient to achieve an improved sustainable development that focuses on intergenerational fairness, as these more defensive approaches may disregard important management opportunities involved with an uncertain future. A perhaps more promising approach, the idea of future options and the ability to respond to changing social and biophysical circumstances (i.e. flexibility) as criteria for sustainable development have only shown a shadowy existence up to now. To further develop the consistency of sustainability concepts, a shift of sustainability approaches from continuity towards flexibility options is proposed.  相似文献
