An enzyme-linked immunosorbent assay (ELISA) was developed to measure total antibody to Toxoplasma gondii in serum samples from macropods. The validity of the assay was established by comparing parasite isolation in mice for 17 Tasmanian pademelons (Thylogale billardierii) and 17 Bennett's wallabies (Macropus rufogriseus rufogriseus). The ELISA was then used to detect antibody against T. gondii in serum from 236 macropods, collected from 21 locations in Tasmania, including Flinders Island. Antibody against T. gondii was detected in 20 animals (15 T. billardierii and 5 M. rufogriseus). There was a significant (p less than 0.01) difference in possession of T. gondii antibodies between adult (greater than or equal to 1 year of age) Tasmanian pademelons and Bennett's wallabies. 相似文献

SUMMARY: Johne's disease was diagnosed in 10 alpacas ( Lama pacos ) in Australia between February 1993 and May 1994. Eight of the animals were between 12 and 24 months of age, one was a 6-year-old female, and one was a 4-year-old male. Five, including the 6-year-old and the 4-year-old alpacas, showed weight loss and diarrhoea before death or slaughter. The other cases showed no clinical signs of Johne's disease but 4 gave a positive result on faecal culture and one gave a positive result on testing with the caprine ACID assay and had acid-fast organisms in its faeces. At necropsy, all cases had grossly enlarged mesenteric lymph nodes. Johne's disease was diagnosed after histological examination of the lymph nodes with conventional culture and polymerase chain reaction testing of tissue samples. This report outlines the clinical, epidemiological, and pathological findings in these cases. 相似文献

The objective of this study was to evaluate different concentrations of growth hormone (GH) on the development of bovine preantral follicles cultured included in the ovarian tissue (in situ) on the rates of morphologically normal, viable, primordial and developing follicles, as well as the oocyte and follicle diameter and ultrastructural analysis. Ovarian fragments collected from cows with no cross‐breeds defined were cultured in situ for 1 and 7 days in minimal essential medium (α‐MEM+) supplemented with different concentrations of recombinant human GH (0, 10, 25, 50 ng/ml). The ovarian fragments non‐cultured (control) and cultured were processed for classic histology, mechanical isolation and electron transmission microscopy (MET). The parameters underwent anova (Tukey′s and Dunnett′s tests) and chi‐square test (χ2). After 7 days of culture, the treatment with 50 ng/ml GH showed no differences with fresh control (p > 0.05) and had greater effectiveness than in the 0, 10 and 25 ng/ml GH concentrations of the morphologically normal follicles. Regarding the primordial follicles, a reduction was observed in the 50 ng/ml GH concentration concomitant with the significant increase in developing follicles, differing from both the fresh control and the other GH concentrations tested. In addition, 50 ng/ml GH showed a larger follicle and oocyte diameter when compared to the other treatments cultured. Similar structures were ultrastructurally observed in the control group, 50 ng/ml GH. Follicles cultured in 10 ng/ml GH showed nuclear invagination, vacuoles and lesioned basal membrane. Hence, it is concluded that 50 ng/ml GH is the most effective concentration for the development of preantral follicles cultured in situ. 相似文献

Following a field campaign to determine the species composition, canopy cover, aboveground annual production and leaf area index (LAI) of the semi-arid savanna of north-western Namibia, we present a production model that can be used by graziers to determine the livestock carrying capacity. The model predicts the annual aboveground net primary production (ANPP) from regression equations of canopy cover by annual production fraction for plant functional classes. We tested the output of the model against another fully independent net primary production (NPP) model, namely the MODIS NPP product. The mean MODIS NPP for the 29 sites was 343 ± 22?kg dry matter (DM) ha?1 y?1 as opposed to 285 ± 142?kg DM ha?1 y?1 for the fAP model that used the regression method (p < 0.01). As a proof of concept, these landscape-scale ANPP values are used to calculate a recommended livestock carrying capacity for the Ehirovipuka Communal Conservancy, a 1 980 km2 communal area with both wildlife and livestock populations. In addition, we also provide details of a field method for predicting landscape-scale LAI from line transect data. This approach can be used to ground reference the LAI values generated from the MODIS LAI product. 相似文献

Wisent, or European bison (Bison bonasus), is listed as “vulnerable” on the IUCN Red List of Threatened Species and is therefore protected by international law. For the first time, a Wisent embryo has been obtained in vitro. Bovine viral diarrhoea virus (BVDV) is an endemic pathogen worldwide and eradication strategies focus on the identification and removal of persistently infected (PI) animals arising after in utero infection. Despite this, acute infections with BVDV can persist for months or years after the removal of the PI source despite repeated screening for PIs and tight biosecurity measures. Recent evidence for a prolonged duration of viraemia in the testicles of bulls following acute BVDV infection suggests the possibility of a form of chronic persistence that may more closely resemble the persistence strategies of hepatitis C virus (HCV). To investigate the potential for virus transmission from infected and recovered cattle to virus naïve hosts we established an acute infection of 5 BVDV-naïve calves and monitored animals over 129 days. Infectious BVDV was detected in white blood cells between days 3 and 7 post-challenge. The animals seroconverted by day 21 post-infection and subsequently were apparently immune and free from infectious virus and viral antigen.Animals were further monitored and purified white blood cells were stimulated in vitro with phytohaemagglutinin A (PHA) during which time BVDV RNA was detected intermittently.Ninety-eight days following challenge, blood was transferred from these apparently virus-free and actively immune animals to a further group of 5 BVDV-naïve calves and transmission of infection was achieved. This indicates that BVDV-infected, recovered and immune animals have the potential to remain infectious for BVDV-naïve cohorts for longer than previously demonstrated. 相似文献

The purpose of our work was to establish an efficient-oriented enucleation method to produce transgenic embryos with handmade cloning (HMC). After 41–42 h oocytes maturation, the oocytes were further cultured with or without 0.4 μg/ml demecolcine for 45 min [chemically assisted handmade enucleation (CAHE) group vs polar body (PB) oriented handmade enucleation (OHE) group respectively]. After removal of the cumulus cells and partial digestion of the zona pellucida, oocytes with visible extrusion cones and/or polar bodies attached to the surface were subjected to oriented bisection. Putative cytoplasts without extrusion cones or PB were selected as recipients. Two cytoplasts were electrofused with one transgenic fibroblasts expressing green fluorescent protein (GFP), while non-transgenic fibroblasts were used as controls. Reconstructed embryos were cultured in Well of Wells (WOWs) with porcine zygote medium 3 (PZM-3) after activation. Cleavage and blastocyst rates were registered on day 2 and day 7 of in vitro culture respectively. Meanwhile, the total blastocyst cell number was counted on day 7. We found that the difference was only observed between blastocyst rates (38.6 ± 2% vs 48.1 ± 3%) of cloned embryos with GFP transgenic fibroblast cells after CAHE vs OHE. With adjusted time-lapse for zonae-free cloned embryos cultured in WOWs with PZM-3, it was obvious that in vitro developmental competence after CAHE was compromised when compared with the OHE method. OHE enucleation method seems to be a potential superior alternative method used for somatic cell nuclear transfer (SCNT) with transgenic fibroblast cells. 相似文献

Cloacal and pharyngeal swabs from 100 tree-nesting Double-crested cormorant (DCC) chicks were examined by culture for commensal and potentially pathogenic bacteria. No Salmonella or Erysipelothrix were isolated from the cloacal swabs. Twenty-two cloacal swabs were positive for Campylobacter, of which 14 were C. jejuni, C. coli, and 1 C. lari. None belonged to common serotypes isolated from humans or animals in recent years in Canada. Tests for antimicrobial drug resistance among 187 commensal Escherichia coli isolates from the cloacal swabs indicated that < or =5% were resistant to any of the 12 antibiotics tested. This contrasts with the frequently high resistance rates among E. coli isolates from poultry. Pharyngeal swabs from DCC were negative for Pasteurella multocida. Culture of cloacal swabs from 100 ground-nesting DCC chicks resulted in the recovery of 19 Salmonella isolates, all of which were S. enterica serotype Typhimurium. None of these isolates were resistant to any of the 12 antibiotics tested. Altogether, these findings suggest that DCC from this region are not being colonized with commensal or potentially pathogenic enteric bacteria from agricultural or human sources and that enteric bacteria isolated from these birds are unlikely to contribute to a gene pool of antimicrobial drug resistance. 相似文献
