Site-specific management (SSM) is a common way to manage within-field variability. This concept divides fields into site-specific management zones (SSMZ) according to one or several soil or crop characteristics. This paper proposes an original methodology for SSMZ delineation which is able to manage different kinds of crop and/or soil images using a powerful segmentation tool: the watershed algorithm. This image analysis algorithm was adapted to the specific constraints of precision agriculture. The algorithm was tested on high-resolution bio-physical images of a set of fields in France.  相似文献

Oak leaves have a high concentration of ellagitannins. These phytochemicals can be beneficial or poisonous to animals. Beef cattle are often intoxicated by oak leaf consumption, particularly after suffering feed restriction. The severity of the poisoning has recently been associated with the ruminal microbiota, as different bacterial populations were found in animals that tolerated oak leaves and in those that showed clinical and pathological signs of toxicity. Intoxication has previously been linked to the production of phenolic metabolites, particularly catechol, phloroglucinol, and resorcinol. This suggested that the microbial metabolism of ellagitannins could also be associated with its tolerance or intoxication in different animals. Therefore, it is essential to understand the metabolism of ellagitannins in cattle. Here we show that ellagitannins are metabolized in the cattle rumen to urolithins. Different urolithins were detected in ruminal fluid, feces, urine, and plasma. Oak leaf ellagitannins declined as they were converted to urolithins, mainly isourolithin A and urolithin B, by the ruminal and fecal microbiota. Urolithin aglycons were observed in rumen and feces, and glucuronide and sulfate derivatives were detected in plasma and urine. Sulfate derivatives were the main metabolites detected in plasma, while glucuronide derivatives were the main ones in urine. The main urolithins produced in cattle were isourolithin A and urolithin B. This is a relevant difference from the monogastric mammals studied previously in which urolithin A was the main metabolite produced. Low molecular weight phenolics of the benzoic, phenylacetic, and phenylpropionic groups and metabolites such as catechol, resorcinol, and related compounds were also detected. There was a large variability in the kinetics of production of these metabolites in individual animals, although they produced similar metabolites in all cases. This large variability could be associated with the large variability in the rumen and intestine microbiota that has previously been observed. Further studies are needed to demonstrate if the efficiency in the metabolism of ellagitannins by the microbiota could explain the differences observed in susceptibility to intoxication by the different animals.  相似文献

In the spring of 2003, a series of sudden deaths in a group of adult lions (Panthera leo) with a previous history of depression, inanition, and lethargy, was investigated. Five animals died within 24 to 36 hours after onset of signs of disease. Serologic screening for viral disease detection was negative, evidence of parasites was not detected, and results of a complete blood count and serum biochemical analysis were within reference intervals in all lions. The most relevant lesions observed were multiple areas of necrosis and hemorrhage in the intestinal outer muscular layer, and cellulitis with an intense bloody edema in the mesenteric and the pericardial fat tissue. On the basis of the fulminant course of the disease, the gross and histologic findings, and the isolation and identification of Clostridium sordellii, a diagnosis of infectious myositis and cellulitis associated with acute clostridiosis was made. To the authors' knowledge, this is the first report of sudden death associated with C. sordellii in felines.  相似文献

BACKGROUND: Didymella bryoniae has a history of developing resistance to single‐site fungicides. A recent example is with the succinate‐dehydrogenase‐inhibiting fungicide (SDHI) boscalid. In laboratory assays, out of 103 isolates of this fungus, 82 and seven were found to be very highly resistant (BVHR) and highly resistant (BHR) to boscalid respectively. Cross‐resistance studies with the new SDHI penthiopyrad showed that the BVHR isolates were only highly resistant to penthiopyrad (BVHR‐PHR), while the BHR isolates appeared sensitive to penthiopyrad (BHR‐PS). In this study, the molecular mechanism of resistance in these two phenotypes (BVHR‐PHR and BHR‐PS) was elucidated, and their sensitivity to the new SDHI fluopyram was assessed. RESULTS: A 456 bp cDNA amplified fragment of the succinate dehydrogenase iron sulfur gene (DbSDHB) was initially cloned and sequenced from two sensitive (BS‐PS), two BVHR‐PHR and one BHR‐PS isolate of D. bryoniae. Comparative analysis of the DbSDHB protein revealed that a highly conserved histidine residue involved in the binding of SDHIs and present in wild‐type isolates was replaced by tyrosine (H277Y) or arginine (H277R) in the BVHR‐PHR and BHR‐PS variants respectively. Further examination of the role and extent of these alterations showed that the H/Y and H/R substitutions were present in the remaining BVHR‐PHR and BHR‐PS variants respectively. Analysis of the sensitivity to fluopyram of representative isolates showed that both SDHB mutants were sensitive to this fungicide as the wild‐type isolates. CONCLUSION: The genotype‐specific cross‐resistance relationships between the SDHIs boscalid and penthiopyrad and the lack of cross‐resistance between these fungicides and fluopyram should be taken into account when selecting SDHIs for gummy stem blight management. Gas chromatography-orthogonal acceleration time-of-flight mass spectrometry (GC-oaTOFMS) is an emerging technique offering a straightforward access to a resolving power up to 7000. This paper deals with the use of GC-oaTOFMS to identify the flavor components of a complex seafood flavor extract and to quantify furanones formed in model Maillard reactions. A seafood extract was selected as a representative example for complex food flavors and was previously analyzed using GC-quadrupole MS, leaving several molecules unidentified. GC-oaTOFMS analysis was focused on these unknowns to evaluate its potential in flavor research, particularly for determining exact masses. N-Methyldithiodimethylamine, 6-methyl-5-hepten-2-one, and tetrahydro-2,4-dimethyl-4H-pyrrolo[2,1-d]-1,3,5-dithiazine were successfully identified on the basis of the precise mass determination of their molecular ions and their major fragments. A second set of experiments was performed to test the capabilities of the GC-oaTOFMS for quantification. Calibration curves were found to be linear over a dynamic range of 10(3) for the quantification of furanones. The quantitative data obtained using GC-oaTOFMS confirmed earlier results that the formation of 4-hydroxy-2,5-dimethyl-3(2H)-furanone was favored in the xylose/glycine model reaction and 2(or 5)-ethyl-4-hydroxy-5(or 2)-methyl-3(2H)-furanone in the xylose/alanine model reaction. It was concluded that GC-oaTOFMS may become a powerful analytical tool for the flavor chemist for both identification and quantification purposes, the latter in particular when combined with stable isotope dilution assay.  相似文献

Although synthetic pesticides are still used to control insect pests, greater efforts have been made to develop healthier and more environmentally friendly pesticides. Because of their insecticidal properties, botanical essential oils (EOs) are considered as promising alternatives to the use of synthetic insecticides. However, little is known about mechanisms underlying the insecticidal activity of most these natural compounds. In the present study, we evaluated the contact toxicity and the modes of action of the EO from Mentha arvensis against the granary weevil, Sitophilus granarius L. (Coleoptera: Curculionidae), a cosmopolitan insect pest that causes extensive damage to stored cereals. M. arvensis EO caused high contact toxicity in S. granarius adults, resulting in a rapid paralysis and rapid alteration of walking behavior. Our label-free quantitative proteomics approach revealed that M. arvensis EO induced dramatic physiological changes in exposed insects. The majority of the differentially expressed proteins (DEPs) were upregulated and are related to the development and functioning of the muscular and nervous systems, cellular respiration, protein synthesis, and detoxification. These results suggest that M. arvensis essential oil is capable of affecting a variety of biological processes, and shed light on the repair mechanisms put in place in surviving insects to counter the damage inflicted. This work opens new perspectives on the proposed mechanisms of insecticidal activity of a promising EO for controlling pests of stored cereals and may represent a first step in the development of novel bio-rational insecticides.

Two Asian longhorn beetles, Anoplophora glabripennis and Anoplophora chinensis, are among the most serious alien invasive species attacking forest and urban trees, both in North America and Europe. Major efforts have been put into preventing further entry and establishment of the two species as well as promoting their successful eradication. Here, we review these efforts, their progress and outcome, and scientific advancements in monitoring and control methods. The combined international activities and harmonizing legislative changes in detection and eradication methods have proven worthwhile, with more than 45% of eradication programmes successful in the last 12 years. Some countries were able to completely eradicate all populations and others managed to reduce the area affected. Although the costs of the eradication programmes can be very high, the benefits outweigh inaction. Attempts to eradicate A. chinensis have been more challenging in comparison with those targeting A. glabripennis. For both species, efforts are hampered by the ongoing arrival of new beetles, both from their native regions in Asia and from other invaded regions via bridgehead effects. The methods used for eradication have not changed much during the last decade, and host removal is still the method most commonly used. On the other hand, detection methods have diversified during the last decade with advances in semiochemical research and use of detection dogs. The next decade will determine if eradications continue to be successful, particularly in the case of A. chinensis, which has been targeted in some countries for containment instead of eradication.

ABSTRACT: This study shows that high pathogenic H5N1 influenza virus infection of chicken induced high levels of bioactive interferon type I in the lung (4.3 × 105 U/mg tissue), plasma (1.1 × 105 U/mL), and spleen (9.1 × 105 U/mg tissue). In contrast, a low pathogenic attenuated H5N1 vaccine strain only induced approximately 24 times less IFN in the lung, 441 times less in the spleen and 649 less in the plasma. This was in the same range as a reassortant carrying the HA from the vaccine strain and the remaining genes from the high pathogenic virus. On the other hand, a reassortant virus with the HA from the high pathogenic H5N1 with the remaining genes from the vaccine strain had intermediate levels of IFN. The level of interferon responses related to the viral load, and those in the spleen and blood to the spread of virus to lymphoid tissue, as well as disease severity. In vitro, the viruses did not induce interferon in chicken embryonic fibroblasts, but high levels in splenocytes, with not clear relationship to pathogenicity and virulence. This, and the responses also with inactivated viruses imply the presence of plasmacytoid dendritic cell-like leukocytes within the chicken immune system, possibly responsible for the high interferon responses during H5N1 infection. Our data also indicate that the viral load as well as the cleavability of the HA enabling systemic spread of the virus are two major factors controlling systemic IFN responses in chicken.  相似文献

Several methanogenic archaea lack cysteinyl-transfer RNA (tRNA) synthetase (CysRS), the essential enzyme that provides Cys-tRNA(Cys) for translation in most organisms. Partial purification of the corresponding activity from Methanocaldococcus jannaschii indicated that tRNA(Cys) becomes acylated with O-phosphoserine (Sep) but not with cysteine. Further analyses identified a class II-type O-phosphoseryl-tRNA synthetase (SepRS) and Sep-tRNA:Cys-tRNA synthase (SepCysS). SepRS specifically forms Sep-tRNA(Cys), which is then converted to Cys-tRNA(Cys) by SepCysS. Comparative genomic analyses suggest that this pathway, encoded in all organisms lacking CysRS, can also act as the sole route for cysteine biosynthesis. This was proven for Methanococcus maripaludis, where deletion of the SepRS-encoding gene resulted in cysteine auxotrophy. As the conversions of Sep-tRNA to Cys-tRNA or to selenocysteinyl-tRNA are chemically analogous, the catalytic activity of SepCysS provides a means by which both cysteine and selenocysteine may have originally been added to the genetic code.  相似文献
