ABSTRACT:   Changes in lipid content and fatty acid composition of muscle, liver and ovary of captive-reared and wild silver Japanese eel Anguilla japonica were examined during artificial maturation induced by salmon pituitary homogenate (SPH) injections. Although the relative levels of n-3 and n-6 highly unsaturated fatty acids (HUFA) in liver and ovary were higher than in muscle in both captive and wild silver eels before SPH injection, these tended to decrease with maturation. The relative levels of n-6 HUFA in muscle, liver, ovary and eggs of wild silver eels were remarkably higher than those in captive eels. Therefore, we attempted to alter the ratio of n-6 HUFA in eggs by feeding eels a diet supplemented with linoleic acid-rich plant oil. Although the percentage of n-6 polyunsaturated fatty acids to total fatty acids in eggs of eels fed the supplemented diet was similar to wild silver eels, the percentage of n-6 HUFA remained remarkably lower than in wild silver eels. Hence, it appears that the supplemented diet affected the fatty acid composition of eggs, but did not result in much conversion of linoleic acid to its higher homologs in eels.  相似文献

This study aimed to re‐evaluate the effect of excessive arginine supplementation on growth and feed efficiency of rainbow trout, and to assess the effect of dietary arginine supplementation on hepatic amino acid composition, expression of arginase Ⅱ (ARG 2), inducible nitric oxide synthase (iNOS), and heat shock protein 70 (HSP70) genes in the intestine, and plasma arginine, ornithine, citrulline and urea levels at 0, 6, 12 and 18 hr‐postprandial. Rainbow trout (body weight: 60.5–65 g) were fed diets containing 1.47 (control [CTRL]), 3.89 (3.89A) and 5.64% (5.64A) arginine for nine weeks. Higher muscle protein content was observed in 3.89A than CTRL (p < 0.05). In postprandial study, plasma arginine of 5.64A kept increased until 12 hr‐postprandial and reached identical value (around 150 µg/ml) until 18 hr‐postprandial in 5.64A. Significant increase of plasma arginine level was only observed in arginine supplemented group. Meanwhile, plasma citrulline level in CTRL was significantly higher than in 5.64A at 18 hr‐postprandial. A significantly higher hepatic citrulline level was also observed in CTRL than in 5.64A (p < 0.05). Significantly higher plasma and hepatic free ornithine level was observed in 5.64A than CTRL while significantly higher expression of intestinal ARG2 and HSP70 was found in arginine supplemented groups. These results suggest that citrulline availability seems to be stimulated by arginine deficiency in the CTRL and the presence of arginase in the intestine to regulate excess dietary arginine.  相似文献

This paper provides an insight into the potential of utilising sago pith waste (SPW) to produce composite material. SPW is a fibrous waste that is produced as a result of the sago starch extraction process. It is made up of more than 60 % in weight of sago starch and was successfully converted into natural fibre reinforced thermoplastic starch composite using twin screw extrusion in the presence of different quantities of glycerol and water as plasticisers. No binder was added throughout the process. Good fibre-matrix compatibility was proven by SEM. Due to phase separation and fibre agglomeration, tensile strength of the composite decreased with increasing glycerol content while elongation at break remained unchanged at low values. Activation energies, Ea of the composites were computed from thermogravimetric analysis (TGA) data using Broido and Coat-Redfern equations. The obtained Ea values indicated that plasticisation led to the reduction in thermal resistance of SPW. It is suggested that blending the plasticised SPW with sago starch and polyvinyl alcohol will greatly improve upon the existing shortcomings, potentially resulting in useful consumer products from SPW, an unutilised, water polluting waste.  相似文献

Background

Previous reports associated 2 mutant SOD1 alleles (SOD1:c.118A and SOD1:c.52T) with degenerative myelopathy in 6 canine breeds. The distribution of these alleles in other breeds has not been reported.

Objective

To describe the distribution of SOD1:c.118A and SOD1:c.52T in 222 breeds.

Animals

DNA from 33,747 dogs was genotyped at SOD1:c.118,SOD1:c.52, or both. Spinal cord sections from 249 of these dogs were examined.

Methods

Retrospective analysis of 35,359 previously determined genotypes at SOD1:c.118G>A or SOD1:c.52A>T and prospective survey to update the clinical status of a subset of dogs from which samples were obtained with a relatively low ascertainment bias.

Results

The SOD1:c.118A allele was found in cross‐bred dogs and in 124 different canine breeds whereas the SOD1:c.52T allele was only found in Bernese Mountain Dogs. Most of the dogs with histopathologically confirmed degenerative myelopathy were SOD1:c.118A homozygotes, but 8 dogs with histopathologically confirmed degenerative myelopathy were SOD1:c.118A/G heterozygotes and had no other sequence variants in their SOD1 amino acid coding regions. The updated clinical conditions of dogs from which samples were obtained with a relatively low ascertainment bias suggest that SOD1:c.118A homozygotes are at a much higher risk of developing degenerative myelopathy than are SOD1:c.118A/G heterozygotes.

Conclusions and Clinical Importance

We conclude that the SOD1:c.118A allele is widespread and common among privately owned dogs whereas the SOD1:c.52T allele is rare and appears to be limited to Bernese Mountain Dogs. We also conclude that breeding to avoid the production of SOD1:c.118A homozygotes is a rational strategy.  相似文献

Bifidobacterial plasmids reported so far are derived from a limited number of strains and plasmids of bifidobacterial type strains isolated from humans are unknown. We found that Bifidobacterium kashiwanohense JCM 15439 (type strain) isolated from a healthy infant contained two cryptic plasmids, designated pBBKW‐1 and pBBKW‐2. We determined and analyzed the complete sequences of both plasmids. pBBKW‐1 (7716 bp) was predicted to replicate by a rolling‐circle mechanism and encode six protein‐coding genes, two of which are putative replication proteins. pBBKW‐1 seems to be a cointegrate plasmid containing two copies of the plasmid pMG1 from Bifidobacterium longum. pBBKW‐2 (2920 bp) was predicted to encode six protein‐coding genes and be a theta‐type replicating plasmid, which has been reported to be more stable than a rolling circle‐type replicating plasmid frequently found in bifidobacteria. Our finding will provide new insights into safe recombinant plasmid constructions for humans.  相似文献

Three known saponins, chiisanoside, 11-deoxyisochiisanoside, and isochiisanoside, and one novel saponin, 3,4-seco-4(23),20(29)-lupadiene-3,28-dioic acid 28-O-alpha-l-rhamnopyranosyl (1-->4)-beta-d-glucopyranosyl (1-->6)-beta-d-glucopyranoside, referred to as sessiloside, were isolated from a hot water extract of Acanthopanax sessiliflorus leaves. All of these saponins were lupane-type triterpene triglycosides, and their concentrations were 4.1, 1.0, 0.5, and 0.4% (w/w) of the total extract, respectively. Sessiloside and chiisanoside inhibited pancreatic lipase activity in vitro, and addition of the saponin-rich fraction to a high-fat diet suppressed the body weight gain of mice. The possibility of application of the lupane-type saponins from A. sessiliflorus leaves to the treatment of obesity is discussed.  相似文献

To detect the predominant lactobacilli in the intestinal flora of healthy thoroughbreds, we isolated lactobacilli from the feces of nine thoroughbreds (five males and four females; 0–15-year-old). The isolated lactobacilli comprise 17 species (37 strains), and they were classified into five groups: Lactobacillus salivarius (6 species), L. reuteri (6 species), Lactobacillus delbrueckii (3 species), L. buchneri (1 species) and L. vitulinus (1 species). On the basis of 16S rRNA gene sequences, we identified 3 other phylogenetic relatives belonging to the genus Lactobacillus . These results suggest that the intestinal flora of thoroughbreds may comprise many species of the genus Lactobacillus . Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) analyses of the 340-bp fragments of the 16S rRNA genes from the same nine fecal samples showed that L. hayakitensis , L. equigenerosi and L. equi are contained in all the samples, suggesting that these species are predominant lactobacilli in the intestinal flora of thoroughbreds.  相似文献
