The therapeutic efficacy of atovaquone against Babesia gibsoni was examined in three dogs experimentally infected with B. gibsoni isolated from naturally infected dogs in Aomori Prefecture, Japan. Once parasitemia reached 10%, atovaquone was administered orally (30 mg/kg twice daily for 7 days). Within 2 days of atovaquone treatment, the parasite disappeared from blood smears without any clinical side effects. Anemia and thrombocytopenia were significantly improved in all the dogs. However, a polymerase chain reaction assay revealed that a B. gibsoni marker gene was intermittently present in peripheral blood after atovaquone therapy, indicating that the organism had not been eliminated, and parasites reappeared in blood smears 33 days after the last treatment. To investigate the change in sensitivity against atovaquone, an in vitro sensitivity test was performed using peripheral blood obtained from an untreated dog that was infected with the original parasite isolate, and from two of the experimentally infected and atovaquone-treated animals (blood was collected at the time of the post-treatment recurrence of the B. gibsoni infection). Atovaquone was added to the culture medium to final concentrations of 0.1, 1, 10, 100, and 1000 nM. For the untreated parasites, complete growth inhibition occurred at 1000 nM of atovaquone, whereas the recurrent parasites were inhibited by only 39.52 +/- 8.34% and 31.31 +/- 8.14% at this concentration after 48 h of incubation. An unusual case diagnosed as connective tissue-type mast cell leukemia with marked mastocyte infiltration into visceral organs in a seven-year-old female Curly-Coated retriever is presented. Acute circulatory collapse, emesis, diarrhea, abdominal enlargement, icterus, cyanosis, dyspnea, pulmonary edema, hepatomegary, ascites, and right ventricular enlargement were observed. Hematologic and biochemical examinations revealed mast cell leukemia, mature neutrophilia, monocytosis, thrombocytopenia, hemolytic hyperbilirubinemia, hyperhistaminemia, renal and hepatic injuries. Mast cells were distributed systemically, but predominantly in the diaphragm and liver with a large mass among the serosa of ileum, cecum and colon. The anaesthetic and cardiopulmonary effects of combinations of medetomidine (Me), midazolam (Mi) and butorphanol (Bu) were evaluated in dogs. The characterization of anaesthetic effects was assessed using a scoring system. The combinations tested were 20 or 40 micrograms/kg Me and 0.5 mg/kg Mi (20Me-Mi or 40Me-Mi) followed by either an intravenous injection of physiological saline solution (PSS) or Bu (0.1 or 0.3 mg/kg). The mixture of Me and Mi was injected intramuscularly, followed 15 min later by an intravenous injection of Bu or PSS in all six groups. The combined Me-Mi induced deep sedation but not profound anaesthesia. The effect of the subsequent Bu administration was observed in the scores related to its analgesic effect. There were no significant differences between the two doses of Bu, following either 20Me-Mi or 40Me-Mi in the duration of anaesthesia, heart and respiratory rates, rectal temperature, and anaesthetic and analgesic scores except for palpebral reflex, and interdigital web clamping scores. Therefore, we concluded that the addition of 0.1 mg/kg Bu to Me-Mi elicits adequate anaesthesia with adequate analgesic effect, and side-effects such as bradycardia, hypertension, and slight respiratory acidosis in some dogs. 相似文献

Recently, developmental exposure to clothianidin (CLO) has been shown to cause reproductive toxicity in male mice, but the effects in female mice remain to be clarified. Pregnant C57BL/6N mice were given a no-observed-adverse-effect-level (NOAEL) dose of CLO until weaning. We then examined ovaries of 3- or 10-week-old female offspring. In the CLO-administered group, morphological changes, a decrease in the immunoreactivity of the antioxidant enzyme glutathione peroxidase 4 (GPx4), and activation of genes in the steroid hormone biosynthesis pathway were observed in 3-week-old mice, and decreases of GPx4 immunoreactivity, 17OH-progesterone and corticosterone levels were observed in 10-week-old mice, along with high rates of infanticide and severe neglect, providing new evidence that developmental exposure to CLO affects juvenile and adult mice differently. 相似文献

ABSTRACT: In order to elucidate the mechanism of the changes in gel forming characteristics of fish meat by pH-lowering, the gelation-temperature curve and the gelation-moisture content curve were examined using the acidified walleye pollack surimi or neutralized one after acidification. In the gelation-temperature curve, the gel strength was highest at 30°C and lowest at approximately 50–60°C, irrespective of pH shifting. The gel strength at 30°C and 80°C decreased with the decrease in pH value. The neutralization of acidified surimi improved the gel strength, but it was considerably lower than the original gel strength. The gel strength at 50–60°C was not affected by pH lowering. The gel strength at 80°C could not be revived to the original by pH readjustment, either in the presence or in the absence of EDTA. These results suggest that irreversible changes of meat protein take place under the low pH, and the oxidation ability of sulfhydryl (SH) groups of protein molecule is not affected by pH-shifting. 相似文献

Changes in physical properties of two-step heated gels on addition of gluconate were investigated in terms of relationships
between breaking strength and gel stiffness. Regression lines between the breaking strength and the gel stiffness were extended
to the x-axis (gel stiffness), and the intercept was defined as SBSO. The SBSO of the two-step heated gels increased with gluconate contents in salt-ground surimis, suggesting that the harder but less elastic gels formed on addition of gluconate were dose-dependent. Conversely, the denaturation
rate constants of myosin in salt-ground surimis during preheating estimated by means of Ca-ATPase inactivation, loss of salt solubility, and decrease of denaturant solubility
were considerably reduced by gluconate. Thus, the progress of myosin denaturation was strongly suppressed. Increments of SBSO (δSBSO) of the two-step heated gels on addition of gluconate were inversely correlated with the denaturation rate constants of myosin
in salt-ground surimis for every index. Thus, the changes in physical parameters of two-step heated gel caused by gluconate may be associated with

ABSTRACT: During the fermentation of mackerel to narezushi , the concentration of peptides required to inhibit 50% of the ACE activity in the assay media (IC50 ), as an index of the angiotensin I-converting enzyme (ACE) inhibitory activity, was remarkably decreased with a rapid increase in peptide contents. Systolic blood pressure (SBP) in spontaneously hypertensive rats (SHR) decreased between 2 and 4 h after the single oral administration of greater than 10 mg peptide/kg narezushi extract, and recovered to the initial level by 8 h thereafter. The SBP decreased at seven successive daily doses of 10 mg/kg of narezushi extract and then recovered to the initial level 5 days after stopping a total of 10 daily administrations. The extract was administered to five-week-old SHR rats for 70 days and SBP decreased 21 days after starting and continued for 28 days after the end of administration. The peptide-rich fraction from narezushi extract had a powerful antihypertensive effect, whereas the other fraction had a similar, but weak effect. 相似文献
