Heparin-binding fractions were taken from the heparin sepharose columns on which extracts of loach fish (Misgurnus anguillicaudatus) embryos from blastula, gastrula, 4–8 and 12–16 somites stages were applied. These heparin-binding fractions, except the fraction derived from 12–16 somite embryos, showed potent mitogenic activity on fibroblast-like cells derived from caudal fin blastema of goldfish. Western blot analysis of these heparin-binding fractions was carried out using monoclonal antibodies against human acidic and basic fibroblast growth factors (FGF-1 and-2). An immunoreactive FGF-1 band at 16 kD was detected in the heparin-binding fraction derived from embryos in each stage of blastula, gastrula and 4–8 somites. An immunoreactive FGF-2 band at 17 kDa was detected exclusively in the heparin-binding fraction derived from 4–8 somite embryos. In the heparin-binding fraction derived from 12–16 somite embryos neither immunoreactive FGF-1 nor-2 member band was detectable. 相似文献

Genetic structure and phylogenetic relationships in East and South Asian melons were analyzed, based on the geographical variation
of five isozymes. The analysis of Indian melon accessions showed a continuous variation in seed length, ranging from 4 to
13 mm. Most of the East Asian melons, vars. makuwa and conomon, were classified as the small seed type with seed length shorter than 9 mm. The frequency of the small seed type increased
from the west to the east in India. Allelic variation was detected at a total of nine loci of five isozymes among 114 melon
accessions. Gene diversity calculated for the nine loci indicated that Indian melon was rich in genetic variation, which decreased
from India towards the east. Clear geographical variation was detected in two enzymes, APS and6-PGDH. Pgd-1
1 and Ap-3
1 were frequent in India and Myanmar, while most of the melons in Laos, China, Korea and Japan carried Pgd-1
3 and Ap-3
3, except var. inodorusin China. Among the latter two alleles, the frequency of Ap-3
3 was more than 50% in the small seed type in north and east India, indicating that vars. makuwa and conomon were related to the small seed type in these areas. It was also suggested that the small seed type with wet tolerance originated
in central India and was selected under wet condition in the east.

Solanum villosum is an important African leafy vegetable whose yield is limited mainly by competition from early and excess fruit-set. Induced
male-sterility is a potential tool to reduce this competition and enhance yields. This study was conducted to investigate
the influence of photoperiod and temperature on the floral dynamics of a season-dependent male-sterile mutant. The mutant,
named T-5, has flowers which are sepaloid, mostly stamenless, indeterminate and partially restored in winter, late-spring,
summer and autumn, respectively. Floral organ restoration was found to be largely independent of photoperiod conditions. Day/night
temperatures of 25/25 and 30/20°C were found to favour restoration of the floral organ but most flowers were stamenless and
infertile. High night temperature favoured the formation of indeterminate flowers both in the growth chamber (30°C) and in
the greenhouse (>25°C). On the other hand, low growth chamber (10°C) and greenhouse (<15°C) night temperature favoured the
formation of sepaloid flowers. The optimum temperatures for floral structure and fertility restoration were between 20°C and
25°C (day) and 15–20°C (night). Propagation of T-5 mutant can thus be achieved by growing in regions or seasons with such
temperature ranges. Under temperatures unfavourable for fruit-set, leaf productivity is expected to be high. 相似文献

The clonality analysis of the bone marrow cells was carried out by detecting the integrated proviruses of feline leukemia virus (FeLV) to understand the pathogenesis of FeLV-associated hematopoietic disorders in cats. Bone marrow cells from 4 cases with acute myeloid leukemia (AML), 9 cases with myelodysplastic syndromes (MDS), 2 cases with pure red cell aplasia (PRCA) and 3 healthy carriers infected with FeLV were subjected to Southern blot analyses using an exogenous FeLV probe. Clonal hematopoiesis was found in all the cases with AML and in 6 of the 9 cases with MDS, but not in the cases with both PRCA and healthy carriers infected with FeLV. In the 2 cases with MDS, it was thought that the same clones of the hematopoietic cells might proliferate before and after the progression of the disease irrespective of the changes of the hematological diagnoses by cytological examination. This study indicates that MDS in cats is a disease manifestation as a result of clonal proliferation of hematopoietic cells and can be recognized as a pre-leukemic state of AML. 相似文献

Wheat starch granules were separated from waxy wheat flour by the acetic acid (pH 3.5) fractionation technique. Waxy wheat starch samples were stained red‐brown using KI/I2 solution. Subsequently, the concentration of the KI/I2 solution was increased to >2.5% KI and 1.0% I2. Then the appearance of the waxy wheat starch granules quickly became ghostlike. The internal structure of the ghost could be observed. This structure consisted of two parts with different colors: a red‐brown central part and a slightly pinkish surrounding part. Sonication of the ghost at 20 kHz for 20 sec separated the central part from the surrounding part. Each of the separated parts were subjected to gel‐filtration chromatography, and their chromatography profiles indicated only amylopectin and no amylose. The central and surrounding parts had molecular weights of 54.3 × 105 and 23.8 × 106 from Berry plots, respectively. 相似文献
